CéramidesUn céramide est un sphingolipide résultant de la combinaison d'un acide gras avec la sphingosine via une liaison amide. On trouve de telles molécules en abondance dans les membranes cellulaires, où elles entrent notamment dans la constitution des sphingomyélines. Les céramides ne jouent pas qu'un rôle structurel dans les membranes biologiques, et peuvent également revêtir des fonctions de signalisation lipidique. Leurs actions les mieux comprises vont de la différenciation cellulaire à la mort cellulaire programmée (apoptose) en passant par la prolifération cellulaire.
Les acides gras qui les constituent ont généralement un nombre pair (de 16 à 24) d'atomes de carbone, sont saturés ou monoinsaturés, et sont souvent α-hydroxylés.
La plupart des céramides n'existent pas à l'état naturel si ce n'est comme précurseurs de la biosynthèse des sphingolipides par une réaction d'addition sur la fonction alcool primaire de la sphingosine. (Wikipedia)

SérineLa sérine est un acide-α-aminé en C3, homologue hydroxylé de l'alanine.
C'est un acide aminé aliphatique hydroxylé, dont le nom en nomenclature systématique est acide 2-amino-3hydroxypropanoïque. Il est présent dans la plupart des protéines et c'est l'un des 20 acides aminés naturels les plus courants sur terre. C'est un acide aminé polaire, faiblement acide à cause de sa fonction alcool primaire en beta. Le groupement OH de la chaîne latérale peut être estérifié en présence d'un groupement phosphate.
La sérine a pour codons : AGU, AGC, UCA, UCU, UCG, UCC.
La D-Sérine, synthétisée par la sérine racémase à partir de L-serine, sert de signal neuronal en activant le récepteur NMDA dans le cerveau.Index. décimale : 668.5 Parfums et cosmétiques Résumé : Objective : A higher trans-epidermal water loss (TEWL) occurs in rough skin, in elder skin and also in atopic dermatitis. An impaired skin barrier function is considered to be caused by an incomplete construction of the intercellular lamellar structure due to the quantitative reduction of ceramides. Since these symptoms coexist with oxidative stress, we hypothesized that impairment of the skin barrier function is accelerated by oxidative stress. Thus, the purpose of this study was to clarify the effect of oxidative stress on ceramide synthesis and to characterize whether antioxidants can improve skin barrier function. 3-O-Laurylglyceryl ascorbate (VC-3LG), which is a newly amphipathic derivative of ascorbic acid, was evaluated as a candidate antioxidant.
Methods : We characterized the mRNA expression levels of serine palmitoyltransferase (SPT) in normal human epidermal keratinocytes (NHEKs) treated with H2O2 using real-time PCR analysis. In order to evaluate the effect of VC-3LG on skin barrier function, we used several assays with reconstructed human epidermis equivalents (RHEEs).
Results : Ceramide synthesis was down-regulated in NHEKs by oxidative stress. Treatment with VC-3LG abrogated the down-regulation of SPT mRNA in NHEKs caused by oxidative stress, and stimulated SPT mRNA expression levels. In experiments characterizing the antioxidative properties of VC-3LG, VC-3LG reduced oxidative stress in NHEKs by up-regulating catalase mRNA expression. In addition, VC-3LG stimulated the skin barrier function in RHEEs, which had lower TEWL values compared with untreated RHEEs. Furthermore, VC-3LG increased the quantity of ceramide in RHEEs.

