Vitamine CIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Wrinkles are the most obvious expression of skin aging and are manifested by numerous changes in the organization and structure of the dermis. To better understand if this tissue modification could be linked to a modification of cell function, contractile and synthesis capacities of normal aged human fibroblasts and those obtained from a biopsy of a forehead wrinkle were studied and compared. The capacity of fibroblasts to adhere to the collagen network and to maintain a three-dimensional structure of the dermis was studied using a three-dimensional model of a collagen gel. The metabolic activity of both cell types was determined immunochemically by quantifying collagen I synthesis. Human fibroblasts from the wrinkle contracted the collagen gel less than normal aged human fibroblasts and synthesized less collagen I. The results show that the metabolic activity of aging fibroblasts decelerates and that aging fibroblasts lose their capacity to adhere to collagen fibers, thus limiting the possibility of organizing dermal tissue.
The potential of an active ingredient to compensate for the reduction of metabolic activity and to restore the contractile capacity of fibroblasts from the wrinkle was investigated. Spectroscopie RamanIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : Hair greying (i.e. canitie) is a physiological process occurring with the loss of melanin production and deposition within the hair shafts. Many studies reported the oxidation as the main biological process underlying this defect of pigmentation. Even though the overall appearance and biomechanical properties of hairs are reported to be altered with greying, there is a lack of information about molecular modifications occurring in grey hair shafts. The aim of this study was thus to investigate the molecular signature and associated changes occurring in greying hair shafts by confocal Raman microspectroscopy.
- Methods : This study was conducted on pigmented, intermediate (i.e. grey) and unpigmented hairs taken from 29 volunteers. Confocal Raman microspectroscopy measurements were acquired directly on hair shafts.
- Results : Automatic classification of Raman spectra revealed 5 groups displaying significant differences. Hence, the analysis of the molecular signature highlighted the existence of 3 sub-groups within grey hair : light, medium and dark intermediate. Among molecular markers altered in the course of greying, this study identified for the first time a gradual modification of lipid conformation (trans/gauche ratio) and protein secondary structure (α-helix/β-sheet ratio), referring respectively to an alteration of barrier function and biomechanical properties of greying hair.

CéramidesUn céramide est un sphingolipide résultant de la combinaison d'un acide gras avec la sphingosine via une liaison amide. On trouve de telles molécules en abondance dans les membranes cellulaires, où elles entrent notamment dans la constitution des sphingomyélines. Les céramides ne jouent pas qu'un rôle structurel dans les membranes biologiques, et peuvent également revêtir des fonctions de signalisation lipidique. Leurs actions les mieux comprises vont de la différenciation cellulaire à la mort cellulaire programmée (apoptose) en passant par la prolifération cellulaire.
Les acides gras qui les constituent ont généralement un nombre pair (de 16 à 24) d'atomes de carbone, sont saturés ou monoinsaturés, et sont souvent α-hydroxylés.
La plupart des céramides n'existent pas à l'état naturel si ce n'est comme précurseurs de la biosynthèse des sphingolipides par une réaction d'addition sur la fonction alcool primaire de la sphingosine. (Wikipedia)

Elle peut avoir différentes causes, comme certaines pathologies (dermatite atopique aussi appelé eczéma atopique), des carences, notamment en Vitamine A, ou des facteurs externes, comme les contacts avec des substances irritantes ou le traitement par certains médicaments.Index. décimale : 668.5 Parfums et cosmétiques Résumé : - OBJECTIF : Bien que la xérose soit un désordre cutané couramment retrouvé au sein de la population, aucune étude in vivo globale n'est disponible. Ainsi, l'objectif de ce travail était de caractériser la peau xérotique de l’échelle macroscopique à l’échelle moléculaire, in vivo, par une approche non-invasive.
- METHODES : 15 volontaires sains avec une peau normale et 19 volontaires sains avec une peau xérotique ont été sélectionnés par un dermatologue grâce à une échelle de score visuelle. Dans un premier temps, la surface de la peau a été caractérisée par mesures biométriques. L’état de sécheresse de la peau a ensuite été évalué par microscopie confocale in vivo. La signature moléculaire de la peau xérotique a été déterminée par microspectroscopie Raman in vivo. Enfin, une identification des lipides du stratum corneum a été réalisée en utilisant la chromatographie liquide en phase normale couplée à deux détecteurs, Corona et spectrométrie de masse / haute résolution.
- RESULTATS : Les mesures biométriques ont démontré que la peau xérotique présente une augmentation de la perte insensible en eau et une diminution du taux d'hydratation. La microscopie confocale révèle une altération de la morphologie cellulaire dans la peau xérotique. De plus, la microspectroscopie confocale Raman démontre, in vivo et de manière non-invasive, un manque d'organisation et de conformation des lipides avec la xérose. Enfin, les analyses HPLC révèlent que les trois sous-classes de céramides (NdS, NS et EOP) sont significativement diminuées dans la peau xérotique. L'ensemble de ces résultats a identifié les paramètres caractéristiques de la peau xérotique comparativement à la peau normale.
- CONCLUSION : Cette étude a mis en évidence des paramètres discriminants de l’échelle macroscopique à l’échelle moléculaire, in vivo et de manière non-invasive, entre les peaux normales et xérotiques.
