VergeturesIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Objectifs : Les vergetures sont des lésions cutanées défigurantes qui posent souvent des problèmes d'estime de soi, mais peu d'efforts ont été consacrés dans l’étude de cette pathologie. Nous avons donc analysé des cultures cellulaires de fibroblastes dermiques isolés d'un stria alba, afin de reconstruire ensuite un modèle de peau avec une pleine épaisseur.
- Méthodes : Des fibroblastes dermiques humains (FDH) ont été isolés d'une lésion de Stria distensae (SD) et d'une peau adjacente sans lésion. Le derme de deux modèles de peau de pleine épaisseur a été reconstruit avec du HDF striae‐ ou normal, tandis que l’épiderme était reconstruit avec des kératinocytes humains normaux.
- Résultats : Principales observations et données pertinentes: L'analyse de l'expression génique de cultures cellulaires a révélé une décomposition généralisée de la matrice extra‐cellulaire (MEC) car les collagènes de types I et III, la lysyl oxydase, le biglycane, le lumican et la fibronectine étaient régulés négativement, tandis que la MMP3 augmentait ses inhibiteurs naturels diminuaient (TIMP1, TIMP2 et PAI‐1). Ces résultats ont été corroborés statistiquement pour les éléments clés de la MEC au niveau de la protéine (COL1, MMP1 et TGFB1). Il est intéressant de noter que les fibroblastes de Striae albae ont conservé un phénotype proinflammatoire, comme le suggère l'augmentation de l'expression des gènes de CXCL8, HAS1 et TNFA. Nous avons ensuite reconstruit un modèle de peau de pleine épaisseur (Stria Reconstructed) avec des fibroblastes dermiques de Striae albae. L'analyse de l'expression génique a montré que la reconstruction de Striae induisait non seulement la décomposition de la MEC, mais également le vieillissement de la peau, comme l'indique la régulation à la hausse de P16, PTGS2 et SOD2. Points de discussion : Bien que l’épiderme ait été construit avec des kératinocytes humains normaux, les stries reconstruites présentaient une atrophie épidermique et une augmentation spectaculaire du taux de β1‐intégrine au niveau de la jonction épidermo‐dermique, fournissant pour la première fois une explication rationnelle qui démontre que les cellules principales impliquées dans la pathologie de la Striae sont les fibroblastes et non les kératinocytes.
- Conclusion : Nouvelles connaissances: Ensemble, nos résultats donnent une nouvelle lumière sur l’étiologie des vergetures et indiquent que le Striae Reconstructed, un nouveau modèle de test in vitro et de dépistage du médicament, pourrait être une avancée pour le traitement des vergetures.Note de contenu : - Cell culture

Centcyamine is an alkaloid present in certain plants whose extracts have an anti-inflammatory effect. Skin fibroblasts are essential to the formation and structure of the dermis.
- Objective : Centcyamine is an indole-based alkaloid composed of coumaric acid, a resveratrol precursor and methoxytryptamine, which can be both a precursor, or a derivative, of melatonin. Given these building blocks and their well-known bioactivities, it was of interest to explore the potential benefits of using this aryl-alkaloid, in cosmetic skincare applications.
- Methods : We tested cultured normal human dermal fibroblasts (NHDF) in vitro to observe how supplementation with centcyamine improves properties of the cells to counteract the effect of ageing. The expression of genes and proteins of interest was quantified. The effect on doubling time and cell function was evaluated following treatment of the cells over several replication cycles. Skin firmness, red spot index and skin isotropy were measured with Dynaskin®, Visia® and Primos® equipment, respectively, and compared over two months in a vehicle-controlled clinical trial on 60 persons.
- Results : Centcyamine activates the expression of the gene KL and the related protein Klotho in dermal fibroblasts. Moreover, centcyamine slows the replicative ageing process of fibroblasts in culture. These cells retain cellular functions identical to those of young cells: the synthesis of lamin B1, a crucial regulatory protein of proliferation, as well as of collagen I and elastin is retained in aged cells. The clinical data are shown to improve skin isotropy in a majority of subjects, to reduce the red spot intensity and to maintain skin firmness in the treated group vs. the vehicle.
- Conclusion : The alkaloid centcyamine induces changes in the metabolism of the ageing process of human dermal fibroblasts. The up-to-now unobserved implication of both Klotho and lamin B1 to maintain homeostasis of the skin opens new venues for the prevention of age-related changes in skin structure. (Wikipedia)Index. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : As skin ages, impaired extracellular matrix (ECM) protein synthesis and increased action of degradative enzymes manifest as atrophy, wrinkling and laxity. There is mounting evidence for the functional role of exogenous peptides across many areas, including in offsetting the effects of cutaneous ageing. Here, using an artificial intelligence (AI) approach, we identified peptide RTE62G (pep_RTE62G), a naturally occurring, unmodified peptide with ECM stimulatory properties. The AI-predicted anti-ageing properties of pep_RTE62G were then validated through in vitro, ex vivo and proof of concept clinical testing.
- Methods : A deep learning approach was applied to unlock pep_RTE62G from a plant source, Pisum sativum (pea). Cell culture assays of human dermal fibroblasts (HDFs) and keratinocytes (HaCaTs) were subsequently used to evaluate the in vitro effect of pep_RTE62G. Distinct activities such as cell proliferation and ECM protein production properties were determined by ELISA assays. Cell migration was assessed using a wound healing assay, while ECM protein synthesis and gene expression were analysed, respectively, by immunofluorescence microscopy and PCR. Immunohistochemistry of human skin explants was employed to further investigate the induction of ECM proteins by pep_RTE62G ex vivo. Finally, the clinical effect of pep_RTE626 was evaluated in a proof of concept 28-day pilot study.
- Results : In vitro testing confirmed that pep_RTE62G is an effective multi-functional anti-ageing ingredient. In HaCaTs, pep_RTE62G treatment significantly increases both cellular proliferation and migration. Similarly, in HDFs, pep_RTE62G consistently induced the neosynthesis of ECM protein elastin and collagen, effects that are upheld in human skin explants. Lastly, in our proof of concept clinical study, application of pep_RTE626 over 28 days demonstrated anti-wrinkle and collagen stimulatory potential.
- Conclusion : pep_RTE62G represents a natural, unmodified peptide with AI-predicted and experimentally validated anti-ageing properties. kératineLa kératine est une protéine, synthétisée et utilisée par de nombreux êtres vivants comme élément de structure, et également l'exemple-type de protéine fibreuse.
La kératine est insoluble, et peut être retrouvée sur l'épiderme de certains animaux, notamment les mammifères, ce qui leur garantit une peau imperméable. Parfois, lors d'une friction trop importante, la kératine se développe à la surface de la peau formant une callosité. Les cellules qui produisent la kératine meurent et sont remplacées continuellement. Les morceaux de kératine qui restent emprisonnés dans les cheveux sont couramment appelés des pellicules.
La molécule de kératine est hélicoïdale et fibreuse, elle s'enroule autour d'autres molécules de kératine pour former des filaments intermédiaires. Ces protéines contiennent un haut taux d'acides aminés à base de soufre, principalement la cystéine, qui forment un pont disulfure entre les molécules, conférant sa rigidité à l'ensemble. La chevelure humaine est constituée à 14 % de cystéine.
Il y a deux principales formes de kératines : l'alpha-kératine, ou α-keratin, présente chez les mammifères notamment, dont l'humain, et la bêta-kératine, ou β-keratin, que l'on retrouve chez les reptiles et les oiseaux. Ces deux types de kératines ne présentent clairement pas d'homologie de séquence.
Chez l'être humain, la kératine est fabriquée par les kératinocytes, cellules se trouvant dans la couche profonde de l'épiderme. Les kératinocytes absorbent la mélanine (pigment fabriqué par les mélanocytes), se colorent et ainsi cette pigmentation de l'épiderme permet de protéger les kératinocytes des rayons ultraviolets du Soleil.

