From the water profile across the epidermis the border between stratum corneum and stratum granulosum can be estimated. This is due to the steep drop in water concentration from the inner to the outer side of the stratum corneum. Water content drops from approximately 70% at the inner stratum corneum to only 30% at the skin surface. This slope of the curve becomes clearly flatter in the stratum granulosum. A second parameter is usually taken from confocal Raman spectroscopy to define the stratum corneum border. This is the content of natural moisturization factor (NMF), which should be present only in the stratum corneum. Located at the depth at which the NMF content levels off and the slope of the water profile curve changes is the stratum corneum border. The goal of this work was to develop stratum corneum thickness detection into a robust and semi-automated measurement relying only on the water profile. Further, the aim was to base the empirical findings of water distribution in the epidermis on a well established theory, Fick’s law of diffusion. A mathematical model was developed to fit the water profile curve for a robust and automated detection of the stratum corneum border. In addition, the new model automatically resulted in an accurately determined slope of the water concentration curve in the stratum corneum. This slope, or more exactly the gradient, is one of two parameters directly related to transepidermal water transport across the stratum corneum. In the present study we investigated the role of epidermal tight junctions in two skin physiological processes, "formation of the epidermal calcium ion (Ca2+) gradient" and polarized lamellar body secretion". The former is based on the fact that the epidermal Ca2+ gradient is closely related to epidermal differentiation and the latter on the knowledge that polarized lamellar body secretion is essential to supply intercellular lipids to the stratum corneum. We hypothesized that tight junctions might form the Ca2+ gradient in the epidermis by sealing cell-cell contact at the stratum granulosum and allow the lamellar bodies to be normally secreted toward the stratum corneum by giving granular cells polarity. In our experiments we discovered that tight junctions control not only both intercellular Ca2+ permeability and fluorescent ceramide analog (Cer-FL) secretion in cultured normal human keratinocyte cells but also both intercellular calcium distribution and polarized lamellar body secretion in the skin equivalent. Thus, tight junctions should be responsible for linking a series of processes from epidermal differentiation to stratum corneum barrier formation. Our findings suggest that tight junctions should have a crucial role not only in epidermal barrier function but also in both epidermal differentiation and stratum corneum barrier function. On a utilise la perte d'eau transepidermique et l'hydratation de la couche cornee comme parametres d'evaluation de l'influence du traitement sur la fonction protectrice. On a en outre effectue apres application un test de la fonction protectrice avec un ester d'acide nicotinique. On a constate des effets stabilisateurs de la fonction protectrice apres application seulement sur peau atopique et apres extraction lipidique mais pas sur peau saine. Les resultats nous conduisent a esperer des effets positifs uniquement en cas d'alteration de la fonction protectrice de la peau. Transmission electron microscopy, scanning electron microscopy, and X-ray diffraction were carried out to analyze the as-sprayed coatings and powders. Mercury intrusion porosimetry was applied to analyze the pore size distribution. Laser flash technique and differential scanning calorimetry were used to examine the thermophysical properties of the nanostructured coatings. The results demonstrate that the as-sprayed nanostructured zirconia coatings consist of the nonequilibrium tetragonal phase. The microstructure of the nanostructured coatings includes the initial nanostructure of powder and columnar grains. Moreover, micron-sized equiaxed grains were also exhibited in the nanostructured coatings. Their evolution mechanisms are discussed. The as-sprayed nanostructured zirconia coating shows a bimodal pore size distribution, and has a lower value of thermal conductivity than the conventional coating. La protection contre les rayons U.V. comprend souvent la production de composes absorbeurs d'U.V. qui, chez les organismes marins, sont en général des acides aminés de type mycosporine (MAA) ayant des maxima d'absorption compris entre 310 et 360 nm. Les coraux d'eau peu profonde produisent de grandes quantités de MAA et leur capacité apparente a supporter une exposition aux U.V. de longue durée suggéré qu'ils utilisent leur chromophore absorbeur d'U.V. à des fins d'écran solaire. Nous présentons une vue d'ensemble de la photophysiologie biochimique de coraux tropicaux formant des récifs, et du développement d'une nouvelle classe d'écran solaire synthétique, les 1-alkyl-3-alcanoyl-1,4,5,6-tétrahydropyridines, basée sur cette protection naturelle contre les U.V. Nous avons analyse la qualité d'écran solaire de plusieurs candidats et teste une sélection limitée pour son usage potentiel dans des produits cosmétiques et dermatologiques pour l'homme.
