PicklageLe picklage consiste à faire absorber à la peau en tripe une quantité importante d'acide, en présence de sel neutre (NaCl) pour réprimer le gonflement que provoquerait l'acidité du milieu.
TrypsineLa trypsine (EC 3.4.21.4) est une enzyme digestive du suc pancréatique qui a pour rôle de digérer les protéines.
Elle est synthétisée par le pancréas sous forme de trypsinogène (proenzyme inactive), puis stockée dans les vésicules enzymatiques des cellules acineuses d'où elle est excrétée au moment de la digestion. L'activation du trypsinogène en trypsine est le résultat de l'hydrolyse d'un propeptide sous l'action de l'entérokinase ou par un effet d'autoactivation de la trypsine par elle-même. La cholecystokinine-pancréozymine active la sécrétion des enzymes (donc de la trypsine) dans le suc pancréatique.
La trypsine est une endoprotéase qui hydrolyse les liaisons peptidiques dans lesquelles un acide aminé basique (Lys-|-Xaa ou Arg-|-Xaa) engage sa fonction acide (sauf dans le cas où l'acide aminé suivant (schématisé ici par "Xaa") est une Proline). Elle coupe en C-terminal de ces acides aminés. En d'autres mots, elle transforme les chaînes polypeptides en chaînes protéiques plus courtes pour permettre la digestion. Efficace à pH 7,5 - 8,5, elle est inactivée et digérée en quelques heures à pH neutre (=7) dans l'intestin.
La trypsine participe à l'activation d'autres enzymes comme l'alpha-chymotrypsine par coupure hydrolytique de la chaîne polypeptidique du chymotrypsinogène.
Cette enzyme sert également lors de la 2e semaine du développement embryonnaire humain. Elle est sécrétée par le trophoblaste afin de digérer la zone pellucide entourant le blastocyste. Ce phénomène s'appelle l'éclosion.Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : Traditionally, universally used pelt bating technologies rely on the application of trypsin, neutral and alkaline microbial proteases but suffer from complicated operation, limited bating efficiency and unsatisfactory leather performance. Therefore, devising a new pelt bating approach to achieve high bating efficiency and excellent leather performance has always been wished for by the leather industry. To pursue this goal, years of persistent research work enabled us to develop a novel approach for pelt bating by means of acidic proteases in pickling process. Initially, basic enzymatic characteristics and bating effectiveness of several typical acidic proteases in pelt pickling medium were investigated; then, the bating effectiveness through the quantitative characterization of protease activity of the optimal acidic protease was compared with that of the conventional bating enzyme. The results indicated that all of the selected acidic proteases had good salt-tolerance and exhibited optimum activity at pH 3.0–4.0. The novel pickling-bating method based on microbial origin acidic protease L80A led to an outstanding performance on pelt bating at the dosage of 150 U/mL of collagenolytic activity. The bating effectiveness of acidic protease L80A was comparable to and even better than that of trypsin BEM due to its moderate proteolytic ability. Moreover, the deep and even penetration of acidic protease in the pelt permitted it to produce soft, organoleptically stable and overall better quality crust leather than that of the conventional trypsin bating method. Additionally, pelt bating was performed along with the pickling process without extra inactivation and washing operation, making the bating operation more efficient, economical, and environment friendly. Results had made us to conclude that this cutting-edge acidic proteases based pickling-bating method could be the first step/ way forward to replace the decades-old traditional pelt bating technology. Bating effectiveness is determined not only by the properties but also by the permeation behavior of the proteases. Imperfect methods to control protease permeation often results in uneven distribution of enzyme proteins in the pelts, leading to excessive enzymolysis of the surface layer and inadequate opening-up of the inner layer. In this study, the relative size of proteases and delimed pelts were analyzed, the permeation behavior of fluorescein-labeled proteases in the pelt was observed using a confocal laser scanning microscope (CLSM), and the effect of electrostatic interaction between protease and collagen proteins on the permeation of protease into the pelt was investigated. The results showed that, after dehairing, liming and deliming operations, the enzyme can easily permeate into the pelts due to the formation of large cavities and interfibrillar gaps. The permeation of protease within the delimed pelt is significantly influenced by the affinity (electrostatic interactions) between the collagen and protease proteins. The isoelectric point (pI) of the protease protein, the collagen and the pH of the solution directly influence the electrostatic properties and interactions. When the enzyme and collagen are similarly charged (electrostatic repulsion), the enzyme can easily permeate into the pelts; when the enzyme and collagen are oppositely charged (electrostatic attraction), the permeation of the enzyme into the inner layer is difficult, resulting in the accumulation of protease on the grain and excessive hydrolysis of the grain layer. Therefore, the established permeation regulation mechanism of protease based on electrostatic interactions between enzyme and collagen could serve as an important basis for the selection of protease and the regulation of the enzymatic bating process. Point isoélectriqueEn biochimie, le point isoélectrique (pI) ou potentiel hydrogène isoélectrique (pHI) est le pH auquel une molécule est sous forme d'ion mixte ou, en physico-chimie, le pH d'une solution aqueuse dans laquelle un solide existe sous un potentiel électrique neutre.
