ProtéasomeLes protéasomes sont des complexes enzymatiques multiprotéiques que l'on retrouve chez les eucaryotes, les archées ainsi que chez quelques bactéries de l'ordre Actinomycetales. Dans les cellules eucaryotes, ils se trouvent dans le noyau, le cytosol et associés au réticulum endoplasmique. Leur fonction principale est de dégrader les protéines mal repliées, dénaturées ou obsolètes de manière ciblée. Cette dégradation se fait par protéolyse, une réaction chimique qui coupe les liaisons peptiques et qui est effectuée par des enzymes appelées protéases. La protéine est ainsi découpée en peptides longs de 7 à 9 acides aminés qui seront ensuite hydrolysés hors du protéasome et recyclés. Les protéines sont marquées pour la dégradation par une protéine appelée ubiquitine. Ce marquage est réalisé par l'action coordonnée de trois types d'enzymes. Une fois le marquage par une première molécule d'ubiquitine réalisé, d'autres ubiquitines vont être rajoutées à sa suite. Il faudra une chaîne d'au moins quatre ubiquitines pour que le protéasome 26S reconnaisse la protéine à dégrader. Il existe un compartiment pour celui-ci.
Le protéasome a une forme de baril et possède une cavité en son centre cernée par quatre anneaux, fournissant ainsi un espace clos pour la digestion des protéines. Chaque anneau est composé de sept protéines : les deux anneaux intérieurs sont constitués de sept sous-unités β qui contiennent le site actif de la protéase, tandis que les deux anneaux extérieurs contiennent sept sous-unités α dont le rôle consiste à maintenir l'ouverture par laquelle les protéines à dégrader pénètrent dans le baril: ces sous-unités α sont capables de reconnaître les marqueurs de polyubiquitine qui régulent le processus de dégradation. KératinocytesLes kératinocytes sont des cellules constituant 90 % de la couche superficielle de la peau (épiderme) et des phanères (ongles, cheveux, poils, plumes, écailles). Ils synthétisent la kératine (kératinisation), une protéine fibreuse et insoluble dans l'eau, qui assure à la peau sa propriété d'imperméabilité et de protection extérieure.
L'épiderme est divisé en 4 couches basées sur la morphologie des kératinocytes (de l'intérieur vers l'extérieur) :
1. stratum germinativum (couche basale à la jonction avec le derme)
2. stratum spinosum
3. stratum granulosum
4. stratum lucidum
5. stratum corneum
Les kératinocytes passent progressivement de la couche basale vers les couches supérieures par différenciation cellulaire jusqu'au stratum corneum ou ils forment une couche de cellules mortes nommées squames, par apoptose. Cette couche constitue une barrière de protection et réduit la perte d'eau de l'organisme.
Les kératinocytes sont en perpétuel renouvellement. Ils mettent environ 1 mois pour aller de la couche basale au stratum corneum mais ce processus peut être accéléré en cas d'hyperprolifération de kératinocyte (psoriasis).

huile d'ahiflowerL'huile ahiflowerMD est extraite des graines du Buglossoides arvensis (L.) I.M.Johnst., communément appelé grémil des champs ou orcanète bâtarde, une plante herbacée annuelle appartenant à la famille des boraginacées originaires d'Europe et d'Asie. Le B. arvensis n'est pas génétiquement modifié, mais produit au moyen de techniques de sélection traditionnelles.
L'huile AhiflowerMD est tirée des graines de plantes contenant des triglycérides (90 %), des diglycérides (2-6 %), des monoglycérides (2-4 %), des acides gras libres (<0,3 %) et une fraction insaponifiable (<1,5 %) composée de phytostérols, dont le β-sitostérol, le campestérol, le stigmastérol et le γ-tocophérol.
