Langues : Anglais (eng) Tags : Anti-inflammatory Clinical study Laser doppler blood flowmetry Pseudopterosins Index. décimale : 668.5 Parfums et cosmétiques Résumé : Pseudopterosins extracted from Caribbean Sea Whip (Pseudopterogorgia elisabethae) were previously shown to inhibit phospholipase A2, an enzyme that functions in the arachidonic acid cascade of inflammation. With an objective to test its in vivo anti-inflammatory potential, we conducted a screening clinical bioassay using randomized analysis on sixteen panelists. An emulsion containing 0.02% of sea whip extract was applied to the volar forearm of subjects prior to or after an ethyl nicotinate challenge. Blood flow rate at the application site monitored using a laser doppler blood flowmeter served as a measure of inflammation induced by ethyl nicotinate. Upon post-challenge treatment, sea whip extract reduced blood flow by 35.04% against untreated control and also delayed the blood flow. More pronounced reduction in blood flow, as low as 59.50%, was noted upon pretreatment. This reduction was significant (p < 0.0095). Pretreatment caused not only a delay in onset of inflammation but also a reduction in the peak time of blood flow. This study further substantiates the previously reported in vitro anti-inflammatory effect of sea whip extract, demonstrating its activity in deep skin layers by affecting blood flow. The preventive application prior to induction of inflammation was more effective when compared to post-treatment. We focus on an equimolar aqueous solution of cetylpyridinium chloride – sodium salicylate, which in simple shear flow exhibits a shear thinning, and at higher shear rates, a pronounced shear-thickening behavior associated with the formation of shear bands [1–6]. An ultrasound Doppler velocimetry method is used to detect these shear bands in pipe flow. The emitted ultrasonic beam is scattered by the moving sample and the time delay between emitted pulse and received echoes as well as the frequency shift is determined. This Doppler shift is related to the speed and the direction of the moving scatters and with this information, the velocity profile across the pipe is calculated [7]. The shape of the obtained velocity profile is used to determine the flow properties of the sample and the formation of shear bands at the tube's wall or within the bulk phase. For the investigated solution we do not observe shear band phenomena. This is contrary to previous investigations [5, 6, 8] where shear bands in the order of few tenth of a millimeter were found. Dans cette étude, nous avons analysé l'irritation provoquée sur 37 volontaires bien portants par des applications locales, répétées, sur une période de 5 jours, de savons et détergents synthétiques présents sur le marché. Les tests ont été effectués et évalués cliniquement et par débimétrie à laser doppler (LDF) selon une méthode standard développée antérieurement.
Les savons et détergents synthétiques pourraient être divisés en groupes d'égale irritation. Nous avons pu observer que les résultats physiques concordaient parfaitement avec les résultats de la LDF. La débimétrie à laser doppler s'est avérée être une méthode précise, fiable et tout à fait adaptée à l'évaluation du degré d'irritation de la peau. Au cours de nos expériences, nous n'avons pu établir de relation nette entre le pH des nettoyants et leur irritation sur la peau. Nous en avons déduit que le résultat des tests d'irritation est grandement influencé par des circonstances telles que la méthode d'applications (durée, fréquence, concentration...) et le mode d'évaluation (durée des tests et méthode). Par conséquent, les conclusions des observateurs peuvent être diverses, et la prudence est nécessaire dans la comparaison entre les différentes études.Permalink : https://e-campus.itech.fr/pmb/opac_css/index.php?lvl=notice_display&id=5194

Langues : Anglais (eng) Tags : Transepidermal water loss Laser doppler velocimetry Surfactants Stripping Occlusion Hydration Skin permeability Human skin Index. décimale : 668.5 Parfums et cosmétiques Résumé : La fonction de barrière du stratum corneum a été évaluée, in vivo chez l'homme, après différents traitements physiques et chimiques, par deux méthodes non invasive : la mesure de la Perte Insensible en Eau (PIE) transépidermique par évaporimétrie à l'aide d'un évaporimètre TM Servo Med et la Vélocimétrie Laser Doppler (VLD), avec le Laser Flo TM BPM 403 T.S.A., pour mesurer le temps de latence avant l'apparition d'une vasodilatation induite par l'application de méthyl et d'hexyl nicotinate.
Les traitements physiques et chimiques réalisés ont été :
1. La délamination du stratum corneum par 8 et 12 strips ; dans ces deux cas, un site non traité sert de témoin.
2. L'occlusion et l'hydratation du stratum corneum, respectivement par un patch occlusif (3 cm2) seul ou avec 400 µl d'eau distillée pendant 5 heures 30.
3. L'application de trois surfactifs, le tween 60, le lauryl sulfate de sodium ( SDS) et le bromure de cétyl triméthylammonium (CTAB), en solution aqueuse à 20 %, sous un volume de 400 µl, sous un patch occlusif pendant 5 heures 30. Un site traité à l'eau distillée dans les mêmes conditions sert de témoin.
L'étude a été effectuée sur les deux avant-bras de 8 à 10 volontaires sains, âgés de 23 à 30 ans. Les mesures de PIE sont réalisées avant et après traitement.
L'augmentation de PIE et la diminution du temps de latence avant vasodilatation mesurées par VLD après tous les traitements confirment les modifications de la perméabilité cutanée. Les variations observées sont plus ou moins significatives selon le traitement, la méthode de mesure et la molécule utilisée pour l'étude de la pénétration percutanée.
Avec le nicotinate de méthyle (hydrophile), plus la PIE augmente, plus le temps de latence diminue. Une diminution significative du temps de latence après application du nicotinate d'hexyle (lipophile) n'est observée que lorsqu'il y a une très forte augmentation de la PIE. La nature lipophile de la barrière cutanée est confirmée in vivo. La bonne utilisation de ces deux méthodes complémentaires. Regarding the blood flow, we found that the areas surrounding the internal canthus where the dark circles are frequently observed had larger blood mass and slower blood flow velocity compared to the other regions. Furthermore, this tendency is particularly evident in the superficial layers of the skin, and also in the lower part and the lateral part of the internal canthus. Our results suggest that naturally the blood flow in the areas surrounding the internal canthus tends to stagnate easily. Concerning the skin color, we found that the areas around the internal canthus tend to appear, particularly the skin color under the internal canthus had a low lightness (L*) value. There was a significant correlation between the differences in the blood mass (ΔMass) and the differences in lightness (ΔL*) in the lower part of the internal canthus and its adjacent regions. Furtheremore, compared to the person with less prominent dark circles, the person with prominent circles had larger differences between the lower part of the internal canthus and its adjacent regions in terms of the blood mass as well as the lightness. From our results, we concluded that retention of blood in the surface of the skin is a significant factor in the development of dark circles around the eyes.
