CollagénasesLes collagénases sont des enzymes capables de rompre les liaisons peptidiques du collagène. Elles facilitent la destruction des structures extracellulaires lors de la pathogenèse bactérienne. Ce sont des exotoxines.
La production de collagénases peut être induite lors d'une réponse immunitaire, par les cytokines qui stimulent les cellules telles que les fibroblastes et les ostéoplastes et occasionnent indirectement des lésions tissulaires.

Matrix metalloproteinases and bacterial collagenases are two kinds of collagenases that can cleave the triple-helical domain of native fibrillar collagens. In this paper, the family members and domain composition of matrix metalloproteinases and bacterial collagenases are summarized. The catalytic mechanism of collagen hydrolysis by collagenases is described, and the methods adopted to date for investigating and regulating collagenases and their inhibitors are reviewed. Furthermore, the applications of collagenases and their inhibitors in biomedicine, food processing and the enzymatic unhairing process in the leather-making industry are presented. Wet-blue (tannage)Peau tannée au chrome (le chrome donne une couleur bleue)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : In this study, alkaline metalloprotease gene 1067 was cloned from Planococcus halotolerans SCU63T and heterologously expressed in Bacillus subtilis SCK6. Using Luria Bertani (LB) broth medium as the initial medium, the optimal medium was obtained through a series of fermentation and culture optimization (g/L): yeast extract (10), soybean powder (15), urea (20), potassium chloride (6.7), calcium chloride (13.3), NaCl (10). On the basis of the optimal medium, the highest enzymatic activity of 1259.21 U/mL could be obtained by culturing at 30°C for 40 h. with pH 8, inoculation amount of 4% and filling amount of 50 mL. EDTA inhibits protease activity and PMSF promotes it, indicating that it was a metalloprotease rather than a serine protease. The optimum reaction temperature of the protease is 70°C, and the optimum pH is 9. The metal ions Zn 2+, Co2+ and surfactant β-ME, Tween 80 can improve the activity of the protease. The results of unhairing, SEM and staining show that this metalloprotease can completely dehair goatskin. Compared with the conventional chemical method, the goatskin after enzymatic unhairing has softer texture and smoother surface, and there is no obvious damage to the goatskin. SCK6. LB broth medium was used as the initial medium and for a series of fermentation optimisations. The most suitable medium components were as follows (g/l) : yeast extract powder (20), casein (13), tryptone (7), NaCl (10), urea (8). When the inoculation amount was 2%, the filling amount was 100mL, the culture time was 370C, and the pH was 7.0 for 40 hours the protease production reached the highest. The enzyme activity was completely inhibited by EDTA. Surfactants, such as Tween 20, Tween 80, and TritonX-100, could improve the enzyme activity, while SDS could inhibit the enzyme activity. Under protease action, goatskin can be completely depilated at 37oc and 28 hours. Compared with the conventional chemical dehairing, the enzymatic dehairing opened up the goatskin elastic fibre without any hair root in the hair follicle, and the skin was of high quality. Gallate d'épigallocatéchineLe gallate d'épigallocatéchine (EGCG) est l'ester d'épigallocatéchine et d'acide gallique. C'est le flavanol le plus abondant du thé, connu pour être un puissant antioxydant. On le trouve essentiellement dans le thé blanc, le thé vert et, dans une moindre mesure, le thé noir — lors de la production de ce dernier, les flavanols sont convertis essentiellement en théaflavines et en théarubigines2. On le trouve également dans de nombreux légumes, dans les fruits à coque ainsi que dans la poudre de caroubier à un taux de 1,09 mg·g-1.

Thé vert et constituantsIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Les plantes ayant des propriétés antioxydantes sont bénéfiques pour prévenir les événements de vieillissement induits par la lumière UV. De nombreux produits à base de Camellila sinensis (thé vert) sont disponibles dans le commerce, dont un grand nombre prétendant contenir des composés bioactifs qui permettraient d'éviter des lésions cutanées provoquées par les UV. Dans cette étude, nous avons testé l'efficacité de cinq extraits de thé vert commercialisés, utilisés pour enrichir les formulations cosmétiques pour la protection des fibroblastes humains et murins contre les effets de rayonnement UV, et comparés par rapport à un extrait fluide, celui préparé selon les recommandations de la pharmacopée brésilienne. En tenant compte du fait que le processus de vieillissement peut être accéléré par le rayonnement solaire excessif par la génération de radicaux libres, conduisant à lʇépuisement des défenses antioxydantes de la peau, et à son effondrement causé par une perturbation du métabolisme des métalloprotéinases, nous avons utilisé leur teneur individuelle en EGCG, l'état de la catalase et de la SOD et le niveau des MMP-1, -9, -13 comme paramètres de comparaison. Les teneurs en EGCG des produits commerciaux ont montré une grande variabilité, allant de niveaux indétectables jusqu'à 58.65 ± 1,12 μg mL−1, contrairement à l'extrait fluide (87,82 ± 1,35 μg mL−1). En outre, seul l'extrait selon la pharmacopée a été en mesure de réduire considérablement la dégradation MMP tout en améliorant les niveaux de SOD et de la catalase. Ces résultats indiquent, pour la première fois, que les méthodes de préparer des mélanges à base de plantes peuvent interférer de façon significative avec des composés doués d' effets photoprotecteurs; l'efficacité des produits contenant des extraits de C. sinensis sensés agir contre les effets du rayonnement solaire peut donc être compromise. Ethylène diamine tétra acétiqueL'EDTA (Ethylène Diamine Tétra Acétique), ou acide éthylène diamine tétraacétique, est un acide diaminotétracarboxylique de formule C10H16N2O8.
L'EDTA comporte six sites basiques, quatre correspondant aux bases conjuguées (carboxylates) des fonctions carboxyliques et deux correspondant aux fonctions amines. Ces sites basiques sont également des centres ligands, faisant de l'EDTA un ligand hexadentate (ou parfois tétradentate, lorsque seuls les sites carboxyliques sont utilisés). C'est d'ailleurs sa principale caractéristique, son fort pouvoir chélatant (ou complexant) par lequel il forme des complexes métalliques très stables, ce qui en fait un traitement en cas d'intoxication aux métaux lourds comme le plomb, avec une concentration adaptée (Voir Applications et chélation). Dans les complexes, l'EDTA est lié aux cations métalliques sous la forme d'une de ses bases conjuguées9.

However, the main difficulty which hinders the utilization of protease in unhairing is the injury to skin caused by the protease. In this article, a collagenase was purified from the protease (2709). The particular collagenase is a metalloprotease which is inhibited by EDTA. On the other hand, 2709 itself is a serine protease which is insensitive to EDTA. A way to reduce the injury to skin was put forward. When 10mM EDTA is added to crude enzyme solutions of 2709, the protease (2709) retains 90.5% of the proteolytic activity, and the unhairing capacity of 2709 has little loss. AI the same time, 2709 loses 66.9% of the collagenolytic activity since the collagenase is inhibited by EDTA. Therefore, the injury to skin will be reduced.