L'huile AhiflowerMD contient environ 20 % d'acide stéaridonique (SDA) (% d'acides gras), un acide gras oméga-3 qui agit comme intermédiaire dans la synthèse de l'acide eicosapentanoïque (EPA) de l'acide alpha-linolénique (ALA). L'huile AhiflowerMD contient également environ 45 % d'ALA et 6 % d'acide gamma-linolénique (GLA). Since the Industrial Revolution, air pollution has been a common daily experience for most people living in cities and for many people living with smokers or near power plants and factories. Much of the reforms of the environmental movement beginning in the 1960s and 1970s focused on cleaning up and preventing air pollution - smoke, smog, particulates, petroleum distillates, car and truck exhausts and brake dusts etc. - at their sources.
These efforts greatly reduced people’s exposures to air pollution and therefore to ongoing risk for cardiovascular and pulmonary diseases including lung cancer, while also reducing ecological impacts like acid rain harming fish and frogs in watersheds downstream from polluted cities. However, the negative health impacts of air pollution (indoor and outdoor) still is the fifth largest contributor to mortality worldwide and over 90% of people globally are impacted by air pollution.
Progress has been made. In the 50 years between 1970 and 2020 in the United States, lung cancer more than halved from the #3 cause of death at 78 per 100,000 (men), to less than 32 per 100,000 of (both sexes). Despite real human health and environmental wins resulting from controlling and reducing air pollution, however, much less attention has been given to the impacts of air pollution on the largest organ in our bodies : the skin.
This article summarizes recent research on the pro-inflammatory effects of common air pollution on human skin and then proposes certain natural skin care and dietary protocols using plant-based omega-3 and omega-6 polyunsaturated fatty acids to help protect the skin from accelerated harmful effects.Note de contenu : - The skin's role in air pollution protection

Skin, as our biggest organ and the barrier between the organism and the outside world, is directly exposed to ambient pollution. We can remove the pollutants by daily cleansing, or protect skin to some extent by applying a physical film to shield it from harmful pollutants, but for full protection we need to strengthen the skin's own barrier function.
We therefore investigated a number of actives for their capacity to increase skin resilience to common airborne pollutants. Based on the results of this skin biological research, we identified actives that demonstrated protective effects and integrated them into a novel concept affording comprehensive anti-pollution protection and relief for vulnerable skin.
Because epigenetic changes may be implicated in changes to phenotypic appearance linked to environmental pollution, we included the new field of epigenetics in out research. Investigation of various potential epigenetic marks revealed that the specific micro-RNA miR-450b-5p, which is associated with cellular senescence, could demonstrate environmentally stressed changes. TélomèresUn télomère est une région hautement répétitive, donc non codante, d'ADN à l'extrémité d'un chromosome. À chaque fois qu'un chromosome en bâtonnet d'un eucaryote est dupliqué, lors de la réplication, qui précède la mitose (division cellulaire), le complexe enzymatique de l'ADN polymérase s'avère incapable de copier les derniers nucléotides : l'absence de télomère signifierait la perte rapide d'informations génétiques nécessaires au fonctionnement cellulaire.
Les télomères raccourcissent avec l’âge, l’inflammation et le stress. Des études ont montré que des télomères courts sont associés à un risque plus élevé de maladies liées à l’âge.Index. décimale : 668.5 Parfums et cosmétiques Résumé : Today, it seems that different factors are interlinked and combined with other processes and on different levels to contribute to the skin ageing phenomenon. On a cell level, telomeres’ shortening is considered one of the main characteristics of ageing. Over the course of cell division, the telomeres gradually shorten until they reach a critical minimum size, triggering cell senescence which contributes to the degeneration of the skin tissue and its ageing.
In skin cells, telomeres are stabilised by a multiprotein complex, called telosome or shelterin. In the absence of the telosome, the weakened telomeres shorten more quickly at each division.
Stress (UV, reactive oxygen species, etc.) also accentuates their wear. This leads to gradual depletion of the cells and its premature ageing.
In this article, we discuss the in vitro models we developed for studying the capacity of a natural active ingredient to limit deterioration of the telosome. This active prevents the accelerated erosion of telomeres induce by stress and slows down premature passage of skin cells into the senescent phase and thus ensures longevity.
This new anti-ageing strategy is supported by proven cosmetic benefits : the marks of ageing on the skin are reduced and the perceived age is improved.Note de contenu : - The telosome : the protective cap of telomeres
