In the present study, a room temperature ionic liquid (IL), 1-butyl-3-methylimidazolium acetate ([BMIM]Ac), was synthesized, and then evaluated as an alternative solvent for type I collagen. Real-time polarizing optical microscope observation indicated complete disintegration of hierarchical structure of collagen aggregates as solubilized in [BMIM]Ac at 25°C. The solubility reached up to approximately 8.0 wt.% at 25°C, more than ten times higher than that in conventional dilute acetic acid. In comparison with dilute acetic acid and recently reported chloridion ILs, high solubility of collagen in [BMIM]Ac at room temperature was ascribed to loose binding between [BMIM]+ and acetate, as well as stronger proton-accepting ability of [BMIM]Ac, which enabled rupture of those intermolecular hydrogen bonds and ionic bonds that stabilized collagen aggregates. However, such bond-rupturing effect was found selective at room temperature. As demonstrated by Fourier transform infrared, circular dichroism, atomic force microscope, and ultrasensitive differential scanning calorimetry analysis, [BMIM]Ac did not destroy the special triple-helical structure of tropocollagen molecules that had been identified as being of importance for the functional and bioactive properties of collagen. According to these results, the discovery of [BMIM]Ac as an ideal solvent for collagen may open up new possibilities for the chemistry and engineering of collagen, which has long been established as a readily accessible and renewable resource with many unique properties. Two of the parameters which represent the capillary radius and the amount of capillaries in a definite area of leather, respectively r and n have been deduced.
Researches on the locational differences in air permeability based on the model, show that different locations of leather have different values for r and n. The capillary radius of samples near the head location ranges from 452.9nm to 479.1nm, while the quantity of capillaries in the same sample is the greatest, from 7.948 x 1011/m2 to 8.617 x 1011/m2. The capillary radius (r) of a sample from a near centre location is the largest at 581.0nm and the capillary amount (n) is the lowest at only 5.262 x 1011/m2. The capillary radii of samples near the tail and centre of the leather are about 530nm.
Neither the thickness nor aperture ratio can successfully represent the air permeability of leathers. The air permeability process is rather complex. ThermogravimétrieIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Genipin is a naturally occurring iridoid compound, it is widely used as an ideal biological protein crosslinking agent due to its low toxicity compared to glutaraldehyde and formaldehyde. Under alkaline condition, genipin could undergo ring-opening polymerization via nucleophilic attack of hydroxyl ions followed by an aldol condensation. Because of the fact that polygenipin could create long-range intermolecular crosslinking between protein chains, preliminary investigations have been carried out to study effect of polygenipin crosslinking on color and thermal stability of hide powder by using colorimetry, differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). The results show that the peak denaturation temperature (Tp) for hide powders obtained from DSC increased to a maximum and then decreased with increasing of the molecular weight of polygenipin. Degree of crosslinking was evaluated, and the results suggest that thermal stability of hide powder is influenced not only by degree of crosslinking, but also by the type of crosslinking. Thermogravimetric analysis also confirms that long-range intermolecular crosslink bridges formed between collagen molecules results in more thermally stable hide powders. This study suggests that polygenipin can be potentially useful in producing eco-friendly leather. ProtéoglycanesUn protéoglycane est une glycoprotéine, combinaison d'une protéine et d'un glycosaminoglycane (GAG). L'association entre les deux types de chaîne s'effectue essentiellement dans l'appareil de Golgi, mais également au niveau du réticulum endoplasmique d'une cellule. La proportion de glucides des protéoglycanes peut atteindre 95 %. Ceux-ci se présentant sous la forme d'une ou plusieurs chaînes de glycosaminoglycanes non ramifiées. Les chaînes de sucres sont très longues mais pas ramifiées. Ils sont O-glycosylés, se lient à l'acide aminé sérine à l'extrémité OH. Les protéoglycanes peuvent être soit transportés à l'extérieur de la cellule par exocytose (s'intégrant alors à la matrice extracellulaire sous forme de chondroïtine-sulfate, kératan-sulfate, héparan-sulfate, dermatan-sulfate, etc.), soit entrer dans la constitution de la membrane plasmique ou du glycocalyx, jouant alors un rôle dans les relations cellule-matrice.
Les PG (protéoglycanes) ont des compositions et poids moléculaire très variés et sont hétérogènes au niveau de leur structure et de leur fonction.