- Methods : A new encapsulated form of D-Biotin was developed with the purpose of improving the water solubility of biotin. This novel form of encapsulated biotin was characterized by its physicochemical properties: particle size, D-Biotin content and solubility in water. Also, proliferation and gene expression in vitro tests in cell culture were performed to evaluate its effectiveness in promoting hair growth, an ELISA test was conducted for hair keratinization and skin lightening property was tested by analysing the intracellular melanin content.
- Results : The developed WS Biotin microcapsules exhibit a particle size range of 2–30 μm with D-Biotin content of ~50% (w/w). The water solubility of WS Biotin was found to be 20-fold greater than free biotin. The obtained in vitro results indicated that WS Biotin enhances the expression of hair-related keratins in hair follicle keratinocytes, as well as the expression of hair growth-promoting genes in dermal papilla cells. Moreover, the melanin content in UVA-exposed epidermal melanocytes was reduced upon exposure to WS Biotin.

DopaLa 3,4-dihydroxyphénylalanine (abréviée en DOPA ou dopa) est une substance intermédiaire dans la synthèse des catécholamines. Elle possède deux isomères optiques, la L-DOPA ou L-dopa (ou lévodopa, dénomination pharmacologique internationale), lévogyre, et la D-DOPA ou D-dopa, dextrogyre.
La L-DOPA est la forme stéréo-isomérique lévogyre métabolisable par l'organisme. Sa décarboxylation par la DOPA-décarboxylase produit la dopamine.
La L-DOPA est capable de franchir la barrière hémato-encéphalique. C'est une substance qui est utilisée dans le traitement de la maladie de Parkinson en augmentant le taux de dopamine.

In hairy skin such as mouse skin, the inter-follicular epidermis is generally flat. In contrast, in human skin, the epidermis is wavy and possesses well-developed rete ridges. It is not well understood what difference exists in cell function between melanocytes present in the inter-rete ridge and those in the rete ridge. To clarify this problem, we first tried to determine the melanocyte and melanoblast populations per unit area of epidermis both in the rete ridge and inter-rete ridge epidermis.
- Methods : Thirty-eight samples from normal skin sites of 28 patients (14 males and 14 females) aged from 5 to 76 years were fixed and processed to dopa and combined dopa-premelanin reactions. The numbers of cells positive to the dopa (melanocytes) and combined dopa-premelanin (melanoblasts and melanocytes) reactions were scored.
Results : The average melanocyte population/0.1 mm2 of the inter-rete ridge was 74 cells, whereas that of the rete ridge was 147 cells. Moreover, the average melanoblast population/0.1 mm2 in the inter-rete ridge was 43 cells, whereas that of the rete ridge was 131 cells. The melanoblast population in the rete ridge differed between female and male, but the melanocyte populations in the rete ridge and inter-rete ridge and the melanoblast population in the inter-rete ridge did not differ between female and male. However, no age difference in the melanocyte and melanoblast populations was observed between the rete ridge and inter-rete ridge.
