VieillissementIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Contexte : Les fibroblastes sont une composante importante de la peau qui détermine ses propriétés. Le N-Acétylglucosamine (NAG) est le substrat pour la synthèse de l'acide hyaluronique et il a également une activité anti-inflammatoire et anti-sénescente dans les cellules mésothéliales.
Methozdes : Nous avons testé dans des fibroblastes de peau humaine en culture, comment la supplémentation du milieu de culture avec 10 mmol L−1 de NAG modifie les propriétés de ces cellules.
Resultats : les fibroblastes cultivés en présence de NAG produisent plus de protéines, principalement due à l'augmentation de la synthèse du collagène (+33% par rapport au témoin, P < 0.05). La synthèse d'acide hyaluronique a été augmentée (+107% par rapport au témoin, P < 0.001), mais l'interleukine-6 synthèse a été réduite (−22% par rapport au témoin, P < 0.05). Les fibroblastes cultivés dans un milieu avec NAG 10 mmol L−1 démontrent une meilleure capacité à guérir la couche de cellules blessées (+34% par rapport au témoin, P < 0.05). En outre, la sénescence réplicative des fibroblastes qui subissent le vieillissement en présence de NAG a été moins prononcée, comme en témoigne la moindre augmentation du temps de doublement de population (−70% par rapport au témoin, P < 0.05).
Conclusion : Nous concluons que les changements induits par NAG dans les propriétés des fibroblastes cutanés peuvent être importants pour la prévention des changements dus au vieillissement de la structure et la fonction de la peau.Note de contenu : - Effect of NAG on function of fibroblasts

StatistiqueIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Skin ageing is an inevitable physiological process, determined by several concomitant factors, resulting in a thinner and drier skin, in the formation of fine wrinkles, and a gradual dermal atrophy.
During this process the skin cells become senescent, they slow down their metabolism and the production of structural matrix components. The Growth Differentiation Factor 11 (GDF11) has been recently identified as a pro-youth factor in several cell types, and in particular it was shown its involvement in the restoration of a young phenotype in skin fibroblasts, by stimulating important ExtraCellular Matrix proteins. Furthermore, its key role in modulating mitochondrial functions in skin cells during ageing has been highlighted. In the present article, we describe the characterisation of a plant extract, obtained from Rosa rugosatissue cultures, able to produce a ‘rejuvenating’ effect in aged skin fibroblasts by inducing GDF11 expression and synthesis. As a new nonionic and amphiphilic ascorbic-acid derivative, 2-glyceryl-3-octyl ascorbate (GO-VC), which is synthesised by introducing an octyl and a glyceryl group into ascorbic acid structure, has been recently developed. Due to its unique chemical structure, in addition to the effects of AsA, GO-VC is expected to have a humectant effect derived from glycerin and an anti-bacterial property derived from octanol, and also is highly likely to have a fibroblast activation ability, and a melanogenesis-suppressing and skin pore and wrinkle improving effect. GO-VS's safety tests, which include a human skin-sensitivity test [repeat insult patch test (RIPT)], ocular irritation test [bovine corneal opacity and permeability (BCOP) assay], genetic toxicity (Ames test), and clinical human skin patch test, have been performed, confirming the safety of GO-VC. "Alors que la génétique correspond à l'étude des gènes, l'épigénétique s’intéresse à une "couche" d’informations complémentaires qui définit comment ces gènes vont être utilisés par une cellule ou… ne pas l'être".
"C'est un concept qui dément en partie la "fatalité" des gènes".
Fibroblastes
Fruit du moineLe fruit de moine ou fruit du moine (Siraitia grosvenorii), est une plante grimpante pérenne originaire du sud de la Chine. C'est l'une des quatre espèces du genre Siraitia. Il est connu sous le synonyme de Momordica grosvenorii et Thladiantha grosvenorii. Les autres espèces du genre Siraitia sont Siraitia siamensis poussant en Thaïlande, Siraitia sikkimensis et Siraitia silomaradjae poussant en Inde, et Siraitia taiwaniana poussant à Taïwan.
La plante pousse de 3 à 5 m de long, s'élevant verticalement en s'accrochant et en s'enroulant sur les autres végétaux. Les feuilles en forme de cÅ“ur font de 10 à 20 cm de long. Le fruit de forme ovale de 5 à 7 cm de diamètre contient une pulpe sucrée, comestible ainsi que de nombreuses graines. Son fruit est plus connu sous son nom chinois luo han guo (罗汉果, 羅漢果, luóhà n guǒ, "fruit du arhat").
Un extrait du fruit est 300 fois plus sucré que le saccharose et est utilisé comme édulcorant naturel alimentaire et un ingrédient de la médecine chinoise traditionnelle. Des hétérosides au pouvoir sucrant intense, le mogroside V, mogroside IV et le siamenoside I qui sont responsables du goût sucré du fruit.

This novel ingredient is based on the Monk fruit (Momordica grosvenorii) which is cultivated in a mountain valley in Guangxi, a place known by the striking longevity and happiness of its inhabitants.
Since emotions are directly linked to the subconscious, the intention was to measure objectively the emotional impact of using this active ingredient in a cosmetic application. To do so, our laboratory used scientific rigour and Artificial Intelligence (AI) taking into account such subconscious evaluations.
But overall wellbeing is seen as integrative beauty that embraces both the emotional health and the physical aspect. Consequently, our laboratory also took a closer look at the causes for aging. While many people say it is all in the genes, Epigenetics suggests that not everything is genetically predisposed. Lifestyle has a major impact and maintaining good habits like exercising, eating plenty of fruits and vegetables, not smoking, and drinking alcohol in moderation can have very positive effects on the aging process.Note de contenu : - Introduction : Epigenetics

KératinocytesLes kératinocytes sont des cellules constituant 90 % de la couche superficielle de la peau (épiderme) et des phanères (ongles, cheveux, poils, plumes, écailles). Ils synthétisent la kératine (kératinisation), une protéine fibreuse et insoluble dans l'eau, qui assure à la peau sa propriété d'imperméabilité et de protection extérieure.
L'épiderme est divisé en 4 couches basées sur la morphologie des kératinocytes (de l'intérieur vers l'extérieur) :
1. stratum germinativum (couche basale à la jonction avec le derme)
2. stratum spinosum
3. stratum granulosum
4. stratum lucidum
5. stratum corneum
Les kératinocytes passent progressivement de la couche basale vers les couches supérieures par différenciation cellulaire jusqu'au stratum corneum ou ils forment une couche de cellules mortes nommées squames, par apoptose. Cette couche constitue une barrière de protection et réduit la perte d'eau de l'organisme.
Les kératinocytes sont en perpétuel renouvellement. Ils mettent environ 1 mois pour aller de la couche basale au stratum corneum mais ce processus peut être accéléré en cas d'hyperprolifération de kératinocyte (psoriasis).
