To pursue this aim, we considered the ingredient's effect on stratum corneum (SC) biomechanics to differentiate between consumers prone to tightness from those that are not and correlate these effects with mechanoreceptor activation.
- Methods : In vivo clinical trials were used to assess the tightness perception dichotomy between groups of Caucasian women; in vitro experiments were used to measure the mechanical stresses induced in the SC after cleanser and moisturizer application; and in silico simulations were used to illustrate how the measured mechanical stresses in the SC result in the development of strains at the depth of cutaneous mechanoreceptors, triggering tightness perceptual responses.
- Results : Before any cream application, women prone to tightness tend to have a more rigid SC than their less sensitive counterparts, however cleanser application increases SC stiffness in all women. Surprisingly, no correlation was found between tightness perception and hydration measurements by the Corneometer or barrier function, as evaluated by transepidermal water loss. Self-declared tightness and dryness scores were strongly associated with a self-described sensitive skin. After application of the optimized moisturizing formula, Osmoskin® containing natural waxes with good filming properties, consumers report a strong decrease in tightness and dryness perception. These results match with laboratory experiments where the cleanser was shown to increase SC drying stresses by 34%, while subsequent application of Osmoskin® decreased stresses by 48%. Finite element modelling, using experimental results as input, elucidates the differences in perception between the two groups of women. It makes clear that Osmoskin® changes the mechanical status of the SC, producing strains in underlying epidermis that activates multiple cutaneous mechano-receptors at a level correlated with the self-perceived comfort.

Peau -- analyseIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Objectif : La rupture de la barrière de la peau se produit souvent dans des conditions de la peau malade et endommagée telles que la dermatite atopique (AD). Nous nous sommes intéressés à la protéine galectine-7 (Gal-7) en tant que biomarqueur de l’état de la peau et avons évalué si le contenu de Gal-7 dans la couche cornée (scGal-7) pouvait être utilisé comme un indicateur de rupture de la barrière de la peau et comme un indice de symptômes cutanés locaux chez les patients atteints de la DA.
Methodes : L'altération des niveaux d'expression de Gal-7 dans des kératinocytes et la teneur en scGal-7 après rupture de la barrière par le dodécylsulfate de sodium ont été évalués in vitro et in vivo, respectivement. La corrélation entre la teneur en scGal-7 et la perte d'eau transépidermique (TEWL) a été examinée dans 126 sujets sains. Nous avons effectué des mesures simples des teneurs en scGal-7 dans 34 patients DA et des mesures en série sur 15 patients hospitalisés parmi eux. Les échantillons du SC ont été recueillis par la méthode d'adhésifs arrachés et la teneur en scGal-7 a été déterminée en utilisant un dosage immuno-enzymatique.
Résultats : L'expression de Gal-7 dans les kératinocytes augmente après rupture de la barrière. La teneur en scGal-7 reflète la perturbation de la barrière cutanée. Les teneurs en scGal-7 et les valeurs TEWL sont corrélées chez les sujets sains. Le niveau de scGal-7 était plus élevé chez les patients AD que chez les sujets sains. Les teneurs en scGal-7 dans la joue et le cou des patients atteints de DA sont significativement corrélés avec les scores totaux et locales de gravité des lésions de la peau. Des mesures en série chez les patients hospitalisés ont montré que les teneurs en scGal-7 dans la joue et le cou ont diminué en tandem avec des scores de gravité locales en réponse au traitement.

CéramidesUn céramide est un sphingolipide résultant de la combinaison d'un acide gras avec la sphingosine via une liaison amide. On trouve de telles molécules en abondance dans les membranes cellulaires, où elles entrent notamment dans la constitution des sphingomyélines. Les céramides ne jouent pas qu'un rôle structurel dans les membranes biologiques, et peuvent également revêtir des fonctions de signalisation lipidique. Leurs actions les mieux comprises vont de la différenciation cellulaire à la mort cellulaire programmée (apoptose) en passant par la prolifération cellulaire.
Les acides gras qui les constituent ont généralement un nombre pair (de 16 à 24) d'atomes de carbone, sont saturés ou monoinsaturés, et sont souvent α-hydroxylés.
La plupart des céramides n'existent pas à l'état naturel si ce n'est comme précurseurs de la biosynthèse des sphingolipides par une réaction d'addition sur la fonction alcool primaire de la sphingosine. (Wikipedia)

pHIndex. décimale : 668.5 Parfums et cosmétiques Résumé : The multiple protective functions of the skin derive from the interactions between epithelial skin and immune cells as well as the commensal microbiota. Developed in the last trimester of intra-uterine life, the skin barrier adapts dynamically after birth. Specific differences in the structure and physiology have been disclosed between infant and adult skin. The stratum corneum of infants is thinner and structured by thicker corneocytes with a more anisotropic surface in comparison to adult skin. Lower levels of the natural moisturizing factor and its constituents, together with the increased protease activity in the epidermis result in dry baby skin and ongoing adaptation of the desquamation to the extra-uterine environment. Infant epidermis is characterized by an accelerated proliferation rate and clinically competent permeability barrier in term neonates, despite the higher baseline values of transepidermal water loss in infants. The skin surface of newborns is less acidic, which could increase susceptibility to diaper and atopic dermatitis. Immediately after birth, skin is colonized by commensal bacteria—a process dependent on the mode of delivery and of major importance for the maturation of the immune system. Skin bacterial diversity and dysbiosis have been related to different pathology such as atopic and seborrheic dermatitis. This paper focuses on the ongoing structural, functional and biochemical adaptation of the human skin barrier after birth. We discuss the interactions on the ‘skin barrier/ microbiota/ immune system’ axis and their role in the development of competent functional integrity of the epidermal barrier. Over time, normal skin ageing is accelerated by external stresses such as smoking, pollution, chemical products and radiation. UV light, in particular UVA, causes DNA damage, apoptosis and morphological modifications, which are responsible for both premature ageing and cancer. The aim of this study was to establish a discriminatory and sufficiently reproducible cutaneous model for evaluating UVA damage, to enable testing for effectiveness of potentially protective compounds.
- Methods : The cutaneous model is based on Human skin explants irradiated with UVA. Deleterious effects on epidermis were observed and quantified by haematoxylin–eosin staining and by immunofluorescence of y-H2Ax, cytokeratin 5, involucrin and loricrin protein. Dermis deterioration was evaluated by transmission electronic microscopy and zymography in situ.
- Results : We were able to observe and quantify deleterious effects associated with UVA irradiation: epidermal and dermal disruption, appearance of Sunburn cells, increased DNA damage and induced apoptosis. The use of this model in the evaluation of protective compounds was first confirmed using sunscreens, then further validated with a panel of active ingredients which showed beneficial effects on epidermis morphology and DNA integrity after UVA exposure.

GalactomannaneLe galactomannane est un polyoside (polymère d'ose) présent dans de nombreuses graines.
Le galactomannane est une fibre végétale soluble et acalorique présente dans les graines et sert de réserve de sucre lors de la germination. Elle est abondante dans l’albumen de graines de légumineuses, telle que la cyamopsis tetragonoloba1, de la Caesalpinia spinosa et de la Ceratonia siliqua.
Le galactomannane est un polymère linéaire composé d'une chaine de monomères de mannose ((1,4)-beta-D-mannopyranose) auxquelles sont ramifiés par un pont 1-6 une unité de galactose.
Les galactomannanes sont utilisés dans l'agro-alimentaire pour modifier la viscosité et la texture des aliments (boissons, crême glacée,...). Ils sont utilisés sous forme de gomme naturelle : gomme de guar (E412), gomme tara (E417) et gomme de caroube (E410).
