Peau -- PhysiologieIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - OBJECTIF : Les cellules de papilles dermiques humaines (HDPCs) jouent un rôle essentiel dans la morphogenèse folliculaire des cheveux et des cycles postnatales de croissance des cheveux. Des rapports antérieurs ont démontré que le facteur de croissance AA dérivé des plaquettes (PDGF-AA) améliore la formation de condensats dermiques pendant le développement folliculaire des cheveux. En outre, PDGF-AA induit/maintient la phase anagène du cycle pilaire. Il est probable que la morphologie et la fonction mitochondriale sont étroitement couplées avec le maintien de ces activités exigeant de l'énergie. Toutefois, on en sait peu sur la régulation mitochondriale dans les HDPCs. Ainsi, nous avons étudié la régulation mitochondriale impliquant le PDGF dans HDPCs.
- METHODES : Les morphologies des HDPCs mitochondriales ont été examinées en présence ou en l'absence de PDGF-AA sous un microscope à fluorescence. Nous avons étudié la production d'ATP cellulaire et la motilité. La relation entre la morphologie mitochondriale et les fonctions cellulaires a été discutée.
- RESULTATS : Nous avons observé que les HDPCs primaires contenaient des mitochondries aux morphologies filamenteux et/ou arrondies. Les deux types de mitochondries ont montré des potentiels de membrane similaires. Fait intéressant, en présence de PDGF-AA, mais pas du PDGF-BB, l'équilibre entre les deux morphologies est déplacé vers la forme filamenteuse. En même temps, à la fois l'activité enzymatique mitochondriale et le niveau de l'ATP cellulaire total ont été augmentés par PDGF-AA. Ces deux paramètres sont étroitement corrélés, suggérant l'implication du PDGF augmenté dans la production mitochondriale d'ATP. De plus, le PDGF-AA a accéléré la migration des HDPCs dans un test de 'scratch', mais ne modifie pas le taux de prolifération cellulaire. Stress oxydatifIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - OBJECTIF : Examiner la capacité d'un extrait issu de la médecine traditionnelle chinoise, le Polygonum multiflorum Radix, à protéger la viabilité des mélanocytes soumis à un stress oxydant, un mécanisme clé dans l'initiation et la progression du blanchissement du cheveu.
- METHODES : Dans le but de tester le potentiel antioxydant de l'extrait Polygonum multiflorum Radix, nous avons traité des mélanocytes humains normaux issus de cultures primaires de prépuce avec une version commerciale de l'extrait de Polygonum multiflorum Radix, puis exposé les cellules au peroxyde d'hydrogène (H2O2). La concentration en espèces réactives de l'oxygène et le ratio glutathion/protéines ont été déterminés pour objectiver l'effet antioxydant de l'extrait. Afin d'améliorer sa solubilité pour une utilisation cosmétique, un nouvel extrait du Polygonum multiflorum Radix a été développé. Le blanchissement des cheveux étant la conséquence d'une disparition des mélanocytes au sein des follicules pileux, nous avons vérifié si le potentiel anti-oxydant du nouvel extrait de Polygonum multiflorum Radix préserve la viabilité des mélanocytes soumis à un stress oxydant de type H2O2, ainsi que la pigmentation au sein de follicules pileux humain.
- RESULTATS : Le traitement de mélanocytes humains normaux issus de cultures primaires de prépuce par la version commerciale de l'extrait de Polygonum multiflorum Radix, a entrainé une diminution de l'accumulation des espèces réactives de l'oxygène intracellulaires en réponse à une exposition à l'H2O2, ainsi qu'une préservation du ratio glutathion/protéines. Ce résultat confirme un potentiel protecteur de l'extrait vis-à -vis des mélanocytes soumis à un stress induit par H2O2 et suggère une capacité à prévenir la mort des mélanocytes face à un stress oxydant. Une diminution de la mort cellulaire a été confirmée lorsque les mélanocytes sont traités par l'extrait amélioré de Polygonum multiflorum Radix, démontrant ainsi un effet cytoprotecteur. Enfin, le contenu en mélanine de follicules pileux humains était significativement supérieur après traitement par la version améliorée de l'extrait de Polygonum multiflorum Radix, en comparaison au contrôle traité par le véhicule, confirmant ex vivo l'effet protecteur de l'extrait sur la pigmentation du cheveu.
- CONCLUSION : L'extrait de Polygonum multiflorum Radix protège in vitro les mélanocytes humains normaux des effets délétères liés à l'exposition au stress induit par H2O2 et a un impact bénéfique sur la pigmentation au sein des follicules pileux humains ex vivo, montrant l'intérêt de l'extrait de Polygonum multiflorum Radix en tant qu'ingrédient actif pour la protection des mélanocytes contre une mort prématurée. Tests de sécuritéIndex. décimale : 668.5 Parfums et cosmétiques Résumé : -Objectif : Ce travail présente l’évaluation des performances du test RHE/IL-18, réalisé sur le système d’essai SkinEthicTM RHE dans le but d'identifier les sensibilisants cutanés.
