This crosstalk is particularly important for the regulation of complex events such as wound healing, but also for continuous tissue repair and regeneration. The messenger molecules involved are mainly cytokines and growth factors. An imbalance in the synthesis and release of these signals as it is observed in elderly skin may lead to a reduced biosynthesis of important extracellular-matrix (ECM) proteins such as collagen and elastin. Here we present a strategy to activate matrix protein production by stimulating growth factor synthesis and release by keratinocytes, which can easily be reached by superficially applied cosmetic compounds. In vitro assays demonstrate that a crocus bulb extract can activate expression of elastin, lysyl oxidase-like 2 enzyme (LOXL2) and the connective tissue growth factor (CTGF). Non-invasive skin structure analysis by two-photon microscopy performed after a two-week treatment with a cream containing the crocus bulb extract demonstrates a clear augmentation of collagen and elastine. Dark circles and puffy eyes still benefit from few efficient treatments. We have shown that Meiyanol, an ecoresponsible of elderberry (Sarnbucus nigra) flowers is capable extract of efficiently reducing bath dark circles and puffy eyes via an original mechanism of action. Indeed, Meiyanol is able to inhibit the expression of the anchoring proteins I-CAM1 and I-CAM1 in the endothelial cells of the capillaries lumen, hence preventing the leukocyte binding and their extravasation. Since this extravasation is responsible for generating leaks of red blood cells and plasma that will in turn form dark circles and puffy eyes respectively, reducing it leads to a significant reduction in these two symptoms. This work opens new alternatives and describes new anti-ageing targets for the development of future cosmetic eye contour actives.
