Microscopie électronique en transmissionIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : The effects of polyacrylic acid (PAA) on the hierarchical structure of sheepskin collagen fibres were investigated by using of transmission electron microscope (TEM), scanning electron microscopy (SEM), X-ray diffraction (XRD) and differential scanning calorimetry (DSC). It was found that although the unique axial periodic (d periodicity) structure of collagen fibrils and orderly arrangement of collagen molecule did not change obviously alter the introduction of PAA, the aggregation behaviour of collagen fibres was greatly influenced. With increasing concentration of PAA, the order of intermolecular lateral packing decreased, the shrinkage of the leather lessened [from changes in surface morphology observed by SEM] and the dry heat stability of collagen fibre declined due to a decrease in the orderly degree of collagen fibre aggregation. Tannage Résumé : The US meat industry currently produces approximately 35 million cattle hides annually as its most valuable coproduct. These hides serve as raw material, first for the leather industry, and then for the gelatin, and biomaterials industries. The conversion of animal hides into leather is a multistep process that has evolved more as art form than as science. Economic or environmental issues typically dictate changes in beam-house processes that prepare the hide for tanning. The tanner evaluates these changes, in terms of impact on tannery costs and quality of leather produced. Thus far, the effects of beam-house processes on the molecular characteristics of collagen have received little attention. The basis for tanning and most biomaterials applications is the stabilization of the collagen matrix, thus any changes to the molecular characteristics of hide collagen may be expected to impact these applications. This study showed that while the effects of different dehairing processes on the structure and stability of monomeric collagen were similar, the effects on the collagen fiber structure were distinct. These results are anticipated to assist the tanner as well as the manufacturers of collagen-based biomaterials and gelatin to better understand their substrate and changes to it that may occur when beam-house processes are altered. GélatineLa gélatine est une substance solide translucide, transparente ou légèrement jaune, presque sans goût et sans odeur, obtenue par l'ébullition prolongée de tissus conjonctifs (peaux) ou d'os d'animaux (principalement porc, bœuf, poisson). Elle possède de nombreuses applications dans le domaine culinaire, la médecine, les industries agroalimentaire et pharmaceutique.
En matière d’étiquetage, la gélatine est considérée par la norme européenne3 comme un ingrédient et non pas comme un additif, c'est pourquoi elle n'a pas de numéro E. Hors Union européenne, elle est considérée par certains pays comme un additif gélifiant et on peut la trouver avec la dénomination E441.
La gélatine est un mélange de protéines obtenu par hydrolyse partielle du collagène extrait de la peau comme la peau de porc (cochon), des os, des cartilages, etc. Les liaisons moléculaires entre les fibres de collagène sont alors brisées. Mélangée à de l'eau, la gélatine forme un gel colloïdal semi-solide thermo-réversible (il fond lorsqu'il est chauffé et recouvre son aspect gélatineux lorsqu'il est refroidi). Sous forme déshydratée, par contre, la gélatine n'a pas de point de fusion et devient friable ou brûle quand elle est chauffée à trop haute températureLa rhéologie de la gélatine se caractérise par un comportement viscoélastique, et des contraintes trop élevées ou appliquées trop rapidement peuvent entraîner une rupture fragile (fracturation) ou ductile6. Le caractère plutôt élastique/fragile ou plutôt visqueux/ductile dépend de la concentration en gélatine de la solution aqueuse et de la température, ainsi que de la durée de la mise sous contrainteLes acides aminés constituant la gélatine sont : la glycine (21 %), la proline (12 %), l'hydroxyproline (12 %), l'acide glutamique (10 %), l'alanine (9 %), l'arginine (8 %), l'acide aspartique (6 %), la lysine (4 %), la sérine (4 %), la leucine (3 %), la valine, la phénylalanine et la thréonine (2 %), l'isoleucine et l'hydroxylysine (1 %), la méthionine et l'histidine (< 1 %) et la tyrosine (< 0,5 %). Ces valeurs sont variables (surtout pour les constituants minoritaires) et dépendent de la source de matériaux bruts et de la technique de préparation. La gélatine est constituée à environ 98-99 % (en poids sec) de protéines et contient 18 acides aminés dont huit des neuf acides aminés essentiels à l'Homme. Elle n'a qu'une relative valeur nutritionnelle du fait de l'absence de tryptophane et de son déficit en isoleucine, thréonine et méthionine; elle possède également un taux inhabituellement élevé d'acides aminés non essentiels, la glycine et la proline (qui sont produits par le corps humain). One can not only observe the various entities and study their morphology but also follow the reactions that a substance undergoes when subjected to several treatments.
