We have focussed particularly on the biochemical and cellular gradients of the stratum corneum (SC) with the aim of discovering new skin moisturization and SC maturation mechanisms, identifying new technologies and/or providing conceptual innovations for ingredients that will improve our understanding and treatment of dry skin. Specifically, we discuss gradients for corneodesmosomes and proteases, corneocyte phenotype-inducing enzymes, filaggrin and natural moisturizing factor (NMF), and barrier lipids. These gradients are interdependent and influence greatly corneocyte maturation. The interrelationship between corneodesmolysis and the covalent attachment of ω-hydroxy ceramides and ω-hydroxy fatty acids to the corneocyte protein envelope forming the corneocyte lipid envelope is especially relevant in our new understanding of mechanisms leading to dry skin. This process is initiated by a linoleoyl-ω-acyl ceramide transforming enzyme cascade including 12R lipoxygenase (12R-LOX), epidermal lipoxygenase-3 (eLOX3), epoxide hydrolase 3 (EPHX3), short-chain dehydrogenase/reductase family 9C member 7 (SDR9C7), ceramidase and transglutaminase 1. Our research has opened the opportunity of using novel treatment systems for dry skin based on lipids, humectants, niacinamide and inhibitors of the plasminogen system. It is clear that skin moisturization is a more complex mechanism than simple skin hydration. CéramidesUn céramide est un sphingolipide résultant de la combinaison d'un acide gras avec la sphingosine via une liaison amide. On trouve de telles molécules en abondance dans les membranes cellulaires, où elles entrent notamment dans la constitution des sphingomyélines. Les céramides ne jouent pas qu'un rôle structurel dans les membranes biologiques, et peuvent également revêtir des fonctions de signalisation lipidique. Leurs actions les mieux comprises vont de la différenciation cellulaire à la mort cellulaire programmée (apoptose) en passant par la prolifération cellulaire.
Les acides gras qui les constituent ont généralement un nombre pair (de 16 à 24) d'atomes de carbone, sont saturés ou monoinsaturés, et sont souvent α-hydroxylés.
La plupart des céramides n'existent pas à l'état naturel si ce n'est comme précurseurs de la biosynthèse des sphingolipides par une réaction d'addition sur la fonction alcool primaire de la sphingosine. (Wikipedia)
kératineLa kératine est une protéine, synthétisée et utilisée par de nombreux êtres vivants comme élément de structure, et également l'exemple-type de protéine fibreuse.
La kératine est insoluble, et peut être retrouvée sur l'épiderme de certains animaux, notamment les mammifères, ce qui leur garantit une peau imperméable. Parfois, lors d'une friction trop importante, la kératine se développe à la surface de la peau formant une callosité. Les cellules qui produisent la kératine meurent et sont remplacées continuellement. Les morceaux de kératine qui restent emprisonnés dans les cheveux sont couramment appelés des pellicules.
La molécule de kératine est hélicoïdale et fibreuse, elle s'enroule autour d'autres molécules de kératine pour former des filaments intermédiaires. Ces protéines contiennent un haut taux d'acides aminés à base de soufre, principalement la cystéine, qui forment un pont disulfure entre les molécules, conférant sa rigidité à l'ensemble. La chevelure humaine est constituée à 14 % de cystéine.
Il y a deux principales formes de kératines : l'alpha-kératine, ou α-keratin, présente chez les mammifères notamment, dont l'humain, et la bêta-kératine, ou β-keratin, que l'on retrouve chez les reptiles et les oiseaux. Ces deux types de kératines ne présentent clairement pas d'homologie de séquence.
Chez l'être humain, la kératine est fabriquée par les kératinocytes, cellules se trouvant dans la couche profonde de l'épiderme. Les kératinocytes absorbent la mélanine (pigment fabriqué par les mélanocytes), se colorent et ainsi cette pigmentation de l'épiderme permet de protéger les kératinocytes des rayons ultraviolets du Soleil.
KératinocytesLes kératinocytes sont des cellules constituant 90 % de la couche superficielle de la peau (épiderme) et des phanères (ongles, cheveux, poils, plumes, écailles). Ils synthétisent la kératine (kératinisation), une protéine fibreuse et insoluble dans l'eau, qui assure à la peau sa propriété d'imperméabilité et de protection extérieure.
L'épiderme est divisé en 4 couches basées sur la morphologie des kératinocytes (de l'intérieur vers l'extérieur) :
1. stratum germinativum (couche basale à la jonction avec le derme)
2. stratum spinosum
3. stratum granulosum
4. stratum lucidum
5. stratum corneum
Les kératinocytes passent progressivement de la couche basale vers les couches supérieures par différenciation cellulaire jusqu'au stratum corneum ou ils forment une couche de cellules mortes nommées squames, par apoptose. Cette couche constitue une barrière de protection et réduit la perte d'eau de l'organisme.
Les kératinocytes sont en perpétuel renouvellement. Ils mettent environ 1 mois pour aller de la couche basale au stratum corneum mais ce processus peut être accéléré en cas d'hyperprolifération de kératinocyte (psoriasis).

PerméabilitéIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Quand on se déplace de la peau vers la région du vermillon de la lèvre et puis vers la cavité buccale, on rencontre la muqueuse buccale. La muqueuse buccale est constituée de tissu conjonctif appelé lamina propria, recouverte par un épithélium pavimenteux stratifié. Dans les régions de la voûte du palais et de la gencive, l'épithélium est kératinisé comme l'épiderme. Dans la région buccale, le plancher de la bouche et de la face inférieure de la langue, l'épithélium n'est pas kératinisé. L'épithélium de la face dorsale de la langue est un épithélium spécialisé, mais peut être vu comme un mosaïque d'épithélium kératinisé et non-kératinisé. Les régions non-kératinisées épithéliales ne produisent pas de stratum corneum. Les noyaux avec de l'ADN intact sont retenus dans les cellules superficielles. Dans toutes les régions, les parties externes de l'épithélium fournissent une barrière de perméabilité de protection, qui varie selon les régions. VetiverLe vetiver désigne un ensemble de plantes de la famille des Poaceae (Graminées). Il s'agit de plusieurs espèces du genre Chrysopogon (et anciennement placées dans le genre Vetiveria). On en connaît une douzaine d'espèces poussant dans les zones tropicales. La plus connue est Chrysopogon zizanioides qui pousse surtout sur le sous-continent indien. Deux autres espèces sont fréquemment cultivées : Chrysopogon nigritanus (Afrique australe) et Chrysopogon nemoralis (Asie du Sud-Est).
La plante se présente sous forme de grandes touffes vertes, dont la racine, se développant verticalement, peut atteindre des profondeurs allant jusqu'à trois mètres.
Usage en parfumerie : Après distillation, la racine de vétiver fournit une essence résineuse très épaisse utilisée en parfumerie. L'essence de vétiver appartient à la famille olfactive des boisés. C'est une essence à la saveur fine et complexe : boisée, aromatique, verte, quelquefois légèrement fumée. Le vétiver est principalement utilisé dans les parfums masculins, plus rarement dans les parfums féminins. Le parfumeur Guerlain crée en 1959 un parfum pour hommes nommé Vétiver, ainsi que Carven qui commercialise alors son parfum homonyme en deux versions homme et femme. Plus récemment, Dominique Ropion crée "Vétiver extraordinaire" pour les Éditions de parfum Frédéric Malle, en exploitant un procédé d'extraction qui limite les notes camphrées et permet d'utiliser l'essence à des concentrations élevées.
En parfumerie, on utilise trois variétés différentes de racines de vétiver : le vétiver Bourbon (un des plus appréciés), le vétiver Haïti et le vétiver de Java (à l'odeur légèrement plus fumée).Index. décimale : 668.5 Parfums et cosmétiques Résumé : Skin lipids quality and quantity are essential for an efficient barrier function and providing skin hydration. It is well described today that these parameters are affected by ageing, resulting in skin sagging, wrinkles and dryness. Due to the strong commitment of our scientists to innovate responsibly and create value, they designed a high value active ingredient named here as Vetiver extract, using a groundbreaking upcycling approach. We have verified that this unique extract was able to reactivate the skin surface lipids production, bringing skin hydration for mature skin. Among the lipids capable of modifying the skin surface, there are those from sebum and skin barrier.
We firstly demonstrated that Vetiver extract was able to increase specific lipids involved in skin hydration such as ceramides and their precursors on Reconstructed Human Epidermis and skin explants. This improvement was then evidenced at clinical level in a panel of mature volunteers with poor hydration level. The extract brought hydration through an increase of lipids content and modification of lipids conformation into the skin as measured by Raman spectroscopy.
Secondly, we demonstrated that Vetiver extract allows sebum production from human sebocytes cells lines. At the clinical level on mature volunteers, we confirmed this improvement with a significant increase of sebum production.
By influencing lipid content and structure at different levels, it was hypothesized that Vetiver extract could impact the interaction between skin lipids and volatile compounds provided by fragrance application. We demonstrated, thanks to an innovative study design, that Vetiver extract lengthens the duration of fragrance by emphasizing fragrances’ heart and base notes over time. In conclusion, we developed an upcycled active ingredient able to bring skin benefits for mature skin and boosting fragrance.Note de contenu : - 1. Interestingly, their role is central in the maintenance of hydration which is related to skin barrier function and in the skin structure through adipose tissue. It is well described today that skin lipids are affected by ageing giving skin sagging, wrinkles and dryness. Thereby, developing cosmetic actives able to reactivate skin lipids would be an efficient ant-ageing strategy. Due to the strong commitment of our scientists to innovate responsibly and create value, they designed a high value active ingredient named here as Vetiver extract, using a ground-breaking upcycling approach. We evidenced that this unique extract was able to reactivate globally the skin lipids production, bringing skin hydration and plumping effect for mature skin.
- Method : In order to demonstrate the global renewal of lipids, we evaluated the lipids synthesis on cutaneous cells that produce lipids such as keratinocytes, sebocytes and adipocytes then on Reconstructed Human Epidermis and skin explants. We evaluated the expression of proteins involved in ceramides transport and barrier cornification. We then evaluated hydration and sebaceous parameters on a panel of mature volunteers.
- Results : We firstly demonstrated that Vetiver extract induced sebum production from human sebocytes cells lines but also improved its quality as observed by the production of specific antimicrobial lipids. Secondly, we demonstrated that Vetiver extract was able to restore skin barrier with the increase of skin lipids neosynthesis on Reconstructed Human Epidermis and skin explants. We also evidenced that Vetiver extract stimulated the lipids transport and epidermal cornification. Finally, Vetiver extract showed a significant effect on adipogenesis and maturation of adipocytes at in vitro and ex vivo models. We confirmed all these activities by showing that Vetiver extract improved sebum production and brought hydration through an increase of lipids content and their conformation. Vetiver extract induced an improvement of skin fatigue and a plumping effect by acting deeply on adipose tissue.
