Given the period during which the case-control studies were conducted, the sunscreens prevalent at that time, and how suncreen is used and applied in practice, it is not surprising that these studies failed to find any association between their use and the risk of melanoma. Modern high SPF, broad spectrum sunscreens, on the other hand, can be expected to be an effective measure in helping to prevent melanoma compared with sunscreens typical of those used 10 to 20 years ago. It is reasonable to suppose that the improvement in performance of modern sunscreens will lead to a worthwhile benefit as a preventative agent against melanoma, although these benefits may not be seen for several decades. PolydiméthylsiloxaneLe polydiméthylsiloxane —[O-Si(CH3)2]n—, ou poly(diméthylsiloxane) selon la nomenclature systématique, communément appelé PDMS ou diméthicone, est un polymère organominéral de la famille des siloxanes souvent présent dans les shampoings. On l'y ajoute pour augmenter le volume des cheveux mais il peut également aller boucher les pores du cuir chevelu et rendre les cheveux gras. C'est une des raisons pour lesquelles se laver les cheveux tous les jours est très déconseillé avec un shampooing contenant des silicones.
Il existe également de l'amodiméthicone, qui est un dérivé du diméthicone.
Le polydiméthylsiloxane est un additif alimentaire (E900), utilisé comme antimoussant dans les boissons (Coca-Cola BlāK).
La chaîne de poly(diméthylsiloxane) forme également la structure de base des huiles et des caoutchoucs silicones.

SiliconesLes silicones, ou polysiloxanes, sont des composés inorganiques formés d'une chaine silicium-oxygène (...-Si-O-Si-O-Si-O-...) sur laquelle des groupes se fixent, sur les atomes de silicium. Certains groupes organiques peuvent être utilisés pour relier entre elles plusieurs de ces chaines (...-Si-O-...). Le type le plus courant est le poly(diméthylsiloxane) linéaire ou PDMS. It exploits the film-forming characteristics of polydimethylsiloxanes on human skin, ensuring an optimal distribution of the attached UV absorbers when applied as a sun cream. This results in remarkably high performance sun care products which are suitable for all skin typer but especially for sensitive skin due to its minimal skin penetration. Protection cutanéeIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Consumers and patients are highly interested in sunscreens that offer both a protection against sunburn and suberythemal cell damages. We report the combination of encapsulated UVB filters and the broadband filter Titanium Dioxide together with the cell protection factor Ectoin. The UV filter particles are supposed to work on the outermost skin layers to protect from visible endpoints such as UV-induced erythema. Ectoin additionally work as a second defence line to protect beyond visible endpoints against UV-induced celle damages in the epidermis. Aim of the present study is to investigate whether an encapsulation inhibits the contact of UV filters with the body tissue, leading to an increase of the Margin of Safety. On the other hand the performance of non-penetrating inorganic TiO2 in the UVB and especially in the UVA should be determined. Furthermore we wanted to explore the ability of Ectoin to protect against UV-induced damages, which may also occur before occurence of an erythema. Derma absorption experiments were carried out with encapsulated UV filters. Test show a tremendous reduction (70%) in total dermal absorption of encapsulated UV filters compared to non-encapsulated OMC. Using the biological endpoint persistent pigment darkening (PPD), TiO2 (T-2000, T-S) exhibit a very good UVA protection factor of 8 at a 10% use level, while a sun protection factor (SPF) of approx. SPF 20 can be reached. The protection potential of Ectoin against UVA induced damages can be shown: Ectoin reduces the number of sunburn cells in organo-typical skin equivalents and prevents the destruction of Langerhans cells after UV irradiation of human skin in vivo. In conclusion, encapsulated sunscreens inhibit the contact of UV filters with the body tissue, leading to an increase of the Margin of Safety. Non-penetrating TiO2 offers real broad-spectrum protection (UVA and UVB). Finally Ectoin acts as a second defence line in protecting the skin cells from sub-erythemal UV-induced skin damages. A combination of encapsulated UV fileters, Titanium Dioxide and Ectoin leads to sunscreens that take safety aspects into maximum account. Note de contenu : - UV-PROTECTING AGENTS

The traditional classification divides sunscreen actives into organic (soluble) filters, such as a) EHMC, b) OCR, c) BMDBM, d) BEMT, etc, which only absorb UV radiation, and inorganic (insoluble) UV filters, such as TiO2 and ZnO which scatter, reflect and absorb the UV radiation. Now, a new class of organic (insoluble) UV absorbers is emerging: Microfine organic particles dispersed in water. Two candidates of this new class of UV absorbers will be discussed: e) MBBT, which is a broadband UVA1 absorber and a new triazine based compound, which is a research sample having UVB/UVA2 absorption. Both particule UV absorbers show pigment properties, which are close to TiO2 or ZnO, but also the typical high UV absorbance of organic UV filters. The presence of these constituents suggests that this active produces a variety of effects which are also used in phytomedicine. Surprisingly, Belides was found to exhibit a strong inhibitory effect on melanogenesis (patent pending). The fraction of polyphenols is known to inhibit tyrosinase enzymatic activity. results of a comparative study conducted with Belides and aubutin, used at equivalent polyphenol concentrations, revealed that Belides was about twice as active as arbutin. Tests carried out on melanoma cells (B16V) have shown that Belides controls transcription of tyrosinase expression, thus inhibiting its synthesis. In addiction, Belides has been found to significantly decrease the release of the peptide hormone endothelin (ET-1), the binding capacity of alpha-MSH (melanocyte-stimulating hormaone) on the melanocortin receptor-1 (MC1-R) and also melanosome uptake mechanisms. Furthermore, results of a pilot study performed on human volunteers demonstrate the in vivo skin-lightening efficacy of Belides.
