Langues : Anglais (eng) Tags : Anti-inflammatory Clinical study Laser doppler blood flowmetry Pseudopterosins Index. décimale : 668.5 Parfums et cosmétiques Résumé : Pseudopterosins extracted from Caribbean Sea Whip (Pseudopterogorgia elisabethae) were previously shown to inhibit phospholipase A2, an enzyme that functions in the arachidonic acid cascade of inflammation. With an objective to test its in vivo anti-inflammatory potential, we conducted a screening clinical bioassay using randomized analysis on sixteen panelists. An emulsion containing 0.02% of sea whip extract was applied to the volar forearm of subjects prior to or after an ethyl nicotinate challenge. Blood flow rate at the application site monitored using a laser doppler blood flowmeter served as a measure of inflammation induced by ethyl nicotinate. Upon post-challenge treatment, sea whip extract reduced blood flow by 35.04% against untreated control and also delayed the blood flow. More pronounced reduction in blood flow, as low as 59.50%, was noted upon pretreatment. This reduction was significant (p < 0.0095). Pretreatment caused not only a delay in onset of inflammation but also a reduction in the peak time of blood flow. This study further substantiates the previously reported in vitro anti-inflammatory effect of sea whip extract, demonstrating its activity in deep skin layers by affecting blood flow. The preventive application prior to induction of inflammation was more effective when compared to post-treatment. It is well known that NO, generated from L-arginine (L-Arg) by the action of NO synthase (NOS), plays a key role in the control of vascular tone and peripheral blood flow. In these studies we found a dose-dependent enhancement of constitutive NO production by L-PCA in human aortic endothelial cells. In contrast, D-PCA had no influence on NO production. Furthermore, L-PCA stimulated L-Arg uptake into human aortic endothelial cells in a dose-dependent manner while D-PCA had no effect, similar to its effect on NO production. L-Arg uptake and its stimulation by L-PCA to human aortic endothelial cells were suppressed with the cationic amino acid transporter. This result suggests that the enhancement of the NO production by L-PCA is due to the increase of L-Arg uptake via cationic amino acid transporter pathway. In conclusion, we found that L-PCA enhances micro blood circulation via a modulation of constitutive NO production. This intrinsic biological effect of L-PCA could lead to the development of innovative cosmetic products to care for problematic skin, such as dark circles under the eyes where a reduced micro blood circulation is one of the underlying causes. Elle permet une analyse rapide, détaillée et non invasive du flux sanguin cutané. Cet appareillage a été utilisé pour mesurer l'effet vasoactif d'un produit cosmétique sur un modèle d'inflammation cutané : l'érythème induit par le nicotinate de méthyle. Les mesures ont été réalisées sur dix volontaires sains, après conditionnement en atmosphère contrôlée, sur la face interne de leurs avant-bras, toutes les 10 minutes après application du vasodilatateur. Le produit cosmétique est efficace pour limiter l'inflammation provoquée par le nicotinate de méthyle. En effet, la diminution des valeurs maximales de perfusion sur le site testé est significativement plus marquée que sur le site témoin (53 % après 50 minutes). Des tests in vitro ont montré que la glycérine empèche la cristallization de la structure lipidique du Stratum Corneumà humidité ambiante basse. La question de savoir si ceci peut affecter la fonction barrière de la peau durant des applications répetées d'une crème pour peau normale contenant de la glycérine n'a pas été demontrée. Par conséquent, une crème contenant 20% de glycérine a été comparée avec son placebo dans un test bilatéral en double aveugle sur 17 volontaires en bonne santé. L'effet hydratant de la glycérine a étéévalué avec l'aide d'un cornéomètre, son influence sur la fonction barrière de la peau avec l'aide d'un évaporimètre (mesurant la Perte Trans-Epidermal en eau) et son effet protecteur après irritation á l'aide d'un agent de surface par la réponse biologique de la peau (mesure de la PTEL et du flux sanguin).
Dix jours de traitement sur une peau normale avec une crème contenant 20% de glycérine ont augmenté les mesures du cornéomètre de manière significative, indiquant une augmentation de l'hydration de la peau.
