TélomèresUn télomère est une région hautement répétitive, donc non codante, d'ADN à l'extrémité d'un chromosome. À chaque fois qu'un chromosome en bâtonnet d'un eucaryote est dupliqué, lors de la réplication, qui précède la mitose (division cellulaire), le complexe enzymatique de l'ADN polymérase s'avère incapable de copier les derniers nucléotides : l'absence de télomère signifierait la perte rapide d'informations génétiques nécessaires au fonctionnement cellulaire.
Les télomères raccourcissent avec l’âge, l’inflammation et le stress. Des études ont montré que des télomères courts sont associés à un risque plus élevé de maladies liées à l’âge.

I would summarise the achievable goals for non-prescription products as wrinkle reduction, reduction of sebaceous pore diameter, skin smoothing, skin textural improvement of dyspigmentation.
Daily use of effective photoprotective products have been clearly shown to reduce accelerated skin ageing as well as skin cancers and pre-skin cancers.
There are goals that I consider unrealistic and include claims for nonprescription products being able to lift faces, lift and enlarge busts, firm and lift buttocks, cure cellulite, and get rid of enlarged veins and telangiectasia.
At present the technology is not available in my opnion to achieve these goals.

Protection cutanéeIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Differentiation with the sun care market has traditionnaly been driven by various factors in the market place, be these formats (sprays, lotions gels etc.), end user (baby, sports, sensitive), application site (body, face hair and scalp) or added value (sand resistance, insect repellent). As we learn more about the effects of UV, and other radiation, on the skin it is encouraging to see that health benefits for the consumer are still the major driver - at the end of the day (or more like during the middle of the day) the products must provide as much protection as possible. As well as the challenge of potential new wavelengths to consider, formulators must also address the tactile and sensory aspects of their products to ensure consumers apply as much product as is necessary to achieve the optimum functionality of the product and also that users comply with any pack instructions such as regular reapplication ot the product over the course of a day. KératinocytesLes kératinocytes sont des cellules constituant 90 % de la couche superficielle de la peau (épiderme) et des phanères (ongles, cheveux, poils, plumes, écailles). Ils synthétisent la kératine (kératinisation), une protéine fibreuse et insoluble dans l'eau, qui assure à la peau sa propriété d'imperméabilité et de protection extérieure.
L'épiderme est divisé en 4 couches basées sur la morphologie des kératinocytes (de l'intérieur vers l'extérieur) :
1. stratum germinativum (couche basale à la jonction avec le derme)
2. stratum spinosum
3. stratum granulosum
4. stratum lucidum
5. stratum corneum
Les kératinocytes passent progressivement de la couche basale vers les couches supérieures par différenciation cellulaire jusqu'au stratum corneum ou ils forment une couche de cellules mortes nommées squames, par apoptose. Cette couche constitue une barrière de protection et réduit la perte d'eau de l'organisme.
Les kératinocytes sont en perpétuel renouvellement. Ils mettent environ 1 mois pour aller de la couche basale au stratum corneum mais ce processus peut être accéléré en cas d'hyperprolifération de kératinocyte (psoriasis).
Mélanocytes
Protection contre le rayonnement ultraviolet
Protection cutanéeIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Le stem cell factor (SCF) et son récepteur à activité tyrosine–kinase, nommé KIT, sont impliqués dans divers processus biologiques et en particulier dans la mélanogenèse. En effet, cette voie de signalisation contrôle la migration des mélanoblastes depuis la crête neurale durant le développement embryonnaire et participe à l’âge adulte, à la communication entre les kératinocytes et les mélanocytes. Au niveau des mélanocytes, la liaison de SCF à son récepteur membranaire entraîne l’activation de voies de signalisation impliquant des protéines kinases qui, au final, vont réguler l’expression de gènes impliqués dans les processus de pigmentation. Nous avons développé un composé nommé IV09.007, que nous avons précédemment décrit comme présentant des propriétés de modulation de la voie SCF/KIT de transduction du signal, avec un effet pro-pigmentant. Dans la présente étude, nous avons examiné l’expression des ARN messagers et la localisation des protéines SCF et KIT, au niveau cutané et dans diverses lignées cellulaires originaires de la peau. Nous avons ensuite exploréà l’aide d’une approche par microarray la capacité du composé IV09.007 à moduler l’expression de gènes dans des kératinocytes et des mélanocytes humains en culture. De cette manière, nous avons pu mettre en évidence la régulation de gènes impliqués dans la réparation de l’ADN; principalement des gènes associés aux voies de réparation par excision de bases ou de nucléotides. Une réponse transcriptionnelle modifiée a aussi été observée pour certains gènes impliqués dans la lutte contre le stress oxydant, dans l’inhibition de l’apoptose et la diminution de la réponse immune inflammatoire. Ces résultats de microarray prédisant au composé IV09.007 la capacité de conférer un effet protecteur, nous avons vérifié cette hypothèse par des tests de comètes sur des kératinocytes et des mélanocytes irradiés par des UVB, et ce afin de démontrer l’efficacité de IV09.007 à prévenir les dommages à l’ADN.