RésorcinolLa résorcine (ou résorcinol, ou benzène-1,3-diol en nomenclature IUPAC) est l'isomère méta du benzènediol. C'est un diphénol (C6H4(OH)2) utilisé comme antiseptique et pour la préparation de certains colorants : bleu de résorcine, vert de résorcine. Le terme est composé à partir de résine et d'orcine.
PROPRIETES CHIMIQUES, TOXICITE :
Le résorcinol donne des réactions de condensation avec de nombreux composés organiques. On obtient, par exemple, par condensation d'alcools primaires avec le résorcinol des benzéines et des naphtaléines. Ces composés, en solution alcaline, présentent une fluorescence qui varie en fonction du nombre et de la position des groupements -OH et -COOH.
Il est utilisé, comme réactif très sensible, pour la recherche de quantités minimes d'acide tartrique et de zinc. Il peut irriter la peau et les muqueuses. Son absorption peut provoquer des troubles sanguins, des convulsions et même la mort.
UTILISATION :
Le résorcinol, obtenu industriellement par fusion de l'acide métabenzène disulfonique avec un excès de soude a de nombreuses utilisations industrielles : tannage, fabrication de résines
Notamment comme colle en construction navale bois,contreplaqué marine,du fait de sa bonne tenue à l'eau,remplacée à l'heure actuelle par les résines époxy : fabrication d'adhésifs, préparation d'acide, para-aminosalicylique, fabrication d'explosifs et de colorants,
cosmétiques, teinture et impression des tissus, antiseptique, antifongique et desséchant.
TyrosinaseIndex. décimale : 668.5 Parfums et cosmétiques Résumé : OBJECTIF : Le 4-n-butylresorcinol est un inhibiteur compétitif de la tyrosinase et a été utilisé en tant qu'agent anti-mélanogénique. Cependant, son mécanisme d'inhibition dans des cellules intactes n'a pas encore été complètement compris. Pour élucider le mécanisme cellulaire, nous avons comparé in vitro et in vivo les effets inhibiteurs de 4-n-butylresorcinol sur l'activité de la tyrosinase.
METHODES : Des cellules de mélanome B16F10 ont été cultivées dans un milieu contenant α-MSH en présence ou en absence de 4-n-butylresorcinol. Les niveaux d’ARNm de tyrosinase, les taux des protéines et de l'activité dans les cellules B16F10 ont été comparés par PCR en temps réel, par immunocoloration et combinés avec transfert de Western et l'analyse colorimétrique, respectivement. La concentration de mélanine a été mesurée par colorimétrie à la fois dans les cellules et dans les médias. La glycolysation de la tyrosinase et la dégradation protéolytique ont été analysées par immunoblot après que les cellules ont été traitées avec Endo H/E64/PNGase et les inhibiteurs du protéasome, respectivement.
RESULTATS : Le 4-n-butylresorcinol inhibe l'activité de la tyrosinase et la synthèse de mélanine plus efficacement dans des cellules intactes que dans les lysats cellulaires. Le transfert de Western en temps réel et la RT-PCR ont montré que le 4-n-butylresorcinol réduit les taux des protéines, mais pas le taux d’ARNm de tyrosinase dans des cellules B16F10. Le 4-n-butylresorcinol n'a montré aucun effet sur le processus de la glycosylation de la tyrosinase ou sur le trafic des mélanosomes. Cependant, le traitement des cellules B16F10 avec E64 ou avec l'inhibiteur de protéasome a abrogé la diminution de la tyrosinase induite par le 4-n-butylresorcinol. Par ailleurs, le 4-n-butylresorcinol active la p38 MAPK, ce qui entraîne une augmentation de l'ubiquitination de la tyrosinase.
CONCLUSION : Le 4-n-butylresorcinol inhibe la mélanogénèse en améliorant la dégradation protéolytique de la tyrosinase, ainsi que via la liaison compétitive à la tyrosinase. The goal of the current work is to demonstrate L-4-thiazolylalaine (L4, Protinol), a non-proteinogenic amino acid shown to stimulate the production of dermal proteins by fibroblasts, is an alternative efficacious topical ingredient for visible signs of ageing.
- Methods : In vitro studies using 3D human skin tissue models were performed to show changes in protein and gene expression of key dermal markers in samples treated with 0.3% L4 compared to vehicle control. In vivo evaluation of skin turnover was measured in volunteers after treatment with L4 compared to retinol. Skin biopsies (n = 30) were taken to investigate epidermal and dermal changes in cases treated with L4 and compared to retinol. Finally, a clinical evaluation (n = 28) was conducted to assess the efficacy of L4 over a base formulation using various ageing parameters within a population of women 46–66 years old with mild-to-moderate wrinkles.
- Results : In vitro studies on 3D tissues displayed significant changes in the dermal matrix via an increase in HA and pro-collagen I production and a decrease in the expression of inflammatory genes. In vivo biopsy studies demonstrated that L4 and retinol independently increased epidermal thickness and collagen remodelling significantly more compared with the base formula. Clinical evaluation showed firmer and smoother skin at day 28 post-treatment with L4 over the vehicle control without causing side effects such as redness or irritation.

Rayonnement ultravioletIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : Recent research has shown that significant levels of cyclobutane pyrimidine dimers (CPDs) in DNA continue to form in melanocytes for several hours in the dark after exposure to ultraviolet radiation (UVR) ends. We document the utility of a new multifunctional ingredient, 3-(4-hydroxy, 3-methoxybenzyl)-pentane-2,4-dione (INCI acetyl zingerone (AZ)), to protect melanocytes against CPD formation after UVR exposure ends.
- Methods : The use of AZ as an intervention to reduce CPD formation after irradiation was assessed in vitro by comparing kinetic profiles of CPD formation for several hours after irradiation in cells that were untreated or treated with AZ immediately after irradiation. Multifunctional performance of AZ as an antioxidant, quencher and scavenger was established using industry-standard in vitro chemical assays, and then, its efficacy in a more biological assay was confirmed by its in vitro ability to reduce intracellular levels of reactive oxygen species (ROS) in keratinocytes exposed to UVA radiation. Molecular photostability was assessed in solution during exposure to solar-simulated UVR and compared with the conventional antioxidant a-tocopherol.
- Results : Even when added immediately after irradiation, AZ significantly inhibited ongoing formation of CPDs in melanocytes after exposure to UVA. Incubation with AZ before irradiation decreased intracellular levels of UVA-induced ROS formation in keratinocytes. Compared with a-tocopherol, the molecular structure of AZ endows it with significantly better photostability and efficacy to neutralize free radicals (.OH, .OOH), physically quench singlet oxygen (1O2) and scavenge peroxynitrite (ONOO-).

Hyaluronique, acideL'acide hyaluronique est un type de polysaccharide (plus précisément une glycosaminoglycane) non fixé à une protéine centrale et largement réparti parmi les tissus conjonctifs, épithéliaux et nerveux animaux.
Il se trouve notamment dans l'humeur vitrée et le liquide synovial. Il est l'un des principaux composants de la matrice extracellulaire ainsi que de certaines mucoprotéines lorsqu'il est associé à une fraction protéique.
Depuis les années 1990, il est très utilisé dans divers dispositifs médicaux, médicamenteux et cosmétiques (présenté dans ce dernier cas comme "antistatique, humectant, hydratant, conditionneur cutané, anti-âge, etc."), bénéficiant d'un effet de mode grandement soutenu par une large publicité commerciale.
Contribuant de façon significative à la prolifération et à la migration des cellules, l'acide hyaluronique est aussi impliqué dans la progression de certains cancers. (Wikipedia)
