Dans cette étude, nous avons étudié les effets de licoricidin sur le photovieillissement des fibroblastes dermiques humains UVA-irradiés (HDFS).
L'activité in vitro de piégeage des ROS, l'effet protecteur cellulaire et l'inhibition de l'activité de l’élastase ont été déterminées par des systèmes de Fe3+ -EDTA / H2O2, par photohemolyse et par dosage de l'activité de l’élastase, respectivement. La capacité anti-oxydante du composé a été évaluée par ELISA fluorescent et dosage du 2 ‘, 7’-dichlorofluorescin-diacétate (DCF-DA). L'expression de la protéine et la phosphorylation ont été examinées en utilisant Western Blot.
La capacité antioxydante totale (OSC50) de licoricidin était de 2,77 μM. Il était de 3,1 fois plus élevée que celle de l'acide L-ascorbique. Ses effets protecteurs ont été confirmés dans une étude des dommages cellulaires 1O2- induite par les érythrocytes humains. La valeur τ50 de 10 μM de licoricidin était de 71,0 min; ceci est nettement plus élevé que celle obtenue avec les α-tocophérol (37,0 min). L'activité inhibitrice de l’élastase de licoricidin (CI50 de 61,2 μM) était de 2,1 fois plus puissante que celle de l'acide oléanolique. Licoricidin réduit nettement les ROS intracellulaires induits par les UVA d'une manière dépendante de la concentration. Western blot a montré que licoricidin atténue l'induction UVA-dépendante de la protéine MMP-1. Du point de vue mécanistique, ceci semblait être due à l'inhibition licoricidin dépendante de la phosphorylation de MAPK, ce qui a entraîné une diminution de l'activation de c-Jun et c-Jun réduite et l'expression de c-Fos.
La licoricidin bloque le vieillissement entraîné par les UVA par un captage des ROS. Cette activité converge pour limiter l'activité de MMP-1. C'est donc un agent blanchissant efficace qui sert de désinfectant et (à haute concentration) d'oxydant ou monergol dans les fusées spatiales.

In this study, both H2O2 (450μM) and UVC (40J/m2) irradiation-induced cell death were detected by MTT assay, and nicotinamide showed the dose dependent inhibition on H2O2 and UVC induced cell death. Morphological study by microscopic observation showed that H2O2 or UVC induced cellular morphological changes including appearence of rounded cells or swelling cells, characteristic og apoptosis. In the presence of nicotinamide, H2O2 and UVC induced morphological changes were attenuated significantly. Prevention of cells from H2O2 and UVC induced cell death was asociated with decrease of apoptotic bodies by flow cytometry analysis and attenuation of the appearance of DNA ladders by DNA electrophoresis. Induction of p53 protein was detected in H2O2-treated NIH3T3 cells and nicotinamide suppressed its increase. However, bcl-2, bax and α-tubulin proteins were remained unchanged. ROS have been demonstrated to be involved in the H2O2 and UVC induced apoptosis. In this study, H2O2 or UVC irradiation increase intracellular peroxide levels detected by DCHF-DA assay and fluorescent microscopic observation, and nicotinamide blocked the peroxide level induced by H2O2 or UVC irradiation. Decrease of H2O2 or UVC-induced 8-OH dG in DNA was also detected in nicotinamide trated cells. The results of present study firstly demonstrated that nicotinamide prevented cells from H2O2 and UVC irradiation induced apoptosis through suppression ROS production, p53 elevation and 8-OH dG formation. Compared to Japanese women living in Akita, Japanese women living in Kagoshima had significantly longer facial wrinkles, higher number of wrinkles, larger hyperpigmented spots, higher number of spots, rougher facial skin texture, more yellow foreheads and upper inner arms, darker foreheads, and less stratum corneum hydration in the cheeks and arms. When compared on an age-for-age basis, the average 40 year-old Kagoshima women has the same level of facial wrinkling as a 48 year-old Akita women, an acceleration of 8 years for living in the more sunexposed southern latitude. For facial hyperpigmentation, the increase in onset is 16 years; the average 40 year-old Kagoshima women has the same level of facial hyperpigmentation as a 56 year-old Akita women. The results further testify to the skin damaging effects of sun exposure and may be useful in public health education to promote everyday sun protection.
