Les protéines oxydées, si elles ne sont pas dégradées par le protéasome, peuvent éventuellement s'accumuler et causer des dommages au niveau du métabolisme, de l'élastose et de la formation de pigment tel que la lipofuscine. Avec l'âge, l'activité du protéasome et des enzymes qui protègent les protéines de l'oxydation diminue. Il en résulte une accumulation de protéines oxydées. Le but de ce travail est d'étudier le pouvoir antioxydant d'un mélange de triacétate de panthényle et de linoléate d'éthyle (complexe bioactif). Ce complexe testé à 3 % sur des biopsies de peau humaine a montré une protection statistiquement significative de l'oxydation des protéines induite par les UV (UVA + UVB) en prétraitement (24 heures avant irradiation UV, 72 % de protection, P < 0.05) et immédiatement après irradiation (78% de protection, P < 0.05). Les rayons UV induisent aussi une diminution significative de l'ARNm utilisé dans la synthèse d'enzymes réparatrices telle la Methionine Sulfoxide Reductase (MSR). Le complexe bioactif stimule l'expression de l'enzyme réparatrice en pré et post application. Nous recommandons l'utilisation de ce nouvel actif pour prévenir l'accumulation de protéines oxydées due au photovieillissement de la peau, la déshydratation du stratum corneum, l'élastose de la peau et la formation de lentigos (pigmentation). The oxidation kinetics exhibits first order kinetics with respect to [Oxidant], fractional order of [Ala] and inverse fractional order of [H+] on the rate of reaction. The rate of reaction decreases with decreasing dielectric constant of the medium. The rate constant increases with increasing CTAB and then constant. Increasing [Hg(OAc)2] and inorganic salts i.e. [Cl–] and [Br–] increased the rate of reaction, whereas a change in ionic strength of the medium had no effect on oxidation velocity. Methyl Cyanide as the main oxidation product has been identified. Thermodynamic and activation parameters have been evaluated. The rate constants, binding constants with surfactant (CTAB) and the index of cooperativity have also been evaluated. Les comportements de teinture-formation pour le système du p-aminophenol (PAP) et du 5-amino-o-cresol (5AOC) avec des agents tensio-actifs, dont les pièces hydrophiles ont la charge différente, ont été comparés en changeant la concentration des agents tensio-actifs. On a trouvé que les mêmes colorants sont produits indépendamment de la charge des agents tensio-actifs supplémentaires et le taux de colorant produit dans le bain de teinture est augmenté en présence des agents tensio-actifs. L'ordre de la vitesse de production est, avec de l'agent tensio-actif anionique > avec de l'agent tensio-actif non ionique > avec de l'agent tensio-actif cationique > sans agent tensio-actif. La relation entre la capacité de teinture des cheveux et le taux de colorant produit dans le dyebath en présence des agents tensio-actifs n'est pas trouvée. Le facteur principal régissant la capacité de teinture des cheveux est différent du mécanisme du colorant accru dans la solution. On a également relevé que le taux teindre-formation est augmenté en immergeant des cheveux dans la solution de réaction et les cheveux fonctionnent comme accélérateur pour la réaction de teinture-formation. In addition to kinetic experiments, conductivity measurements have also been done to obtain critical micelle concentration and other thermodynamic properties. The kinetic observations indicate fractional order and first order dependence, with respect to [Acetone] and [NBP] respectively. Cationic surfactant (CTAB) strongly catalysed the oxidation of acetone in aqueous acetic acid medium and typical kobs and [CTAB] profile was observed i.e. with the progressive increase in [CTAB], the reaction rate increased, at higher concentration constancy in kobs was observed, whereas anionic surfactant (SDS) was found to show no considerable effect on reaction rate. Mercuric acetate and phthalimide exhibited nil effect on the reaction rate. The presence of inorganic salts (KCl, KBr) exhibits positive effect in the reaction rate. The various activation parameters in presence of CTAB and SDS have been also evaluated. A suitable mechanism consistent with the experimental findings has been proposed. Les protéines carbonylées sont les principaux produits oxydatifs des protéines et peuvent être introduits par réaction avec les aldéhydes dérivées du péroxyde lipidique. L'acroléine est l'un des aldéhydes le plus réactif engendré pendant la dégradation des péroxydes lipidiques et des protéine-acroléine adduits ont été trouvés dans la lésion endommagée oxydativement comprenant les peaux endommagées par les UV. Des études récentes ont révélé que les protéines carbonyleés étaient également détectées dans les fines couches supérieures de la peau, à savoir le stratum corneum (SC). Toutefois, l'effet de la carbonylation de protéine sur la fonction du SC n'était pas encore évident. Dans cette étude, nous avons traité des plaques d'épiderme reconstitué humain et d'épiderme porcin avec l'acroléine dans des conditions expérimentales pour étudier l'influence de la carbonylation de protéine sur le SC. Les plaques humaines et porcines du SC traitées avec l'acroléine ont montré une transmission à la lumière visible inférieure à celles non traitées. La spectroscopie infrarouge de réflexion totale atténuée (ATR-IR) avec l'analyse d'ajustement de la courbe sur des amides région I a montré que l'acroléine a induit des altérations dans la structure secondaire des protéines des plaques du SC porcin, qui étaient accompagnées d'une structure fibreuse diminuée de la kératine observée par microscopie électronique à transmission. Ces résultats montrent la possibilité que la carbonylation du SC causée par des facteurs environnementaux soit l'un des éléments participant à la modification de la structure fibreuse de la kératine, et à la diminution de la transmission de la lumière du SC, modifiant la qualité d'aspect de la peau.
