Travail de rivière (cuir)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : In this study, a metalloproteinase gene from Planococcus halotolerans SCU63 was cloned and expressed in Bacillus sp. SCK6. LB broth medium was used as the initial medium and for a series of fermentation optimisations. The most suitable medium components were as follows (g/l) : yeast extract powder (20), casein (13), tryptone (7), NaCl (10), urea (8). When the inoculation amount was 2%, the filling amount was 100mL, the culture time was 370C, and the pH was 7.0 for 40 hours the protease production reached the highest. The enzyme activity was completely inhibited by EDTA. Surfactants, such as Tween 20, Tween 80, and TritonX-100, could improve the enzyme activity, while SDS could inhibit the enzyme activity. Under protease action, goatskin can be completely depilated at 37oc and 28 hours. Compared with the conventional chemical dehairing, the enzymatic dehairing opened up the goatskin elastic fibre without any hair root in the hair follicle, and the skin was of high quality. HexamidineL'hexamidine, (hexaméthylènedioxy)4,4'dibenzamidine bis-(hydroxy-2 éthanesulfonate), Numérotation CAS : 3811-75-4, est une molécule utilisée en médecine pour ses propriétés antiseptiques. Elle est présente dans un certain nombre de préparations pharmaceutiques utilisées pour la désinfection de plaies non ouvertes, dans certains collyres et pulvérisations.
Cet antiseptique, utilisé sur peau saine, n'est pas retrouvé sous la couche cornée.
De nombreuses préparations à base d'hexamidine sont vendues sous divers noms commerciaux, la plus connue étant Hexomédine qui a été commercialisée en France à partir de 1949 sous diverses formes 3 : solution hydroalcoolique à 0,1 % pour applications locales, pommade et gel, poudre pour solution aqueuse, collutoire.
L'Hexomédine transcutanée 0,15 % est un antiseptique local sous forme de solution. Il est utilisé dans le traitement des infections de la peau et des ongles.
Ce composé est un agent tensioactif, c’est-à -dire un savon, qui tue les bactéries de type Gram+. Il est aussi utilisé dans un certain nombre de produits cosmétiques comme conservateur.
L'hexamidine peut provoquer une dermite de contact. Il est considéré comme non toxique dans les cosmétiques pour des concentrations de 0,03 % à 0,1 %.

SérineLa sérine est un acide-α-aminé en C3, homologue hydroxylé de l'alanine.
C'est un acide aminé aliphatique hydroxylé, dont le nom en nomenclature systématique est acide 2-amino-3hydroxypropanoïque. Il est présent dans la plupart des protéines et c'est l'un des 20 acides aminés naturels les plus courants sur terre. C'est un acide aminé polaire, faiblement acide à cause de sa fonction alcool primaire en beta. Le groupement OH de la chaîne latérale peut être estérifié en présence d'un groupement phosphate.
La sérine a pour codons : AGU, AGC, UCA, UCU, UCG, UCC.
La D-Sérine, synthétisée par la sérine racémase à partir de L-serine, sert de signal neuronal en activant le récepteur NMDA dans le cerveau.Index. décimale : 668.5 Parfums et cosmétiques Résumé : HtrA1 is serine protease which has been identified in the epidermis. Although the precise function of HtrA1 in the epidermis is still unclear, some reports have suggested that HtrA1 is involved in programmed cell death, apoptosis and anoikis.
On the other hand, it is generally known that excess apoptosis caused by UVB exposure is involved in the accumulation of cell damage in the skin epidermis, and it adversely affects skin epidermis homeostasis that causes chronic cell inflammation and parakeratosis. However, the precise mechanism underlying how UVB induced apoptosis adversely affects epidermal alterations has not been elucidated. This study aims to clarify the behaviour of HtrA1 in the apoptosis of epidermal cells induced by UVB exposure. Enzymes protéolytiquesUne enzyme protéolytique est une enzyme capable de couper une protéine en plusieurs fragments ou peptides. La trypsine, la papaïne, la pepsine, la chymotrypsine, la plasmine, la subtilisine... sont capables de couper une protéine, chaque enzyme étant spécifique de certains sites particuliers de cette protéine. C'est ainsi, par exemple, qu'une immunoglobuline G est découpée par la papaïne en un fragment Fc et deux fragments Fab, comme l'a montré Porter en 1959.

SurfactantsIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Most proteases exhibit broad-spectrum activities to all of the protein components of skins, thus, the indiscriminate application of proteases will bring out undue and non-selective loss of skin structural proteins, especially collagen and elastin, and lead to loose and damaged grain. Finding the effective ways to control proteases’ activities against collagen and elastin in leather processes is very important. In this work, the influence of typical surfactants on the collagenolytic and elastinolytic activities of frequently used proteases was investigated, and the mechanism was also discussed. The results indicated that the nonionic surfactants had slight effects and anionic surfactants exhibited quite different behaviors on the collagenolytic and elastinolytic activities of selected proteases. Both collagenolytic and elastinolytic activities of trypsin preparations were obviously inhibited by all selected anionic surfactants, especially SDS and SDBS. For the bacteria proteases, their elastinolytic activities were significantly activated and their collagenolytic activities were inhibited to a variable extent by anionic surfactants. Tanners may effectively control the selective action of proteases to collagen and elastin to achieve different requirements through correctly utilizing surfactants.
