Environmental pollution now-a-days is considered one of the main causes of skin ageing. Among the pollutants, tobacco smoke and PM2.5 (particulate matter) are thought to be the major health hazardous components responsible for skin in-flammation, pigmentation, and wrinkling, as evident from sporadic clinical and epidemiological studies. Our objective was to develop a plant-based natural ingredient that could suppress the detrimental effects of those two pro-minent pollutants on human skin tells. Normal human epidermal keratinocytes (NHEK) and normal human dermal fibroblasts (NHDF) were exposed to PM2.5 and tobacco smoke, respectively, for the induction of inflammation and collagenase activity. Various plant extracts were screened for their efficacy in showing anti-ageing properties against these pollutants. We showed that our unique extract, Ougon, was able to suppress the tobacco smoke-induced expression of matrix metalloproteinase-1 (an enzyme responsible for the degradation of extracellular matrix) in NHDF by inhibiting the expression of CYP1B1 (a member of the cytochrome p450 family). Moreover, Ougon was found to have strong efficacy in inhibiting the PM2.5-induced expression of some inflammatory mediators (e.g. IL-113, IL-6, and IL-8) in NHEK in concentration-dependent manners. Therefore, we suggest that Ougon extract may protect the MMP-1 and inflammation-mediated ageing of skin induced by environmental stress factors. |