Les protéoglycanes sont des composants essentiels de la matrice extracellulaire. Ce sont des pièges à eau qui sont importants pour les propriétés mécaniques des tissus cartilagineux par exemple. Les héparan sulfates peuvent avoir un rôle dans la signalisation : ce sont des co-récepteurs pour les FGF (Fibroblast Growth Factor). Les protéoglycanes jouent aussi un rôle dans la diffusion des molécules de signalisation (Wnt, Shh), ou bien en interagissant avec des inhibiteurs (Noggin). (Wikipedia)

UréeIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : In the beamhouse, liming might directly affect the structure and performance of collagen, as well as the quality of resultant leather. However, the influences of composition and content of liming agents on liming mechanism are quite complicated. In this study, calcium hydroxide and urea were utilized in liming. The solution pH, hide swelling ratio and non-collagenous protein removal were quantitatively analyzed. The morphologies of both limed and fresh hides were studied by optical microscopy. The reaction mechanism of fiber opening up of cattle hides was analyzed and speculated by the combination of thermogravimetric analysis and Fourier transformation infrared spectroscopy. It was found that the fiber bundles of hides limed by urea/calcium hydroxide have a better opening up effect than that by pure calcium hydroxide. The mechanism of liming in an urea/calcium hydroxide solution system was proposed. GélatineLa gélatine est une substance solide translucide, transparente ou légèrement jaune, presque sans goût et sans odeur, obtenue par l'ébullition prolongée de tissus conjonctifs (peaux) ou d'os d'animaux (principalement porc, bœuf, poisson). Elle possède de nombreuses applications dans le domaine culinaire, la médecine, les industries agroalimentaire et pharmaceutique.
En matière d’étiquetage, la gélatine est considérée par la norme européenne3 comme un ingrédient et non pas comme un additif, c'est pourquoi elle n'a pas de numéro E. Hors Union européenne, elle est considérée par certains pays comme un additif gélifiant et on peut la trouver avec la dénomination E441.
La gélatine est un mélange de protéines obtenu par hydrolyse partielle du collagène extrait de la peau comme la peau de porc (cochon), des os, des cartilages, etc. Les liaisons moléculaires entre les fibres de collagène sont alors brisées. Mélangée à de l'eau, la gélatine forme un gel colloïdal semi-solide thermo-réversible (il fond lorsqu'il est chauffé et recouvre son aspect gélatineux lorsqu'il est refroidi). Sous forme déshydratée, par contre, la gélatine n'a pas de point de fusion et devient friable ou brûle quand elle est chauffée à trop haute températureLa rhéologie de la gélatine se caractérise par un comportement viscoélastique, et des contraintes trop élevées ou appliquées trop rapidement peuvent entraîner une rupture fragile (fracturation) ou ductile6. Le caractère plutôt élastique/fragile ou plutôt visqueux/ductile dépend de la concentration en gélatine de la solution aqueuse et de la température, ainsi que de la durée de la mise sous contrainteLes acides aminés constituant la gélatine sont : la glycine (21 %), la proline (12 %), l'hydroxyproline (12 %), l'acide glutamique (10 %), l'alanine (9 %), l'arginine (8 %), l'acide aspartique (6 %), la lysine (4 %), la sérine (4 %), la leucine (3 %), la valine, la phénylalanine et la thréonine (2 %), l'isoleucine et l'hydroxylysine (1 %), la méthionine et l'histidine (< 1 %) et la tyrosine (< 0,5 %). Ces valeurs sont variables (surtout pour les constituants minoritaires) et dépendent de la source de matériaux bruts et de la technique de préparation. La gélatine est constituée à environ 98-99 % (en poids sec) de protéines et contient 18 acides aminés dont huit des neuf acides aminés essentiels à l'Homme. Elle n'a qu'une relative valeur nutritionnelle du fait de l'absence de tryptophane et de son déficit en isoleucine, thréonine et méthionine; elle possède également un taux inhabituellement élevé d'acides aminés non essentiels, la glycine et la proline (qui sont produits par le corps humain). Tannage Résumé : Collagen fibers obtained from solid fibrous wastes generated in tannery have a high potential of being used in developing green composites. Earlier studies in our laboratory demonstrate that nonwoven composites can be derived from collagen fiber network using paper-making technology. The purpose of this work was to fabricate green composites based on gelatin and vegetable-tanned collagen fibers (VCF), using a non-toxic and naturally occurring compound, genipin, as cross-linking agent. VCF were obtained from split hide that has been tanned with vegetable tannins by mechanical milling technique. The VCF content in the networks of gelatin was varied from 10 to 40% (w/w) in dry weight relative to that of the gelatin. The structure, mechanical properties, thermal properties and water resistance of the composite films have been investigated with Fourier transform infrared spectroscopy (FTIR), mechanical testing, thermogravimetric analysis and swelling measurement. The results from those studies revealed that VCF and gelatin could mix with each other homogeneously under alkaline condition and the mechanical properties of the composite films at wet state were improved significantly over that of pure gelatin film. In addition, the composite films showed higher water resistance and thermal resistance than the control. FTIR analysis confirmed the molecular interactions between gelatin and VCF and the formation of cross-links between primary amino groups on gelatin and VCF. Our results indicated that VCF can be used as environmentally friendly and cost-effective potential reinforcing agent for green composites, providing better properties than the original biopolymer matrix. The genipin cross-linked gelatin/VCF composite film can be a promising candidate for the biomedical and packaging applications.