- Méthodes : Un ensemble de 18 substances et mélanges a été testé sur ce modèle d’épiderme en suivant le protocole du test RHE/IL-18. Les résultats ont été évalués selon 5 schémas d’interprétation pour déterminer le modèle prédictif optimal pour ce système d’essai. Une attention particulière a été accordée à l’analyse du taux basal de relargage d’IL-18 et aux calculs de performances obtenues par rapport à trois références : LLNA, HRIPT et une référence composite prenant en compte l'ensemble des résultats disponibles.
- Résultats : Les performances du test obtenues avec le système d’essai SkinEthicTM n’ont pas montré de différences importantes selon les trois références utilisées, par ailleurs, l’analyse de ces performances montre que ce système d’essai pourrait être considéré comme une alternative à d’autres épidermes reconstruits (Epi-DERMTM, EpiCSTM, et VUMC-EE) en évaluation pour la réalisation du test RHE/IL-18.
- Conclusion : Le modèle de prédiction à utiliser avec le système d’essai SkinEthicTM a été amélioré. Vieillissement cutané -- PréventionIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : One of the main approaches to preventing skin ageing is to protect fibroblast cells from oxidative stress. The promoting effect of the human amniotic membrane extract (hAME) on re-epithelization, proliferation and migration of cells in wound healing has been already well studied. This experimental study aimed to investigate the antioxidant activity of hAME against hydrogen peroxide (H2O2)-induced dermal fibroblast damage.
- Methods : Here, to establish the ageing model, human foreskin fibroblasts (HFFs) were exposed to 200 μM H2O2 for 2 h. HFFs were treated with 0.1 mg/ml AME for 24 or 48 h before or/and after H2O2 exposure. A total of 48 h following the H2O2 treatment, we measured cell proliferation, viability, senescence-associated β-galactosidase (SA-β-Gal), antioxidants and preinflammatory cytokine (IL-6) levels, as well as the expression of senescence-associated genes (P53 and P21).
- Results : The obtained results indicated that under oxidative stress, AME significantly increased cellular viability and not only promoted the cell proliferation rate but also attenuated apoptotic induction condition (p < 0.001). AME also significantly reversed the SA-β-Gal levels induced by H2O2 (p < 0.001). Additionally, both pre- and post-treatment regimen by AME down-regulated the expression of senescence marker genes (p < 0.001). Moreover, AME declined different oxidative stress biomarkers such as superoxide dismutase and catalase and increased the glutathione amount.

TranspirationIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : Axillary wetness represents an unwanted effect of the physiologically vital sweating mechanism, especially when it becomes excessive. Cosmetic products reducing sweat secretion rely on aluminium salts as the active ingredient acting by physically blocking the sweat gland. Driven by the interest to better understand the sweat mechanism and to develop alternative technologies against excessive sweating a search for an effective testing approach started as up to now, cost- and time-consuming in vivo studies represent the standard procedure for testing and identifying these alternatives.
Material and methods : The herein described in vitro test system is based on the measurement of intracellular changes of the ion equilibrium in cultured eccrine sweat gland cells. Subsequently, in vivo studies on the back of volunteers were conducted to verify the sweat-reducing effect of in vitro newly discovered substance.
- Results : In this study, we describe an effective cell-based in vitro method as a potent tool for a more targeted screening of alternatives to aluminium salts. Testing the commonly used aluminium chlorohydrate as one example of an aluminium-based active in this screening procedure, we discovered a distinct influence on the ion equilibrium: Intracellular levels of sodium ions were decreased while those of chloride increased. Screening of various substances revealed a polyethyleneimine, adjusted to pH 3.5 with hydrochloric acid, to evoke the same alterations in the ion equilibrium as aluminium chlorohydrate. Subsequent in vivo studies showed its substantial antiperspirant action and confirmed the high efficiency of the polyethyleneimine solution in vivo. Further, specific investigations connecting the chloride content of the tested substances with the resulting sweat reduction pointed towards a substantial impact of the chloride ions on sweating.
- Conclusion : The newly described in vitro cell-based screening method represents an effective means for identifying new antiperspirant actives and suggests an additional biological mechanism of action of sweat-reducing ingredients which is directed towards unbalancing of the ion equilibrium inside eccrine sweat gland cells.Note de contenu : - MATERIALS AND METHODS : Ethics statement - Cell culture - Determination of cell viability - Ion flux assays with fluorescence dyes - Statistical analysis