The advantages of modern electron microscopy include, apart from the require ment of small quantities of sample, stereoscopic view, possibility of heating or cooling the sample, and observation of the specimen under moist conditions.

EnzymesUne enzyme est une protéine dotée de propriétés catalytiques. Pratiquement toutes les biomolécules capables de catalyser des réactions chimiques dans les cellules sont des enzymes ; certaines biomolécules catalytiques sont cependant constituées d'ARN et sont donc distinctes des enzymes : ce sont les ribozymes.
Une enzyme agit en abaissant l'énergie d'activation d'une réaction chimique, ce qui accroît la vitesse de réaction. L'enzyme n'est pas modifiée au cours de la réaction. Les molécules initiales sont les substrats de l'enzyme, et les molécules formées à partir de ces substrats sont les produits de la réaction. Presque tous les processus métaboliques de la cellule ont besoin d'enzymes pour se dérouler à une vitesse suffisante pour maintenir la vie. Les enzymes catalysent plus de 5 000 réactions chimiques différentes2. L'ensemble des enzymes d'une cellule détermine les voies métaboliques qui peuvent avoir lieu dans cette cellule. L'étude des enzymes est appelée enzymologie.
Les enzymes permettent à des réactions de se produire des millions de fois plus vite qu'en leur absence. Un exemple extrême est l'orotidine-5'-phosphate décarboxylase, qui catalyse en quelques millisecondes une réaction qui prendrait, en son absence, plusieurs millions d'années3,4. Comme tous les catalyseurs, les enzymes ne sont pas modifiées au cours des réactions qu'elles catalysent, et ne modifient pas l'équilibre chimique entre substrats et produits. Les enzymes diffèrent en revanche de la plupart des autres types de catalyseurs par leur très grande spécificité. Cette spécificité découle de leur structure tridimensionnelle. De plus, l'activité d'une enzyme est modulée par diverses autres molécules : un inhibiteur enzymatique est une molécule qui ralentit l'activité d'une enzyme, tandis qu'un activateur de cette enzyme l'accélère ; de nombreux médicaments et poisons sont des inhibiteurs enzymatiques. Par ailleurs, l'activité d'une enzyme décroît rapidement en dehors de sa température et de son pH optimums.

Wet-blue (tannage)Peau tannée au chrome (le chrome donne une couleur bleue)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : To characterize the reacting performances of proteases against chrome-tanned leather, the methods to quantitatively measure protease activities against chrome-tanned elastin and collagen fibers were established through detecting the produced amount of Desmosine (DES) or Hydroxyproline (Hypro) in unit time. Chrome-tanned elastin and collagen fiber substrates with different Cr content were prepared, and the influence of the Cr content on the enzymatic hydrolysis resistance of tanned substrates was investigated. The activities of several typical proteases against chrome-tanned elastin and collagen fibers were tested based on the methods, and the relationship between the activities and bating effectiveness on wet blue was also preliminarily investigated. The results showed that the two kinds of activities of proteases decreased with the increase of Cr content in the substrates. When the Cr contents in elastin or collagen fiber reached 0.5% or 2.0%, the corresponding activity approached to the lowest level, respectively. Different protease showed different activity decrease extend towards tanned protein fibers. Meanwhile, the bating effectiveness of proteases on wet blue was positively related to their activities towards the chrome-tanned protein fibers measured by the established methods, i.e. proteases with highly chrome-tanned elastin and collagen fiber activities could significantly improve the softness of leather. The new methods can be used as an available tool to correctly select proteases and optimize process parameters for wet blue bating. Note de contenu : - MATERIALS AND METHODS : Materials - Methods.
