Toxicologie génétiqueIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Objectifs : La surveillance de la toxicité subaiguë et chronique est essentielle lors du développement de nouvelles molécules cosmétiques. Compte tenu du manque de méthodes alternatives validées, nous avons développé un modèle de cytotoxicité à doses réitérées in vitro sur cellules TPH-1.
Methodes : Cultivées en suspension, les cellules ont été traitées avec des produits chimiques pendant 14 jours, à raison de 3 applications par semaine, et le taux de viabilité cellulaire a été déterminé par le test MTT. Nous avons commencé par étudier les effets à long terme de produits chimiques induisant différents degrés de cytotoxicité : le Paraquat, le 3-acide nitropropanoique (3-NPA) et le dodecyl sulfate de sodium (SDS). A partir d'une étude de cytotoxicité aigüe, des doses comprises entre 1 et 10 μg ml−1 ont donc été choisis pour réaliser notre étude de cytotoxicité subaigüe. Une étude comparative du potentiel genotoxique du Paraquat et du peroxyde d'hydrogène (H2O2) a été menée sur les cellules TPH-1. Plusieurs tests des comètes ont été réalisés : à 1 h (4°C), après une période de récupération de 24 h (37°C) et après un traitement de 14 jours (37°C). Une fois adapté pour tester des extraits végétaux ou de molécules fortement diluées, certains de nos produits cosmétiques ont été testé avec ce model.
Resultats : Comme pressentie, après 14 jours de traitement au Paraquat, le taux de viabilité cellulaire a fortement diminué pour des doses supérieures à 3 μg ml−1, alors que 10 μg ml−1 de 3-NPA ou de SDS n'induisaient pas plus de 44 % de mortalité cellulaire. Etonnamment, les résultats des tests des comètes ont montrés que les "tails moments" dus à l'H2O2 restent stables, alors que ceux liés au Paraquat augmentent de manière "dose-dépendante". D'autre part tous les composés testés entre 0.5 et 5 μg ml−1 ont été classés comme inoffensifs et ceci même avec un seuil placé à 90% de viabilité cellulaire.
Conclusions : En conclusion, ce test pourrait présenter un intérêt pour l'étude de cytotoxicité subaigüe et génotoxique des produits appliqués quotidiennement et suggère que le Paraquat en serait un contrôle positif de choix.Note de contenu : - MATERIALS AND METHODS : Cell culture maintenance - Cell viability assessment after acute cytotoxicity assay - Cell viability assessment after subacute cytotoxicity assay - Comet assay after 1h of exposure at 4°C - Comet assay after 24h or 14 days of exposure at 37°C - Statistical analysis

StatistiquesIndex. décimale : 668.5 Parfums et cosmétiques Résumé : OBJECTIF : Examen de la réparation de la barrière cutanée/processus de cicatrisation en utilisant un modèle d’équivalent de peau vivante (LSE) et l'imagerie par spectrométrie de masse assistée par désorption‐ionisation laser (MALDI‐MSI) pour identifier les lipides directement impliqués comme biomarqueurs potentiels. Ces biomarqueurs peuvent être utilisés par exemple pour déterminer si une plaie in vivo va guérir si elle est infectée.
METHODES : Un modèle de LSE in vitro a été blessé avec une lame de scalpel et analysé au jour 4 après la blessure par histologie et MALDI‐MSI. Ces échantillons de tissu étaient soit fixés au formol et inclus dans la paraffine (FFPE) pour l'histologie, soit congelés (FF) pour l'analyse MSI.
RESULTATS : L'utilisation d'un modèle de peau blessée in vitro couplé avec la MSI a permis l'identification des lipides impliqués dans le processus de réparation de la barrière cutanée/cicatrisation. La technique a permis de mettre en évidence les lipides directement au niveau de la plaie et de distinguer les différences de distribution des lipides entre l’épiderme et le site de la plaie.
CONCLUSION : Cette nouvelle méthode de couplage d'un modèle LSE in vitro avec la MSI a permis une analyse moléculaire approfondie de la réparation de la barrière cutanée/processus de cicatrisation. La technique a permis l'identification de lipides directement impliqués dans la réparation de la barrière cutanée/processus de cicatrisation, indiquant que ces biomarqueurs peuvent être potentiellement utilisés en clinique. Textiles et tissus à usages techniques -- Propriétés mécaniquesIndex. décimale : 668.4 Plastiques, vinyles Résumé : The demand for small-diameter blood vessel substitutes has been increasing due to a shortage of autograft vessels and problems with thrombosis and intimal hyperplasia with synthetic grafts. In this study, hybrid small-diameter vascular grafts made of thermoplastic polyurethane (TPU) and silk fibroin, which possessed a hybrid fibrous structure of an aligned inner layer and a random outer layer, were fabricated by the electrospinning technique using a customized striated collector that generated both aligned and random fibers simultaneously. A methanol post-treatment process induced the transition of fibroin protein conformation from the water-soluble, amorphous, and less ordered structures to the water-insoluble ?-sheet structures that possessed robust mechanical properties and relatively slow proteolytic degradation. The methanol post-treatment also created crimped fibers that mimicked the wavy structure of collagen fibers in natural blood vessels. Ultrafine nanofibers and nanowebs were found on the electrospun TPU/fibroin samples, which effectively increased the surface area for cell adhesion and migration. Cyclic circumferential tensile test results showed compatible mechanical properties for grafts made of a soft TPU/fibroin blend compared to human coronary arteries. In addition, cell culture tests with endothelial cells after 6 and 60 days of culture exhibited high cell viability and good biocompatibility of TPU/fibroin grafts, suggesting the potential of applying electrospun TPU/fibroin grafts in vascular tissue engineering. Tests de sécuritéIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - OBJECTIF : Optimiser les rendements d'extraction d'huile essentielle des fleurs de Fagraea fragrans Roxb. par hydro-distillation en utilisant une conception composite centrale (CCD) et d'évaluer ses activités biologiques pour la parfumerie et les applications cosmétiques.
- METHODES : La CCD a été appliquée pour étudier les influences des paramètres opérationnels [ratios pondéral eau/ fleurs (X1) et temps de distillation (X2)] sur les rendements d'huile essentielle (Y). Les compositions chimiques de l'huile essentielle extraite de l'état optimisé ont été identifiées par chromatographie en phase gazeuse-spectrométrie de masse. Les activités anti-oxydantes de l'huile essentielle ont été déterminées contre les radicaux ABTS • + et DPPH • et les effets cytotoxiques ont été évalués sur des cellules de rein embryonnaire humain (HEK293) par l'utilisation du dosage de MTT. En outre, les propriétés aromatiques de l'huile essentielle ont été évaluées par cinq volontaires sains formés.
- RESULTATS : Les meilleures conditions pour obtenir le rendement maximum d'huile essentielle étaient 7,5 mL g−1 (X1) et 215 min (X2). Le rendement expérimental de l'huile essentielle (0,35 ± 0,02% v/w) était proche de la valeur prédite par un modèle mathématique (0,35 ± 0,01% v / w). 3-Octadecyne, Z, Z, Z-7,10,13-hexadecatrienal, E-nérolidol, pentadecanal, et le linalol sont les principaux constituants de l'huile essentielle. L'huile essentielle a montré les capacités anti-oxydantes modérées avec aucun effet toxique sur les cellules HEK293 entre 1-250 ug mL−1. En outre, l'huile essentielle présentait un arôme très fort et a été classé pour être haut en notes moyennes.
- CONCLUSION : Les résultats offrent les conditions effectivement opérationnelles dans l'extraction de l'huile essentielle de F. fragrans utilisant l'hydro-distillation.
