The human patch test is an appropriate method for this type of assessment, but it cannot be used for the higher throughput needed for screening innovative formulations and a distinct assessment is often difficult. The aim of this study was therefore to investigate the feasibility of using an in vitro approach for compatibility testing to determine the irritancy of surfactants. Test samples were provided by the German Society of Cosmetic Chemists (DGK), which conducted a human patch test study with the same set of samples in parallel. This gave us the unique opportunity to correlate in vitro with in vivo data. To assess irritant effects in vitro, reconstructed human epidermis was exposed to seven coded test samples consisting of individual anionic surfactants, blends of surfactants, and controls. A multiple endpoint analysis was established comprising the viability, cytotoxicity, histology, cytokine release and differential gene expression. Using this test strategy, a very good correlation was determined for our in vitro assessment of compatibility with a theoretical ranking and the human patch test data. Les quelques études conduites sur la signification de l’état cutané apparent vis-à-vis de l’apparence physiques ont concernés le visage seul ou des images de zones précises du visage. On en connaît peu à propos de l’impact de la perception du visage et ses correspondance avec d’autres parties du corps ou si la peau du corps affecte la perception général de l’âge ou l’attrait quand elle est présentée en association avec la peau de la face. Nous avons formulé l’hypothèse que la présentation indépendante de visages féminins, de poitrines et de bras (incluant Les mains) - coupé d’une image de visage complet et de la partie supérieure du corps - aboutirait à des différences significatives dans la perception de l’âge et dans l’ attrait. En outre, nous avons cherchéà examiner si une peau d’aspect relativement jeune et attractive sur les parties choisies du corps affectait la perception complète. Les photographies numériques de 52 femmes âgées de 45 à 65 ans ont été rassemblées et traitées pour obtenir quatre jeux d’images : Un jeu a montré l’ensemble de toutes les zones, c’est-à-dire, le visage, la poitrine et les bras, tandis que les trois autres ont été coupés soigneusement pour ne montrer que chaque partie du corps indépendamment. Un total de 240 participants a jugé ces visages pour l’âge perçu et l’attrait. Nos résultats ont montré des différences significatives de la perception : la poitrine et les bras étant jugés significativement plus jeunes que le visage ou l’image composée des mêmes femmes. De plus, les images des bras et de la poitrine ont été perçues comme plus séduisantes que celles du visage et celles des visages composés. Finalement, l’analyse en régression a indiqué que les différences entre les valeurs perçues et les valeurs chronologiques de perception d’âge complet pourraient être prédites par la perception de l’âge du visage et des bras. Ces résultats continuent à soutenir l’importance de la perception de l’âge du visage dans l’évaluation de l’âge d’une femme, mais le point à souligner est le rôle de la peau du corps dans l’impression d’âge global. These two parameters, as well as the measurement of cellular viability, were used to evaluate the effects of UV irradiation on a reconstructed human epidermis (RHE) model, with and without the photoprotection of a broad spectrum sunblock. Reconstructed epidermis models were irradiated by solar spectrum (420 mJ cm–2) or UVA (20 J cm–2). In the absence of irradiation, and with irradiation doses of 420 mJ cm–2 and 20 J cm–2, viability was estimated at 95, 12 and 70% respectively. Sunburn cells per cm were evaluated at 0, 41 and 22, respectively under these same conditions. The apoptotic response was studied through the expression of p53, which increased at first and was then followed by a specific cleavage, 24 h after irradiation at 420 mJ cm–2. However, in the presence of a broad spectrum sunblock preparation (a combination of OMC, MBTBP, TiO2 and ZnO) and after irradiation at 420 mJ cm–2 and 20 J cm–2, viability increased to 40 and 85% respectively. The number of sunburn cells (SBC) per cm fell to 12 and nine respectively. The photoprotection provided by the sunblock limited the increase in the expression of p53 and resulted in the total disappearance of the cleavage. This study shows that human reconstructed epidermis is a valid skin model for use in the evaluation of the effects of solar and UVA irradiation, as well as in the testing of the efficacy of sunfilters. Special care was taken to standardize the physical history of the samples. With increasing temperature, isotherms, while preserving their shape, shift to lower regains while the hysteresis effect decreases. This is in accordance with expectations derived from the Rosenbaum-model for water sorption. The differential heat of water sorption changes from dry to wet hair between 1020 and 50 J/g H2O. Diffusion coefficients are of the order of 10-9 cm2/s. The coefficients increase with temperature and show a pronounced maximum in the range of medium regains. The temperature dependence follows the Arrhenius equation and exhibits activation energies that change from dry to wet hair between 55 and 30 kJ/mol. The sorption performance of strongly bleached hair, with and without an additional treatment with cetyltrimethylammonium bromide, was investigated. No significant effects either of the cosmetic treatment or of the ingredient were found. It is concluded that human hair exhibits a rather robust static and dynamic water sorption performance that, against initial expectations, is not readily changed by cosmetic processes and ingredients.
