Pigmentation de la peauTags : 'Anti-âge' L* 'Age de la peau' 'Eclat 'Couleur Index. décimale : 668.5 Parfums et cosmétiques Résumé : Une étude de vieillissement a été conduite en utilisant la capturer des paramètres de couleur de peau dans le système Lab sur des sujets caucasiens des deux sexes et tous âges adultes disponibles. Cette étude a montré une corrélation linéaire entre L* et l’âge comme suit (L* la valeur) = −0.13 × (l’âge en années) +63.01. Des études précédentes ont mis en évidence des changements de la couleur de peau liés à l’âge. Ce travail présente un nouveau modèle linéaire quantitatif corréléà la luminosité de la peau par lequel il est possible de communiquer au grand public les changements liés à l’âge. Deux études d’évaluation de produits sont aussi présentées ici, démontrant la capacité de produits anti-âges à apporter des améliorations objectives et subjectives dans la luminosité de la peau. Il a été montré qu’il est possible d’utiliser la corrélation âge, valeur fondamentale L* pour décrire chez les caucasiens des avantages produit d’une nouvelle façon quantitative et appropriée au grand public, par la description d’un calcul d’un “recul d’années”. The CC-516 has a high proportion of proline (13%) of free amino acid content. The inhibitory effect of CC-516 on the elastase exhibited 37 to 90% inhibition by 10 to 250 μg/ml concentration ; the IC values with 40.8 μg/ml for porcine pancreatic elastase (PPE) and 48.1 μg/ml for human leukocyte elastase (HLE), respectively. One of the effects of elastase is that it is known to reduce the number of elastin fibers at the level of the enzyme deposition. The number of elastin fibers was increased when we drift from the deposit number of elastase with 100 mg/ml of CC-516. CC-516 showed protection of elastic fiber against degradation by the enzyme in an ex vivo assay. The CC-516 increased proliferation of human fibroblast cell by 85% at 10 concentration, compared with control, whereas the increase by ascorbic acid was 50%. The collagen synthesis was increased by 40% at 10% concentration of CC-516. The treatment with CC-516 improved skin hydration, the skin elasticity, and skin wrinkles. From this study, we suggest that CC-516 can be used as a new anti-aging component for cosmetics. Their expression increases with aging and is further enhanced by UV irradiation. Blackberry (Rubus fruticosus) leaf extract has been shown to suppress MMP-1, -2 and -9 in cell-free assays. We have now further explored the activity of this extract. The effect on MMP-1 expression at the protein and the mRNA level was investigated using non-irradiated and UVA-irradiated human dermal fibroblasts. The extract showed a dose-dependent reduction of the MMP-1 protein level of both irradiated and non-irradiated cells with an almost complete inhibition at a dosage of 0.1%. MMP-1 mRNA expression of UVA-irradiated cells was decreased after pre-incubation as well as after post-incubation with the extract. Best results were obtained with combined pre- and post-treatment (0.1% extract led to 59% reduction in comparison with the respective control). Moreover, we found that blackberry leaf extract inhibits IL-1α, a cytokine known to induce MMP expression. At a dosage of 0.2% the inhibition was 40% compared with that of the stimulated control. The extract also exhibited a potent radical scavenging activity comparable to that of α-tocopherol as was shown by the ABTS assay. Taken together, these biological activities make blackberry leaf extract a highly efficient multifunctional anti-aging active. The need for high efficacy paired with excellent stability appeared contradictive but was resolved by designing complete redox systems rather than antioxidants alone. We here introduce a new class of cosmetic redox pairs, each comprising the initial antioxidant plus its oxidation product. The chemical properties of the oxidation product are the key to enabling the system to maintain or even increase its initial efficacy. A series of antioxidant tests were used to carefully characterize single redox pair components for comparison with traditional antioxidants such as vitamin C and E derivatives (DPPH, lipid and TEAC assays). Kinetic studies on the photobehaviour of a selected redox pair were carried out in cosmetic emulsions with solar simulation and monitored by UV spectroscopy and high performance liquid chromatography. Hydroxy dimethoxybenzyl malonate (HDBM) is a pure transparent cosmetic oil. It shows excellent cosmetic stability coupled with extraordinary antioxidant properties. This controlled activity can be explained well by the presence of its oxidation product hydroxy dimethoxybenzylidene malonate (HDBMox), which provides ongoing antioxidant activity plus excellent UVA absorbing properties. The redox pair described here provides the advanced on-demand benefit of a non-UV-absorbing photoprotector that is converted into its UV-absorbing equivalent upon light exposure. Cependant, même si de nombreuses stratégies ont été proposées pour lutter contre le vieillissement cutané ou son impact esthétique depuis le début de l’ère de la « cosmétique scientifique », rares sont les produits dont l’efficacité découle de l’observation de situations pathologiques. La maladie la plus « visiblement » liée au vieillissement cutané (notamment) est la progéria, ou syndrome de Hutchinson-Gilford, qui se caractérise par un vieillissement accéléré conduisant rapidement à la mort du sujet. Dans cette pathologie la lamine-A, une protéine participant (avec d’autres lamines) à la formation de la lamina nucléaire et impliquée dans la stabilité du noyau, de la structure de la chromatine et de l’expression des gènes, est présente sous une forme tronquée appelée progérine. Dans cette étude, après avoir mis au point un dosage ELISA sensible et spécifique de la progérine, nous avons pu quantifier cette protéine dans des cellules cutanées humaines normales issues de sujets jeunes et matures. Parallèlement à l’étude de la production cellulaire de lactate choisi comme marqueur du vieillissement, nous proposons donc ici la première étude quantitative de l’expression cutanée de la progérine au cours du vieillissement. De plus, alors que les extraits d’algues sont aujourd’hui largement utilisés en cosmétiques afin de lutter contre le vieillissement de la peau, nous avons pu démontrer qu’un extrait d’Alaria esculenta était capable de significativement réduire l’expression de la progérine dans des cellules cutanées issues de sujets matures. Enfin, et puisque cet extrait d’algue est déjà commercialement disponible, nous confirmons qu’une meilleure compréhension des pathologies cutanées représente un outil puissant pour le développement de composés efficaces et pertinents utilisables en cosmétique, mais pas uniquement.
