Le but de cette étude était de déterminer les effets dose–réponse de nature hydratante et tenseur de formulations cosmétiques enrichies de composants extraits d'algues et de collagène de poisson. Divers facteurs ont été étudiés parmi lesquels le type de formulation (crème ou sérum), la concentration en ingrédients actifs, l'effet d'applications répétitives, ainsi que l'effet résiduel des formulations après cessation des applications. Le sérum enrichit en composants marins avait un meilleur effet hydratant à court terme. La crème est apparue plus active par après, particulièrement à la suite d'applications répétitives. Un effet tenseur (« liftant ») persistant a été observé pendant le traitement avec la crème et le sérum. Cependant, aucun effet tenseur persistant n'a été perçu après l'arrêt des traitements. Panthenol, the alcohol form of pantothenic acid (vitamin B5), is believed to act as a humectant and improve the flexibility and strength of nails. A liquid nail treatment formulated with panthenol (2%) was compared to a solution of panthenol (2%) in water. Fingernail specimens were dosed daily for 7 days with either the nail treatment (non-lacquer film forming) formulation or aqueous solution with sampling performed every 24 h. Panthenol concentrations were determined in the dorsal surface, interior (by drilling and removal) and in the supporting bed under the human nail. Panthenol levels in the dorsal nail (R2 = 0.87 ; P < 0.001), nail interior (R2 = 0.94 ; P < 0.001) and nail supporting bed (R2 = 0.79 ; P < 0.003) showed a significant linear increase with each day of dosing. Significantly more panthenol was delivered into the interior nail and supporting bed by a nail treatment formulation than from an aqueous solution. The film acts not only as a reservoir of panthenol, but also acts to increase the hydration of the nail and the thermodynamic activity of panthenol as well, thereby enhancing diffusion. The oxidation kinetics exhibits first order kinetics with respect to [Oxidant], fractional order of [Ala] and inverse fractional order of [H+] on the rate of reaction. The rate of reaction decreases with decreasing dielectric constant of the medium. The rate constant increases with increasing CTAB and then constant. Increasing [Hg(OAc)2] and inorganic salts i.e. [Cl–] and [Br–] increased the rate of reaction, whereas a change in ionic strength of the medium had no effect on oxidation velocity. Methyl Cyanide as the main oxidation product has been identified. Thermodynamic and activation parameters have been evaluated. The rate constants, binding constants with surfactant (CTAB) and the index of cooperativity have also been evaluated. In addition to kinetic experiments, conductivity measurements have also been done to obtain critical micelle concentration and other thermodynamic properties. The kinetic observations indicate fractional order and first order dependence, with respect to [Acetone] and [NBP] respectively. Cationic surfactant (CTAB) strongly catalysed the oxidation of acetone in aqueous acetic acid medium and typical kobs and [CTAB] profile was observed i.e. with the progressive increase in [CTAB], the reaction rate increased, at higher concentration constancy in kobs was observed, whereas anionic surfactant (SDS) was found to show no considerable effect on reaction rate. Mercuric acetate and phthalimide exhibited nil effect on the reaction rate. The presence of inorganic salts (KCl, KBr) exhibits positive effect in the reaction rate. The various activation parameters in presence of CTAB and SDS have been also evaluated. A suitable mechanism consistent with the experimental findings has been proposed. En général, on peut distinguer les effets thermodynamiques du vecteur dus à différentes façons de libérer le produit des vecteurs et les effets améliorant la pénétration des vecteurs dus à une modification de la structure de la couche cornée, ce qui se manifeste par une augmentation du coefficient de diffusion du produit et/ou de sa solubilité dans cette barrière. Puisque le temps de latence du début d'un effet pharmacodynamique, habituellement utilisé comme valeur réciproque, représente un paramètre de réponse adéquat dans certaines circonstances, cette étude a été effectuée pour évaluer ce paramètre plus en détail vis-à-vis de la mesure des données de la biodisponibilité relative et de l'amélioration de la pénétration obtenues à partir de courbes dose–réponse (et activité) simulés faisant l'hypothèse de conditions de dosage infinies, c'est-à-dire d'une cinétique de pénétration d'ordre zéro et en prenant des temps de latence variables pour la pénétration de la substances. Les résultats indiquent que les facteurs de biodisponibilité et d'amélioration peuvent être déterminés précisément à partir de la distance horizontale entre les courbes dose–réponse (ou activité) d'une référence et d'une préparation testée tant que les courbes sont parallèles l'une à l'autre, comme c'est le cas avec les temps de latence uniformes de pénétration du produit. Des courbes non parallèles observées avec des temps de latence variables indiquent une influence des vecteurs sur le coefficient de diffusion du produit dans la barrière. On peut obtenir les facteurs d'amélioration à partir de ces courbes après détermination des temps de latence dans la zone plateau des courbes, puis soustraction de ces valeurs des données de temps de latence mesurées, et finalement tracé des données réciproques en fonction de l'activité de la substance. Les facteurs d'amélioration correspondent alors au logarithme inverse des distances horizontales entre les courbes parallèles résultantes.
