N-acétyl aspartique, AcideIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - CONTEXTE : L'acide aspartique acétylé (A-A-A) a été identifié en utilisant la technologie des puces à ADN combinée a une analyse en 'connectivity mapping' (Cmap), pour ces propriétés potentielles anti-âge. Ces propriétés ont été testées par une approche in silico et in vitro sur modèles cellulaires humains.
- METHODES : La prédiction in silico, utilise un logiciel de relation 'Structure - Activité', qui permet d'orienter la conception des expérimentations in vitro. Ces tests ont été réalisés sur kératinocytes pour les tests de réparation cellulaire et sur fibroblastes pour l’étude de l'inhibition des métallo-protéinases ainsi que l'expression de l'actine-F, mesurée en fonction de l’âge puis après traitement avec l'A-A-A.
- RESULTATS : Les prédictions in silico ont permis de déterminer que l'A-A-A présente des capacités de réparation cellulaire et d'inhibition des métallo-protéinases (MMP). La validation de ces activités a été confirmée respectivement via un test in vitro de migration cellulaire sur les kératinocytes et les activités MMP sur les fibroblastes. L'augmentation significative de la mobilité cellulaire après stimulation avec l'A-A-A, nous a conduit à tester l'expression de l'actine –F tout d'abord en démontrant une augmentation de son expression en fonction de l′âge, induisant une rigidité mécanique du fibroblaste. Nous avons ensuite confirmé que la stimulation des fibroblastes âgés avec l'A-A-A durant 72 heures réduisait significativement l'expression de ce marqueur de rigidité. En l'occurrence, l'A-A-A réduit l'expression de l'actine-F sur des fibroblastes de 50 ans de 40% ce qui équivaut à une expression de l'actine-F des fibroblastes de 30 ans.
- CONCLUSION : En combinant une approche in silico et in vitro, nous avons confirmé le potentiel de l'A-A-A à améliorer des marqueurs cellulaires du vieillissement dans les compartiments épidermique et dermique via la régénération cellulaire sur kératinocytes, l'inhibition des métalloprotéinases et la réduction de l'expression de l'actine-F, comme marqueur de rigidité cellulaire liée à l'âge sur fibroblastes.Note de contenu : - METHODS : In silico study : SAR analysis using PASS program - Examination of MMP release using Luminex - Scratch mobility test (wound healing assay) using immortalized keratinocytes (HaCAT cells) - Immunofluorescence staining of polymerized actin (F-actin) in HDF

Especially for particulate matter and polycyclic aromatic hydrocarbons (PAHs) negative influence on skin integrity has been shown. The natural stress-protection molecule Ectoin® is well known to strengthen the skin barrier function, to work against variable stressors such as UVA/UVB radiation, IR-A, visible light or allergens, and for its anti-inflammatory activity. This study demonstrates the global protection and prevention efficacy of Ectoin from air pollution induced skin damage. Ectoin is well suited for topical antipollution products targeting skin pigmentation and accelerated ageing as well as particle stressed and irritated skin. Vitamine AIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Dermal active substances, in particular anti-aging ingredients, find frequent use in cosmetics for their beneficial effects on the skin. But they can only have these effects if they are bioavailable. That means that the substance must efficiently penetrate the skin in a meaningful and effective concentration. Furthermore, the substance must be protected from degradation due to external influences such as light or oxygen, both as a raw material and in formulation. Retinol is a highly effective antioxidant and has long been known as an anti-aging ingredient. However, its chemical instability makes it a challenge for formulators. Furthermore, in high concentrations retinol causes skin irritation and redness. Retinol can be stabilised to a substantial degree by embedding it in a semi-crystalline, unordered matrix of selected solid and liquid lipids in the form of submicron particles in an aqueous suspension. This also gives efficient, controlled penetration of retinol from the particles into the skip over a prolonged period, minimising skin irritation. At the same time, with their effective adhesion to the skin, the lipid particles help restore the damaged skin barder. This adhesive property also has the benefit of extending the release of retinol. GlabridineLa glabridine est un composé chimique qui se trouve dans l'extrait de racine de réglisse (Glycyrrhiza glabra). La glabridine est un isoflavane , un type d'isoflavonoïde. Ce produit fait partie d'une plus grande famille de molécules d'origine végétale, les phénols naturels.
Elle est utilisée comme ingrédient dans les cosmétiques et est répertoriée dans la Nomenclature internationale des ingrédients cosmétiques (INCI).
La glabridine est une poudre brun jaunâtre. Elle est insoluble dans l'eau, mais soluble dans les solvants organiques tels que le propylène glycol. (Wikipedia)

For this, formulators have numerous cosmetic active ingredients at their disposal. However, a powerful active ingredient can only achieve the desired effect on the skin if it has the relevant bioavailability. The active ingredient must, therefore, be able to penetrate the skin efficiently and build up a specific concentration of the active ingredient there.
Glabridin is a natural whitening agent extracted from the liquorice root (Glycyrrhiza glabra). It intervenes in the biosynthesis of the skin’s own pigment (melanin) and lightens the skin. Its efficacy has been confirmed in numerous publications. However, it is difficult to formulate this active ingredient as it has a very low solubility in lipids. If attempts to incorporate the active ingredient homogeneously into the facial care formulation do not succeed, it will not achieve the relevant bioavailability. This is not the desired outcome given the high price of the active ingredient (five figure amounts per kg). Efficient, controlled penetration of the skin by Glabridin can be achieved by embedding Glabridin in submicron particles made of a semi-crystalline, unordered matrix of selected liquid and solid lipids. Furthermore, the lipid particles help to repair a damaged skin barrier by adhering to the skin thanks to effective adhesion. This adhesion offers the added advantage of prolonged release.
