Produits antisolairesTags : Nanoparticules 'Microscopie Raman' 'Pénétration peau' 'Filtre UV' 'Méthylène tétraméthylbutylphénol bis-benzotriazolyle' MBBT Stripping Index. décimale : 668.5 Parfums et cosmétiques Résumé : Le stripping en combinaison avec la microscopie Raman a été utilisé pour l’évaluation de la distribution latérale et verticale d’un filtre UV organique, le MBBT (méthylène tetramethylbutylphenol bis-benzotriazolyle) dans une formule de protection solaire. 1 mg cm−2 de produit qui contient 10% de MBBT a été appliqué sur l’avant-bras de 3 volontaires, ensuite la quantité moyenne de MBBT a été mesurée par la microscopie Raman dans 15 strips consécutifs. Le recouvrement par le MBBT était 91.1% avec 30.2% localisé sur la surface de la peau (1ère bande), 42% dans la couche cornée supérieure (bandes 2–5) et de 3.6 à 0.8% dans chacune des 10 couches consécutives suivantes. La concentration des dépôts à la surface en MBBT étaient différente d’un facteur de 300 entre les plis, les sillons et les pores sur la première main et les zones adjacentes sur l’autre main. 75% des particules appliquées ont occupé un cinquième de la zone évaluée – dans les plis et les sillons oùétait la forte concentration – cela était confirmé par la reconstruction 3D. 8.6% de MBBT était distribué sous forme de films très minces préférentiellement sur les crêtes adjacentes. Le MBBT qui était localiséà des zones qui n’étaient pas connectés à la surface, comme les pores tronqués, ou sous forme de matériel potentiellement pénétré s’élevait à 0.06% ou à 20ème du 1.4% qui était trouvé dans les strippings les plus bas de la peau. La microscopie Raman combinée au stripping a documenté quantitativement la distribution latérale et verticale et à la résolution latérale cellulaire (12.5 μm). Nos résultats ont confirmé un rapport précédent sur la distribution verticale des particules organiques qui ont été appliques sur la peau et il a été similaire dans les rapports sur la ligne de distribution de TiO2. Their size ranges typically from 50 to 1000 nm. They differ from nanoemulsions in that the lipid core is a solid. The matrix consists of solid lipids or mixtures of lipids. Over the past years it has been demonstrated that solid lipid nanoparticles appear to be a promising drug carrier system for the future. Their occlusion properties reduce transepidermal water loss and can enhance penetration of active ingredients through the stratum corneum. As with all new technologies, some problems with the solid lipid nanoparticle technology need to be solved. One major problem is the homogeneous incorporation of amphiphilic active ingredients into the crystal matrix of the nanoparticles. Actives with an amphiphilic character like tocopherol or retinol cannot be kept homogeneously distributed in the wax structure during the emulsification process. Due to their hydrophilic head group they accumulate at the exterior layer of the nanoparticles together with the surfactant system used. Consequently, homogeneous release over time is not guaranteed and a burst release has to be expected. A second disadvantage is the manufacturing process. Solid lipid nanoparticles can be produced only under high pressure conditions. Also the concentration of the solid particles in the dispersion, which is added to an emulsion, is quite low. To overcome these problems membrane structured solid nanoparticles (MSSN) have been developed. These MSSN systems consist of liquid crystalline membrane systems with extremely low surfactant concentrations. The lateral movement of actives is controlled by amphiphilic solid actives such as ceramides and solid emollients. This guarantees maintenance of the advantageous properties of solid lipid nanoparticles such as retarded release of actives and their protection against chemical decomposition, but it also allows the homogeneous incorporation of amphiphilic actives. Membrane structured solid nanoparticles are produced using a continuous three-phase emulsification technique. This allows protection of heat-sensitive actives against decomposition. The concentration of nanoparticles in the MSSN dispersion can be kept higher than 60% (w/w). Even at these concentrations the nanodispersions keep their flow properties. This paper describes our initial attempts to explain the influence of the emulsifier system on the dermal delivery of an active ingredient.
Some of the emulsifiers studied in this investigation induced liquid crystal formation in the formulations and were found to interact in two different ways. On the one hand, they prevented the evaporation of water from the formulation when applied on the skin and in doing so prolonged the delivery phase of water-soluble active ingredients as only solubilized molecules penetrate the skin at reasonable rates. On the other hand, when combined with lipophilic active ingredients, the location to which the active ingredient is delivered was changed. This can be explained by a possible interaction of these emulsifiers with the skin lipid bilayers. These are present in an orthorhombic, an hexagonal and a liquid packing, which co-exist simultaneously and are characterized by a relatively low, an intermediate and a high permeability, respectively. The results obtained in this study suggest that the use of these liquid crystal formation inducing surfactant systems favors the presence of the more permeable packing states. In a given time, an active ingredient will therefore penetrate deeper into skin with emulsifier systems that induce liquid crystal formation than with systems that do not. It is therefore suggested that such emulsifier systems be used when fast delivery of a lipophilic active ingredient or more delivery of a hydrophilic active ingredient is required, and non-liquid crystalline systems if slower or less delivery is required. The use of liquid crystalline emulsifier systems therefore allows cosmetic formulators to regulate the extent (hydrophilic active ingredients) or speed (lipophilic active ingredients) of active ingredient delivery and thus to formulate for fast efficacy.Permalink : https://e-campus.itech.fr/pmb/opac_css/index.php?lvl=notice_display&id=10511

La faisabilité d'une mesure séparée de la perméabilité du solvant et du soluté est démontrée, et la déconvolution des coefficients de diffusion et de partage de ces deux espèces est théoriquement possible. Là où les espèces diffusées changeraient la nature de la membrane, une formule de correction peut être appliquée en évaluant l'état de la membrane en fonction du temps.
