The human patch test is an appropriate method for this type of assessment, but it cannot be used for the higher throughput needed for screening innovative formulations and a distinct assessment is often difficult. The aim of this study was therefore to investigate the feasibility of using an in vitro approach for compatibility testing to determine the irritancy of surfactants. Test samples were provided by the German Society of Cosmetic Chemists (DGK), which conducted a human patch test study with the same set of samples in parallel. This gave us the unique opportunity to correlate in vitro with in vivo data. To assess irritant effects in vitro, reconstructed human epidermis was exposed to seven coded test samples consisting of individual anionic surfactants, blends of surfactants, and controls. A multiple endpoint analysis was established comprising the viability, cytotoxicity, histology, cytokine release and differential gene expression. Using this test strategy, a very good correlation was determined for our in vitro assessment of compatibility with a theoretical ranking and the human patch test data. It is well known that NO, generated from L-arginine (L-Arg) by the action of NO synthase (NOS), plays a key role in the control of vascular tone and peripheral blood flow. In these studies we found a dose-dependent enhancement of constitutive NO production by L-PCA in human aortic endothelial cells. In contrast, D-PCA had no influence on NO production. Furthermore, L-PCA stimulated L-Arg uptake into human aortic endothelial cells in a dose-dependent manner while D-PCA had no effect, similar to its effect on NO production. L-Arg uptake and its stimulation by L-PCA to human aortic endothelial cells were suppressed with the cationic amino acid transporter. This result suggests that the enhancement of the NO production by L-PCA is due to the increase of L-Arg uptake via cationic amino acid transporter pathway. In conclusion, we found that L-PCA enhances micro blood circulation via a modulation of constitutive NO production. This intrinsic biological effect of L-PCA could lead to the development of innovative cosmetic products to care for problematic skin, such as dark circles under the eyes where a reduced micro blood circulation is one of the underlying causes. The feel of natural and damaged hair was studied after treatment with classical conditioning formulations. A cetrimonium chloride/cetyl alcohol conditioner and shampoos based on sodium lauryl ether sulphate and polyquaternium-10 or dimethiconol, respectively, were used. In addition to a broad-based round robin test to determine the hair feel in 12 test centres, suppleness measurements, atomic force microscopy and scanning electron microscopy, combing force measurements and shine measurements were performed. The cetrimonium chloride conditioner and the silicone shampoo produced the best hair feel. There was a correlation between hair feel and the suppleness measurement. The coating of the hair surface after shampooing with polyquaternium-10 and dimethiconol could be seen with atomic force microscopy and scanning electron microscopy.
