Toxicologie génétiqueIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Objectifs : La surveillance de la toxicité subaiguë et chronique est essentielle lors du développement de nouvelles molécules cosmétiques. Compte tenu du manque de méthodes alternatives validées, nous avons développé un modèle de cytotoxicité à doses réitérées in vitro sur cellules TPH-1.
Methodes : Cultivées en suspension, les cellules ont été traitées avec des produits chimiques pendant 14 jours, à raison de 3 applications par semaine, et le taux de viabilité cellulaire a été déterminé par le test MTT. Nous avons commencé par étudier les effets à long terme de produits chimiques induisant différents degrés de cytotoxicité : le Paraquat, le 3-acide nitropropanoique (3-NPA) et le dodecyl sulfate de sodium (SDS). A partir d'une étude de cytotoxicité aigüe, des doses comprises entre 1 et 10 μg ml−1 ont donc été choisis pour réaliser notre étude de cytotoxicité subaigüe. Une étude comparative du potentiel genotoxique du Paraquat et du peroxyde d'hydrogène (H2O2) a été menée sur les cellules TPH-1. Plusieurs tests des comètes ont été réalisés : à 1 h (4°C), après une période de récupération de 24 h (37°C) et après un traitement de 14 jours (37°C). Une fois adapté pour tester des extraits végétaux ou de molécules fortement diluées, certains de nos produits cosmétiques ont été testé avec ce model.
Resultats : Comme pressentie, après 14 jours de traitement au Paraquat, le taux de viabilité cellulaire a fortement diminué pour des doses supérieures à 3 μg ml−1, alors que 10 μg ml−1 de 3-NPA ou de SDS n'induisaient pas plus de 44 % de mortalité cellulaire. Etonnamment, les résultats des tests des comètes ont montrés que les "tails moments" dus à l'H2O2 restent stables, alors que ceux liés au Paraquat augmentent de manière "dose-dépendante". D'autre part tous les composés testés entre 0.5 et 5 μg ml−1 ont été classés comme inoffensifs et ceci même avec un seuil placé à 90% de viabilité cellulaire.
Conclusions : En conclusion, ce test pourrait présenter un intérêt pour l'étude de cytotoxicité subaigüe et génotoxique des produits appliqués quotidiennement et suggère que le Paraquat en serait un contrôle positif de choix.Note de contenu : - MATERIALS AND METHODS : Cell culture maintenance - Cell viability assessment after acute cytotoxicity assay - Cell viability assessment after subacute cytotoxicity assay - Comet assay after 1h of exposure at 4°C - Comet assay after 24h or 14 days of exposure at 37°C - Statistical analysis

Produits chimiques -- Suppression ou remplacementIndex. décimale : 667.6 Peintures Résumé : Looking into the history of anticorrosive pigments (ACO-pigments), the driving innovation factors were always performance and legislation in combination with costs. The toxic behaviour of red lead or chromates already opened the door to non-toxic alternatives 30 years ago. The stepwise ongoing stricter handling of for example the Cr6 ended up in an almost complete ban in Europe for the benefit of all consumers : Chrome-based pigments are SVHCs (substances of very high concern) in the European Union and currently the sunset date for eg Strontiumchromate and Zinctetraoxychromate is set for January 2019 with the possibility to apply for an authorisation until July 2017.
These pigments show an excellent cost/performance ratio. Nevertheless, due to their toxicity, for more than 30 years, alternatives like zinc phosphates have made inroads. Standard products with universel applicability significantly reduced the consumption of chromates, however they did net always reach the required performance due to a different working mechanism. All phosphate pigments have in common the fact that they build an anodic passivation via precipitation of phosphate complex layers in combination with metal hydroxide barriers (see Figure 1), white Crelons additionally act cathodically.
Therefore, standard zinc phosphate has been improved by two major steps : The first step was the modification of standard zinc phosphate changing, for example, pH, solubility, adding synergistic values etc. Second was the introduction of polyphosphates for higher sophisticated applications, such as coil coatings, aircraft primers etc, which can also be supported by the use of silica-based ion-exchange pigments.
All improved products are highly linked to specific binder systems, therefore more universel zinc-containing pigments have been developed as 'wide-spectrum-anticorrosives', combining the different improvement factors into a single pigment or even developing different modifications.Note de contenu : - How to use zinc-containing aco-pigments to achieve a labelling-free paint ?

N-méthyl-2-pyrrolidoneLe N-méthyl-2-pyrrolidone ou 1-méthyl-2-pyrrolidone, généralement abrégé en NMP, est un liquide incolore ou jaune clair. C'est un amide cyclique (lactame) souvent utilisé en tant que solvant organique.C'est un solvant hygroscopique, miscible avec l'eau et la plupart des solvants organiques. C'est une base faible: une solution aqueuse contenant 10% de NMP a un pH de 7,7-86.
Utilisation : Ce composé est principalement utilisé comme solvant à cause de ses propriétés physiques: faible volatilité, stabilité thermique, solvant polaire et aprotique. Ses propriétés toxicologiques en font un bon candidat pour le remplacement de solvants chlorés.
Les domaine d'utilisation sont vastes : en pétrochimie, il sert de solvant d'extraction pour les composés aromatiques et le butadiène ou de purification des gaz (récupération du dioxyde de carbone et du sulfure d'hydrogène); en électronique, il sert de solvant de nettoyage pour les wafers de silicium; en chimie, il est utilisé comme solvant de réaction ou de formulation ; comme agent de nettoyage dans l'industrie des métaux (dégraissage).
