Tests de sécuritéIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - OBJECTIF : Cette étude visait à caractériser in vitro et in vivo une crème cosmétique avec 6 % d'extrait aqueux de bourgeon de pommier sauvage, contenant 3,5 % d'acides alpha‐hydroxylés (AHA) et de composés polyphénoliques, stabilisés par des émulsifiants d'alkylpolyglucosides biodégradables.
- METHODES : La caractérisation in vitro de la crème comportait une analyse organoleptique physico‐chimique (valeurs de pH et conductivité électrique), une estimation de l'activité antioxydante (AA; à l'aide du test DPPH) sur une période de conservation de 180 jours, à une température de 22 ± 2 °C, et la détermination des substances actives cosmétiques (AHA) contenues dans la crème (via une analyse par CLHP). L'estimation in vivo du potentiel d'irritation cutanée, après application de la crème sous pansement occlusif, pendant 24 h, a été étudiée auprès de 12 volontaires sains, via des méthodes biophysiques : efficacité de l'hydratation, perte en eau transépidermique (TEWL), pH cutané, érythème (EI) et indice de mélanine (MI) chez 10 volontaires sains, après d'application de la crème sur 28 jours, et efficacité du blanchiment chez 10 volontaires sains, après application de la crème sur 7 jours, suite à une hyperpigmentation cutanée induite artificiellement.
- RESULTATS : La crème cosmétique analysée était de couleur beige pâle et inodore, avait une consistance semi‐solide et un aspect homogène, avec des valeurs de pH de 6,53 ± 0,14, une conductivité électrique supérieure à 50 μS/cm−1 et une AA de 24,96 %RSC après préparation; ces caractéristiques sont restées stables durant toute la période d'analyse. Les mesures in vivo ont révélé une absence d'irritation cutanée, après application de la crème sous pansement occlusif, dans le cadre de son profil de sécurité. L'hydratation de la peau a augmenté de 17,28 ± 12,23, après application de la crème sur 14 jours, et de 21,19 ± 7,59, après application sur 28 jours. En outre, au cours de l'analyse effectuée, les valeurs de TEWL et de pH cutané sont restées stables durant l'application de la crème. L'application de la crème après une hyperpigmentation artificielle a entraîné une réduction du MI (la ΔMI après 7 jours était de ‐45,30 ± 18,55).
- CONCLUSION : La crème cosmétique formulée avec 6 % d'extrait standardisé de bourgeon de pommier sauvage et stabilisée par des émulsifiants d'alkylpolyglucosides biodégradables a démontré une bonne stabilité, un niveau acceptable d'activité antioxydante in vitro, une absence d'irritation cutanée après application de la crème sous pansement occlusif et des effets positifs sur la peau de l'homme suite à l'application (effets hydratants et blanchissants). En vertu des propriétés mentionnées, la crème est adaptée à une potentielle utilisation comme produit cosmétique, pour prévenir les lésions cutanées dues au stress oxydatif, pour hydrater les peaux sèches et blanchir la peau en cas d'hyperpigmentation.Note de contenu : - MATERIALS AND METHODS : Materials - Formulation and preparation of investigated samples - HPLC methods for analysis and standardization of AHAs in investigated extract and cream - In vitro methods for characterization and stability analysis of investigated creams - In vitro analysis of antioxydant activity of investigated creams - In vivo characterization of irritation potential and efficiency of investigated cream with extract of wild apple fruit - biophysical methods - Stastistical analysis

Toxicologie cellulaireIndex. décimale : 667.3 Teinture et impression des tissus Résumé : Fenton oxidative wastewater treatment of CI Reactive Black 5 and CI Reactive Blue 19 effluent was performed after a simulated laboratory-scale dyeing process, and the cytotoxicity of the treated effluent was evaluated using human skin cell lines. Among the components for Fenton oxidation, the human skin cell results showed that iron(II)sulfate at 150 mm did not show any significant cytotoxic effect, while other components, such as Glauber's salt solution (20 g l−1; 14%), CI Reactive Black 5 (30 mg l−1; 24%), caustic soda (5 g l−1; 30%), CI Reactive Blue 19 (30 mg l−1; 32%), hydrogen peroxide (0.01 m) and soda ash (5 g l−1) showed cytotoxic potential; the reagent sodium sulfite (30 mm; 48%) exhibited the strongest cytotoxicity level. Fast decolorisation (>95%) was achieved within 10 min for CI Reactive Black 5, while for CI Reactive Blue 19 it took longer (1.5 h) to achieve the same decolorisation. Studies showed that decolorisation for both dyes followed second-order kinetics. In spite of the remarkable efficacy of the Fenton oxidation process in removing colour within a short period of time, the resulting treated wastewater (within a reaction time of 1.5 h) also showed cytotoxicity towards the human HaCaT skin keratinocyte cell line. This observation can be explained by the strong oxidant and intermediate species produced during the advanced oxidation process, and a treatment step using sodium sulfite and a prolonged residence time can help to reduce the cytotoxicity. Latest alterations in the European legislation emphasise this fact. Whether a cosmetic product is well tolerated or causes irritations or allergic reactions must be proven by dermatological tests. The range of test methods starts with simple questionnaires and ends with complex physiological measurements. The quality of dermatological reports directly depends on the seriousness of the commissioned dermatologists. Pitfalls occur whenever non qualified scientific results are generously used for advertising campaigns like 'dermatologically tested', tested', 'hypo-allergen' etc. Additionally a lot of reports are scientifically insufficient. Dermatological reports on cosmetics thérefore must be valid in scientific methods and practical execution.
