(Wikipedia)Index. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : As skin ages, impaired extracellular matrix (ECM) protein synthesis and increased action of degradative enzymes manifest as atrophy, wrinkling and laxity. There is mounting evidence for the functional role of exogenous peptides across many areas, including in offsetting the effects of cutaneous ageing. Here, using an artificial intelligence (AI) approach, we identified peptide RTE62G (pep_RTE62G), a naturally occurring, unmodified peptide with ECM stimulatory properties. The AI-predicted anti-ageing properties of pep_RTE62G were then validated through in vitro, ex vivo and proof of concept clinical testing.
- Methods : A deep learning approach was applied to unlock pep_RTE62G from a plant source, Pisum sativum (pea). Cell culture assays of human dermal fibroblasts (HDFs) and keratinocytes (HaCaTs) were subsequently used to evaluate the in vitro effect of pep_RTE62G. Distinct activities such as cell proliferation and ECM protein production properties were determined by ELISA assays. Cell migration was assessed using a wound healing assay, while ECM protein synthesis and gene expression were analysed, respectively, by immunofluorescence microscopy and PCR. Immunohistochemistry of human skin explants was employed to further investigate the induction of ECM proteins by pep_RTE62G ex vivo. Finally, the clinical effect of pep_RTE626 was evaluated in a proof of concept 28-day pilot study.
- Results : In vitro testing confirmed that pep_RTE62G is an effective multi-functional anti-ageing ingredient. In HaCaTs, pep_RTE62G treatment significantly increases both cellular proliferation and migration. Similarly, in HDFs, pep_RTE62G consistently induced the neosynthesis of ECM protein elastin and collagen, effects that are upheld in human skin explants. Lastly, in our proof of concept clinical study, application of pep_RTE626 over 28 days demonstrated anti-wrinkle and collagen stimulatory potential.
- Conclusion : pep_RTE62G represents a natural, unmodified peptide with AI-predicted and experimentally validated anti-ageing properties. Hyaluronique, acideL'acide hyaluronique est un type de polysaccharide (plus précisément une glycosaminoglycane) non fixé à une protéine centrale et largement réparti parmi les tissus conjonctifs, épithéliaux et nerveux animaux.
Il se trouve notamment dans l'humeur vitrée et le liquide synovial. Il est l'un des principaux composants de la matrice extracellulaire ainsi que de certaines mucoprotéines lorsqu'il est associé à une fraction protéique.
Depuis les années 1990, il est très utilisé dans divers dispositifs médicaux, médicamenteux et cosmétiques (présenté dans ce dernier cas comme "antistatique, humectant, hydratant, conditionneur cutané, anti-âge, etc."), bénéficiant d'un effet de mode grandement soutenu par une large publicité commerciale.
Contribuant de façon significative à la prolifération et à la migration des cellules, l'acide hyaluronique est aussi impliqué dans la progression de certains cancers. (Wikipedia)

These shifting sentiments put active ingredients in the spotlight and demand innovation. Biodesign is rising to the challenge. This approach to product development combines advances in computational biology and precision fermentation to deliver the best of nature's depth and diversity to consumers. White many are familiar with biotechnology's first breakthrough ingredient — hyaluronic acid (HA) — proteins like collagen have rapidly emerged as the pioneering products enabled by biodesign.
HA from microbial fermentation paved the way for future fermentation-derived bioactive ingredients. In 1998 scientists at Savient Pharmaceuticals successfully produced high molecular weight HA, historically sourced from rooster combs, through precision fermentation of the microorganism Streptococcus zooepidemicus' Biotechnology unlocked the potential to create sustainable alternatives to animal-derived molecules that eliminate unethical practices, complicated supply chains, and the use of environmentally harsh chemical processes.
Collagen is another popular ingredient in beauty and an important target for biodesign as it is the most abundant protein in the human body, responsible for structure, stability, and strength within the dermal layers. In addition to its essential structural function, collagen is a signaling molecule defining cellular activities.

Taches pigmentaires (dermatologie)Index. décimale : 668.5 Parfums et cosmétiques Résumé : Dermal-epidermal junction (DEJ) is a key compartment of the skin, and its role is to attach the epidermis to the dermis in order to ensure good cohesion and resistance to the skin. With age, this DEJ may be altered, hence leading to premature ageing (loss of skin elasticity, impaired wound healing...).
Here we show that Sirhamnose, a rhamnose derivative, is capable of improving the expression of DEJ constituents (laminin, collagen IV, and collagen VII) hence leading to a thicker and healthier DEJ. In order to measure the impact of a healthy DEJ on the skin, a clinical test was performed and the 35 healthy volunteers received a daily treatment with Sirhamnose. Global skin improvements were observed, especially for hydration, firmness and skin tone uniformity. This data confirms that DEJ is a central actor in skin health, and that a specific treatment negates the effects of premature aging. The DEJ-specific product Sirhamnose is therefore a novel and original candidate for any anti-ageing treatment.Note de contenu : - Design of a bioavailable rhamnose derivative

The CC-516 has a high proportion of proline (13%) of free amino acid content. The inhibitory effect of CC-516 on the elastase exhibited 37 to 90% inhibition by 10 to 250 μg/ml concentration ; the IC values with 40.8 μg/ml for porcine pancreatic elastase (PPE) and 48.1 μg/ml for human leukocyte elastase (HLE), respectively. One of the effects of elastase is that it is known to reduce the number of elastin fibers at the level of the enzyme deposition. The number of elastin fibers was increased when we drift from the deposit number of elastase with 100 mg/ml of CC-516. CC-516 showed protection of elastic fiber against degradation by the enzyme in an ex vivo assay. The CC-516 increased proliferation of human fibroblast cell by 85% at 10 concentration, compared with control, whereas the increase by ascorbic acid was 50%. The collagen synthesis was increased by 40% at 10% concentration of CC-516. The treatment with CC-516 improved skin hydration, the skin elasticity, and skin wrinkles. From this study, we suggest that CC-516 can be used as a new anti-aging component for cosmetics.
