StatistiqueIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : It was previously reported that a highly resistant structure, which functions as a barrier against the penetration of dyes, is present at the interface between the cuticle and the cortex of human hair. That structure was named CARB, cuticle anchored resistant base. The goal of this study was to clarify the formation and composition of the structure CARB.
- Methods : Cuticular substructures were observed from the keratinized area of each hair to its root end. The positions where the CARB structure appeared were isolated, and the barrier ability before and after that structure was evaluated. The distributions of glycolipid and cystine were measured using a nano-IR and a transmission electron microscope (TEM).
- Results : Cuticular substructures were fully constructed several mm from the hair bulb of the hairs observed. The results show that keratinization at the distal side of the cuticle cell precedes that of the proximal side, and CARB is fully constructed last among the substructures. Glycolipid was preferentially distributed at CARB in the keratinized area. The cystine content of CARB is lower than that of the A-layer; however, it is slightly higher than that of the exocuticle and the inner layer.
- Conclusion : These results demonstrate that CARB is produced in the final stage of keratinization of the cuticle layers. PolymèresIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Objectif : Le cheveu humain est un déterminant majeur de différenciation ethnique visuelle. Bien que les types de cheveux soient célébrés dans le cadre de notre diversité ethnique, l'approche des soins de cheveux a fait l'hypothèse que les types de cheveux sont structurellement et chimiquement similaire. Même si ce n'est clairement pas le cas au niveau macroscopique, l'intervention de nombreux traitements pour les cheveux est à l'échelle nanoscopique et moléculaire. L'objectif du travail présenté ici est d'identifier les principales différences hiérarchiques nanoscopiques et moléculaires à travers cinq types de cheveux ethniques différents, de fibres capillaires prises exclusivement du cuir chevelu. Ce sont des cheveux de type “Afro” (subdivisés en «caoutchouc» élastique et non élastique plus “soft”), Chinois, Européen et Mulatto (métis).
Méthodes : La diffusion de rayons X à petit angle (Small Angle X-ray Scattering=SAXS) est une technique capable de résoudre des variations nanostructurales dans les matériaux complexes. Les fibres capillaires individuels des différents types de cheveux ethniques ont été utilisées pour étudier les caractéristiques structurelles trouvées en commun et aussi spécifiques à chaque type. En même temps, la technique du Wide Angle X-Ray Scattering (WAXS) a été utilisée pour analyser la structure au niveau moléculaire sous la kératine fibreuse présente. L'ensemble des données provenant de ces deux techniques a été analysé par l'analyse en composantes principales (ACP) pour identifier les variables sous-jacentes.
Resultats : L'analyse en composantes principales des données de SAXS et de WAXS s'est montrée capable de discriminer le signal de diffusion entre les différents types de cheveux. Les résultats de la diffusion des rayons X montrent une structure commune sous-jacente du filament de kératine intermédiaire (KIF). Toutefois, des différences marquées ont été observées dans l'orientation préférentielle et dans l'intensité du signal du composant lipidique du cheveu. En outre, des différences ont été observées dans la répartition de l'intensité de la diffusion entourant le beamstop pour des échantillons à l'angle très faible.
Conclusion : Les résultats indiquent que la structure des fibres de kératine reste cohérente entre les types de cheveux ethniques. Les hiérarchies établies par ceux-ci peuvent être modulées par la variation de la teneur en Kératine Associée Protéines (KAP) et en lipides qui modifient les structures d'interface et conduisent à des différences macroscopiques de la morphologie des cheveux.Note de contenu : - MATERIALS AND METHODS : Experimental X-ray scattering - Hair sample preparation and experimental setup - Data correction and reduction

Spectroscopie RamanIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : Hair greying (i.e. canitie) is a physiological process occurring with the loss of melanin production and deposition within the hair shafts. Many studies reported the oxidation as the main biological process underlying this defect of pigmentation. Even though the overall appearance and biomechanical properties of hairs are reported to be altered with greying, there is a lack of information about molecular modifications occurring in grey hair shafts. The aim of this study was thus to investigate the molecular signature and associated changes occurring in greying hair shafts by confocal Raman microspectroscopy.
- Methods : This study was conducted on pigmented, intermediate (i.e. grey) and unpigmented hairs taken from 29 volunteers. Confocal Raman microspectroscopy measurements were acquired directly on hair shafts.
- Results : Automatic classification of Raman spectra revealed 5 groups displaying significant differences. Hence, the analysis of the molecular signature highlighted the existence of 3 sub-groups within grey hair : light, medium and dark intermediate. Among molecular markers altered in the course of greying, this study identified for the first time a gradual modification of lipid conformation (trans/gauche ratio) and protein secondary structure (α-helix/β-sheet ratio), referring respectively to an alteration of barrier function and biomechanical properties of greying hair.

Régénération (biologie)Index. décimale : 668.5 Parfums et cosmétiques Résumé : OBJECTIF : Les interactions mésenchymateuses- épithéliales sont importantes dans le contrôle de la croissance des cheveux et du cycle pilaire. La voie β-caténine des cellules des papilles dermiques (CPD) joue un rôle essentiel dans la morphogenèse et la régénération normale des follicules pileux. La suppression de la β-caténine dans la papille dermique réduit la prolifération des cellules souches du follicule pileux qui génèrent la tige du cheveu et induit une apparition précoce de la phase catagène. Dans cette étude, un modulateur de l'activité/β-caténine Wnt a été étudiée dans METHODES : L'effet des graines de mauve crépue (Malva verticillata) sur les CPD humaines a été étudié par un système de dosage de l'activité du gène rapporteur Wnt/β-caténine (TCF-β-caténine/gène rapporteur LEF) et l'analyse de la prolifération cellulaire. La synthèse des facteurs liés à la croissance des cheveux et au cycle a été mesurée à la fois au niveau d'ARNm et des protéines par PCR semi-quantitative et une analyse par Western blot, respectivement.
RESULTATS : Un extrait des graines de M. verticillata a augmenté l'activité de Wnt rapporteur d'une manière dépendante de la concentration et a également conduit à des niveaux accrus de β-caténine dans les CPD humaines en culture. L'acide myristoléique, identifié comme étant un composé efficace des graines de M. verticillata, a stimulé la prolifération des CPD d'une manière dépendante de la dose ainsi que l'augmentation des niveaux des cibles en aval de l'IGF-1, KGF, VEGF, HGF et leur transcription. L'acide myristoleïque a CONCLUSION : Globalement, les données suggèrent que cet extrait des semences de M. verticillata pourrait être un bon candidat pour le traitement de la perte de cheveux en modulant la voie/β-caténine dans les CPD.Note de contenu : - MATERIALS AND METHODS : Cell culture - Extraction, isolation and structure determination of active compounds - Wnt/β-catenin reporter activity assay - Western blot analysis - Proliferation of DPCs - RNA extraction and reverse transcription polymerase chain reaction (RT-PCR) - Statistical analysis