En physico-chimie : Selon Bolger, le caractère acide ou basique d'une surface s'exprime par son point isoélectrique " Is ou IEPS (Iso Electric point for the surface) " ou point de charge nulle " PCN ou PZC (Point of Zero Charge) ", défini comme étant le pH de la solution aqueuse dans laquelle le solide existe sous un potentiel électrique neutre. Si le pH de la solution est basique, la surface est acide, et inversement. La différence entre le PZC et l'IEPS est basée sur le phénomène d'adsorption spécifique. On peut considérer que si la grandeur mesurée ne dépend pas de la solution utilisée pour la mesurer (pH, concentration, nature des ions), alors on a affaire à un PZC. Dans le cas contraire, c'est un IEPS que l'on mesure. Par exemple, quand la mesure de goutte sessile à deux liquides est utilisée, on considère en général qu'il n'y a pas adsorption des ions de cette goutte et que la goutte déplace complètement l'alcane qui sert de deuxième liquide: on est alors en présence d’un PZC. Au contraire, dans les mesures de potentiel d'écoulement (streaming potential), la solution joue un rôle important et c'est un IEPS que l'on mesure. Enfin, la charge nette se définit grâce au pH de la solution aqueuse dans laquelle la surface métallique existe, dans un état électriquement neutre (c’est-à -dire [M-OH2+ surf]=[M-O- surf]) et au PZC.

- Si pH > PZC alors la charge nette est négative.
Il existe plusieurs méthodes expérimentales permettant de décrire l’état acido-basique de la surface : la mesure du potentiel d’écoulement, la photoélectrochimie, la mesure de l’angle de contact, et la spectroscopie XPS.

A thorough understanding of the basic characteristics of typical acidic proteases and the interaction between enzyme proteins and wet blue fibers will help to improve bating technology for wet blue by selecting the suitable proteases. In this paper, the enzymatic characteristics, molecular weight (Mr) and isoelectric point (pI) of several proteases and their bating effectiveness were investigated. The results indicated that there are two main factors which may affect the wet blue bating effectiveness of acidic proteases. First, the common acidic proteases exhibited low activity towards chrome-tanned collagen fiber which lead to inefficient bating effect through normal dosage. Nonetheless, when the dosages of chrome-tanned collagen fiber activity reached up to 50 U/mL, these acidic proteases also can achieve a good bating effect, the caseinolytic activity has been reached up to 1000 U/mL-4000 U/mL. Second, because of the large molecular weight and the charge repulsion between enzyme proteins and wet blue fibers, the enzymatic hydrolysis process, the penetration and distribution of acidic protease proteins, into wet blue is very difficult. Additionally, neutral proteases have more prospects in wet blue bating process due to the higher chrome-tanned collagen fiber activity and less charge repulsion than acidic proteases. PerméationIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Enzymes have been widely used as alternatives to conventional chemicals in cleaner leather processes due to their advantages of meeting increasing environmental demands. Especially, enzymatic unhairing based on protease has been applied to leather-making for a long time, however, it still has the key problem of slow permeation in the animal hide, resulting in slow unhairing rate, poor hair removal effect, excessive proteolysis of hide collagen, and decreased leather quality. Aiming at the key problem of bio-unhairing technology, fluorescent labeling technique and confocal laser scanning microscopy were used to investigate the protease permeation behaviors into the animal hide based on well-prepared labeled proteases, as well as the quantitative analysis of the protease amount in different hide layers. The results show that the protease mainly