Tanins végétauxIndex. décimale : 668.5 Parfums et cosmétiques Résumé : The cosmetics industry is in the midst of a significant shift, driven by consumer demand for both natural and sustainable ingredients. This transformation is accelerated by concerns on conventional components like UV filters, known for potential environmental degradation and endocrine disruption. Despite the urgency, natural UV-absorbing alternatives based on chemical light absorption remain scarce. Secondary plant substances (SPS), particularly water-soluble tannins, present a promising possibility. This study investigated the reducing capacities, antioxidant activity, tannin contents, and UV-absorption properties of various plant-based extracts mainly obtained from by-products and residues from the agricultural and food industry. By utilizing side-streams, emerging ingredients are potential factors to contribute to the sustainability of cosmetics. The results revealed that bark and tea grounds exhibited the highest reducing capacities and antioxidant activities, making them potential natural sources. Fruit peels, however, showed relatively low antioxidant capacities. Tannin contents varied significantly among the samples, with wine pomace and bark having the highest levels. The UV-absorption measurements indicated that the extracts had the ability to block UV-A and even some visible light, except for fruit peels and nut shell extracts, which were less suitable for UV protection. More than a trend in today's cosmetic industry, natural actives have become a necessity. Therefore, sources of natural actives and natural molecules are a key field of research. In the 1990s, about 80% of the known natural products were from plant origin. 'More specifically, each year, a large proportion of newly described molecular structures are from higher plants.
BiotechMarine, a subsidiary of Seppic, is specialised in hi-tech bioactive natural substances, extracted from seaweeds and marine plants. It proposes to cosmetics laboratories a wide range of 80 active ingredients, extracts and stem cells for personal care. Established in France on the northern coast of Brittany in 1988, its expertise in the world of top marine active ingredients and cutaneous engineering is recognised around the world. For 25 years, BiotechMarine's values remain innovation, safety, efficiency, sustainable development and respect of the biodiversity.
BiotechMarine started developing actives from marine plants stem cells 10 years ago. It was one of the first companies to offer such products in the cosmetic industry, first with cellulosome Crithmum maritimum, followed by cellulosome Eryngium maritimum. KératinocytesLes kératinocytes sont des cellules constituant 90 % de la couche superficielle de la peau (épiderme) et des phanères (ongles, cheveux, poils, plumes, écailles). Ils synthétisent la kératine (kératinisation), une protéine fibreuse et insoluble dans l'eau, qui assure à la peau sa propriété d'imperméabilité et de protection extérieure.
L'épiderme est divisé en 4 couches basées sur la morphologie des kératinocytes (de l'intérieur vers l'extérieur) :
1. stratum germinativum (couche basale à la jonction avec le derme)
2. stratum spinosum
3. stratum granulosum
4. stratum lucidum
5. stratum corneum
Les kératinocytes passent progressivement de la couche basale vers les couches supérieures par différenciation cellulaire jusqu'au stratum corneum ou ils forment une couche de cellules mortes nommées squames, par apoptose. Cette couche constitue une barrière de protection et réduit la perte d'eau de l'organisme.
Les kératinocytes sont en perpétuel renouvellement. Ils mettent environ 1 mois pour aller de la couche basale au stratum corneum mais ce processus peut être accéléré en cas d'hyperprolifération de kératinocyte (psoriasis).

Stress oxydatifIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - OBJECTIF : Examiner la capacité d'un extrait issu de la médecine traditionnelle chinoise, le Polygonum multiflorum Radix, à protéger la viabilité des mélanocytes soumis à un stress oxydant, un mécanisme clé dans l'initiation et la progression du blanchissement du cheveu.
- METHODES : Dans le but de tester le potentiel antioxydant de l'extrait Polygonum multiflorum Radix, nous avons traité des mélanocytes humains normaux issus de cultures primaires de prépuce avec une version commerciale de l'extrait de Polygonum multiflorum Radix, puis exposé les cellules au peroxyde d'hydrogène (H2O2). La concentration en espèces réactives de l'oxygène et le ratio glutathion/protéines ont été déterminés pour objectiver l'effet antioxydant de l'extrait. Afin d'améliorer sa solubilité pour une utilisation cosmétique, un nouvel extrait du Polygonum multiflorum Radix a été développé. Le blanchissement des cheveux étant la conséquence d'une disparition des mélanocytes au sein des follicules pileux, nous avons vérifié si le potentiel anti-oxydant du nouvel extrait de Polygonum multiflorum Radix préserve la viabilité des mélanocytes soumis à un stress oxydant de type H2O2, ainsi que la pigmentation au sein de follicules pileux humain.
- RESULTATS : Le traitement de mélanocytes humains normaux issus de cultures primaires de prépuce par la version commerciale de l'extrait de Polygonum multiflorum Radix, a entrainé une diminution de l'accumulation des espèces réactives de l'oxygène intracellulaires en réponse à une exposition à l'H2O2, ainsi qu'une préservation du ratio glutathion/protéines. Ce résultat confirme un potentiel protecteur de l'extrait vis-à -vis des mélanocytes soumis à un stress induit par H2O2 et suggère une capacité à prévenir la mort des mélanocytes face à un stress oxydant. Une diminution de la mort cellulaire a été confirmée lorsque les mélanocytes sont traités par l'extrait amélioré de Polygonum multiflorum Radix, démontrant ainsi un effet cytoprotecteur. Enfin, le contenu en mélanine de follicules pileux humains était significativement supérieur après traitement par la version améliorée de l'extrait de Polygonum multiflorum Radix, en comparaison au contrôle traité par le véhicule, confirmant ex vivo l'effet protecteur de l'extrait sur la pigmentation du cheveu.
- CONCLUSION : L'extrait de Polygonum multiflorum Radix protège in vitro les mélanocytes humains normaux des effets délétères liés à l'exposition au stress induit par H2O2 et a un impact bénéfique sur la pigmentation au sein des follicules pileux humains ex vivo, montrant l'intérêt de l'extrait de Polygonum multiflorum Radix en tant qu'ingrédient actif pour la protection des mélanocytes contre une mort prématurée. Ces données in vitro fournissent des preuves mécanistiques en accord avec les études in vivo existantes pour l'utilisation de l'extrait de Polygonum multiflorum Radix comme stratégie dans la prévention du blanchissement du cheveu induit par le stress oxydant, et en lien avec l'utilisation traditionnelle du Polygonum multiflorum.Note de contenu : - METHODS : Reagents - Polygonum multiform radix root extracts - Melanocyte ROS inhibition - Measurement of reduced glutathione rate in normal human primary foreskin melanocytes - Melanocyte viability assay - Human hair follicle isolation - Evaluation of melanin content in hair follicles
