ÉpichlorohydrineL'épichlorohydrine ou épichlorhydrine ou 1-chloro-2,3-époxypropane, est un composé organochloré de formule semi-développée CH2CHOCH2Cl, liquide volatil toxique, incolore à forte odeur d'ail. Un atome de carbone de l'épichlorohydrine est chiral, il existe donc deux formes énantiomères de ce composé.
GélatineLa gélatine est une substance solide translucide, transparente ou légèrement jaune, presque sans goût et sans odeur, obtenue par l'ébullition prolongée de tissus conjonctifs (peaux) ou d'os d'animaux (principalement porc, bœuf, poisson). Elle possède de nombreuses applications dans le domaine culinaire, la médecine, les industries agroalimentaire et pharmaceutique.
En matière d’étiquetage, la gélatine est considérée par la norme européenne3 comme un ingrédient et non pas comme un additif, c'est pourquoi elle n'a pas de numéro E. Hors Union européenne, elle est considérée par certains pays comme un additif gélifiant et on peut la trouver avec la dénomination E441.
La gélatine est un mélange de protéines obtenu par hydrolyse partielle du collagène extrait de la peau comme la peau de porc (cochon), des os, des cartilages, etc. Les liaisons moléculaires entre les fibres de collagène sont alors brisées. Mélangée à de l'eau, la gélatine forme un gel colloïdal semi-solide thermo-réversible (il fond lorsqu'il est chauffé et recouvre son aspect gélatineux lorsqu'il est refroidi). Sous forme déshydratée, par contre, la gélatine n'a pas de point de fusion et devient friable ou brûle quand elle est chauffée à trop haute températureLa rhéologie de la gélatine se caractérise par un comportement viscoélastique, et des contraintes trop élevées ou appliquées trop rapidement peuvent entraîner une rupture fragile (fracturation) ou ductile6. Le caractère plutôt élastique/fragile ou plutôt visqueux/ductile dépend de la concentration en gélatine de la solution aqueuse et de la température, ainsi que de la durée de la mise sous contrainteLes acides aminés constituant la gélatine sont : la glycine (21 %), la proline (12 %), l'hydroxyproline (12 %), l'acide glutamique (10 %), l'alanine (9 %), l'arginine (8 %), l'acide aspartique (6 %), la lysine (4 %), la sérine (4 %), la leucine (3 %), la valine, la phénylalanine et la thréonine (2 %), l'isoleucine et l'hydroxylysine (1 %), la méthionine et l'histidine (< 1 %) et la tyrosine (< 0,5 %). Ces valeurs sont variables (surtout pour les constituants minoritaires) et dépendent de la source de matériaux bruts et de la technique de préparation. La gélatine est constituée à environ 98-99 % (en poids sec) de protéines et contient 18 acides aminés dont huit des neuf acides aminés essentiels à l'Homme. Elle n'a qu'une relative valeur nutritionnelle du fait de l'absence de tryptophane et de son déficit en isoleucine, thréonine et méthionine; elle possède également un taux inhabituellement élevé d'acides aminés non essentiels, la glycine et la proline (qui sont produits par le corps humain). Transition vitreuseIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : To improve the cross-linking efficiency of gelatin molecules, sodium acetate trihydrate/urea deep eutectic solvent with 30% water (SAT/U-DES30W, based on DES weight) was proposed as the solvent of gelatin in this paper. The system could overcome the gelation effect of gelatin at low temperature and expose more active groups of gelatin molecules, resulting in better cross-linking effect. Firstly, the optimal reaction concentration of epichlorohydrin (ECH) was determined by boiling water solubility of cross-linking products, and the cross-linking efficiency of gelatin under two systems was judged together with the Glass transition temperature method. Secondly, the water resistance of the cross-linked product was investigated by swelling capacity and water contact angle. The water absorption rate of GEW and GEDES decreased from 4614% to 1500% and 929% at 12 h, respectively, and the water contact angle also obviously decreased, indicating that the cross-linking efficiency of gelatin under DES system was higher. Finally, the cross-linking mechanism of gelatin and ECH were discussed and it was found the thermal stability of GEDES was significantly improved, which would provide a theoretical basis for gelatin-based materials. Chlorure de cyanureLe chlorure cyanurique est un composé chimique de formule (NCCl). C'est un dérivé chloré de la 1,3,5-triazine. C'est un trimère du chlorure de cyanogène.
- Propriétés : Le chlorure cyanurique se présente sous la forme d'une poudre cristalline blanche à l'odeur de dichlore ou d'acide chlorhydrique. À l'air sec, même par température élevée il est stable, mais dès la présence d'humidité, il s'hydrolyse en acide cyanurique et acide chlorhydrique. La vitesse de cette hydrolyse dépend fortement de la température et du pH. Dans une solution tamponnée à pH = 7 d'eau glacée, il est stable pendant plusieurs heures, mais dès que la température dépasse 10 °C, l'hydrolyse se produit, d'autant plus rapidement que la température augmente, aidée par le fait qu'elle est autocatalysée par l'acide chlorhydrique. Phosphorique, AcideL’acide phosphorique est un composé chimique de formule H3PO4. C'est un oxoacide trifonctionnel (triacide) important en chimie inorganique et fondamental en biochimie. Il s'agit d'un acide minéral obtenu par traitement de roches phosphatées ou par combustion du phosphore.

Cross-linking of collagen phosphoric acid hydrolysates was demonstrated by the decrease of the number of the free primary amino groups, the increase in the denaturation temperature, collagenase resistance and changes in the FTIR spectra. The effects of concentration of the cross-linking agent, pH of the solution and the reaction time were studied. The cross-linked collagen hydrolysates showed higher cross-linking densities compared to non-cross-linked collagen hydrolysates. The denaturation temperature was elevated from 24°C for non-cross-linked DSC samples to 28°C for cross-linked samples. Collagen hydrolysates showed the increase resistance to in vitro collagenase degradation. The mixture of collagen hydrolysates with cyanuric chloride gives the possibility of producing new materials for potential biomedical applications. Réticulation (polymérisation)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : Genipin(GP) has been used as a sort of perfect naturel cross-linking agent for collagenous biomaterials, but the cross-linking mechanism between collagen and GP is not clear. The aldehyde groups can be formed in aqueous solution of Genipin (GP), which can react with Cu(OH)2 and be used in the silver mirror reaction. About 0.2506 moi aldehyde groups were determined by Decolor Fuchsin Spectrophotometry in 1mol GP. In neutral solution, the ultraviolet absorption peak of GP appeared at 242.10nm, whereas in alkaline solution, it GP moved to 271.70nm due to the generation of enolates structure. Compared with the neutral condition, the GP molecular size almost doubled in alkaline solution, therefore il was speculated that dimers were formed, In this work, porcine acellular dermal matrix (pADM) was employed as a model of collagen to react with Genipin. The colour of GP cross-linked pADM (GP-pADM) turned into yellow from white initialty and finally into dark blue and a new absorption peak was round at 2356.79cm-1. With an increasing amount of GP, the shrinkage temperature (Ts) of GP-pADM increased slowly from 55.4°C to 80°C (6% dosage). Regarding the formation of dimers, the size of GP molecule was increased which fitted the distance of two aminos of the collagen for cross-linking. Furthermore, due to the ring structure of genipin and cross-linking bonds, the amount of rigid structures of collagen increased, consequently improved collagen stability was achieved. Fibroblasts grew well on GP-pADM with great morphology, and the vitro cytotoxicity was grade 0.