permeates the bovine hide through transfollicular routes from the hair side, and although the intercellular-interfibrillar pathway also exists, it does not play an important role. Additionally, the protease permeation behaviors into the hide are greatly impacted by the charge states of the hide and protease proteins, depending on the isoelectric points (pI) of the proteins and solution pH values. When the solution pH is not between the pI values of the two proteins, the similarly charged protease can quickly and deeply penetrate the hide because of the electrostatic repulsion. The established mechanism provides a theoretical basis for developing an efficient enzymatic unhairing technology for leather-making, and this can also be applied to other processes involving the enzyme permeation into the hide or leather. GlycosidasesLes Glycoside hydrolases (ou glycosidases) catalysent l'hydrolyse de liaisons glycosidiques et libèrent au moins un composé osidique. Ce sont des enzymes extrêmement communes, dont les rôles sont aussi divers que la dégradation des glucides complexes (cellulose et hémicellulose), les défenses anti-bactériennes (ex. le lysozyme), la pathogénicité (ex. les neuraminidases virales) ou le métabolisme cellulaire (amylases ou enzyme de débranchement du glycogène pour le métabolisme énergétique, mannosidases comme modulateur de glycosylation des protéines, ...). Les glycosyltransférases et les glycosides hydrolases sont les deux classes majeures d'enzymes impliquées dans la synthèse et la rupture des liaisons osidiques.

ProtéoglycanesUn protéoglycane est une glycoprotéine, combinaison d'une protéine et d'un glycosaminoglycane (GAG). L'association entre les deux types de chaîne s'effectue essentiellement dans l'appareil de Golgi, mais également au niveau du réticulum endoplasmique d'une cellule. La proportion de glucides des protéoglycanes peut atteindre 95 %. Ceux-ci se présentant sous la forme d'une ou plusieurs chaînes de glycosaminoglycanes non ramifiées. Les chaînes de sucres sont très longues mais pas ramifiées. Ils sont O-glycosylés, se lient à l'acide aminé sérine à l'extrémité OH. Les protéoglycanes peuvent être soit transportés à l'extérieur de la cellule par exocytose (s'intégrant alors à la matrice extracellulaire sous forme de chondroïtine-sulfate, kératan-sulfate, héparan-sulfate, dermatan-sulfate, etc.), soit entrer dans la constitution de la membrane plasmique ou du glycocalyx, jouant alors un rôle dans les relations cellule-matrice.
Les PG (protéoglycanes) ont des compositions et poids moléculaire très variés et sont hétérogènes au niveau de leur structure et de leur fonction.
Les protéoglycanes sont des composants essentiels de la matrice extracellulaire. Ce sont des pièges à eau qui sont importants pour les propriétés mécaniques des tissus cartilagineux par exemple. Les héparan sulfates peuvent avoir un rôle dans la signalisation : ce sont des co-récepteurs pour les FGF (Fibroblast Growth Factor). Les protéoglycanes jouent aussi un rôle dans la diffusion des molécules de signalisation (Wnt, Shh), ou bien en interagissant avec des inhibiteurs (Noggin). Glycosidases have been used in leather making processes to reduce pollutants and improve leather quality. But the selection of glycosidases is still blind because the related mechanisms are not well understood yet. Hence, the animal skin structures and glycoconjugates components, the advances in the methods and mechanisms of removing glycoconjugates related to unhairing and fiber opening-up in leather manufacture, the kinds, compositions, structures and functions of typical glycoconjugates in skin are summarized. Then the approaches to destroy them, possible glycosidases suitable for leather making and their acting sites are analyzed based on the recognition of glycoconjugates in skin and the specificities of glycosidases toward substrates.
