kératineLa kératine est une protéine, synthétisée et utilisée par de nombreux êtres vivants comme élément de structure, et également l'exemple-type de protéine fibreuse.
La kératine est insoluble, et peut être retrouvée sur l'épiderme de certains animaux, notamment les mammifères, ce qui leur garantit une peau imperméable. Parfois, lors d'une friction trop importante, la kératine se développe à la surface de la peau formant une callosité. Les cellules qui produisent la kératine meurent et sont remplacées continuellement. Les morceaux de kératine qui restent emprisonnés dans les cheveux sont couramment appelés des pellicules.
La molécule de kératine est hélicoïdale et fibreuse, elle s'enroule autour d'autres molécules de kératine pour former des filaments intermédiaires. Ces protéines contiennent un haut taux d'acides aminés à base de soufre, principalement la cystéine, qui forment un pont disulfure entre les molécules, conférant sa rigidité à l'ensemble. La chevelure humaine est constituée à 14 % de cystéine.
Il y a deux principales formes de kératines : l'alpha-kératine, ou α-keratin, présente chez les mammifères notamment, dont l'humain, et la bêta-kératine, ou β-keratin, que l'on retrouve chez les reptiles et les oiseaux. Ces deux types de kératines ne présentent clairement pas d'homologie de séquence.
Chez l'être humain, la kératine est fabriquée par les kératinocytes, cellules se trouvant dans la couche profonde de l'épiderme. Les kératinocytes absorbent la mélanine (pigment fabriqué par les mélanocytes), se colorent et ainsi cette pigmentation de l'épiderme permet de protéger les kératinocytes des rayons ultraviolets du Soleil.

Tests de toxicitéIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Un peptide, fragment de la protéine cuticulaire kératine type II, le “keratine peptide” (KP), a été étudié comme possible agent de renforcement pour des cheveux défrisés affaiblis. Le peptide a été préparé en solution aqueuse (WF) et dans des formulations aux solvants organiques (OF), afin de déterminer l’effet des solvants organiques sur l’interaction du peptide avec les cheveux et les différences sur la restitution du cheveu. Les deux formulations du peptide ont montré une amélioration des propriétés mécaniques et thermiques des cheveux affaiblis; les formulations aux solvants organiques se montrant plus efficaces. La cytotoxicité et la génotoxicité du peptide ont également été évaluées dans les fibroblastes de la peau, par des méthodes différentes (dosage au bleu d’Alamar, sonde au 2′-7′- dichlorofluorescéine, morphologie et prolifération des cellules, évaluation des dommages à l’ADN par une version alcaline du test COMET). Ces tests sont des indicateurs du potentiel irritant du peptide ou d’être cancérigène, respectivement. Le peptide dans WF n’a pas provoqué de cytotoxicité ou de génotoxicité, à aucune des concentrations testées. La présence de OF, cependant, a induit une diminution de 20% de la viabilité cellulaire dans l’ensemble de la gamme des concentrations utilisées après incubation 72h. En outre, une inhibition de la croissance cellulaire a été observée et la formulation a été considérée comme génotoxique lors du premier contact avec des cellules. Le peptide a donc été considéré comme un agent prometteur de renforcement pour les cheveux et s’est avéré inoffensif lorsqu’il est appliqué dans WF. Tests de toxicitéIndex. décimale : 668.3 Adhésifs et produits semblables Résumé : Waterborne reactive polymers that contain pendant 2-oxazoline groups were prepared by radical copolymerization of 2-alkenyl-2-oxazoline and unsaturated co-monomers. By varying the composition of co-monomers, a series of emulsion polymers with the various glass transition temperatures (Tg) or water-soluble polymers with high oxazoline contents were prepared. The crosslinking reactions of these oxazolidine-functional polymers with waterborne carboxylated polymers were examined.
As a result, the oxazoline-functional polymers show sufficient reactivity and stability to be used as a low-toxic, one-packed crosslinker providing improved properties for waterborne carboxylated polymer. The pH value of the formulation has an important effect on its reaction rate and pot life. It was also found that the mechanical property of cured film can be controlled by selecting the composition of backbone polymer in the oxazoline-functional polymer. PolypeptidesUn polypeptide est une chaîne d'acides aminés reliés par des liaisons peptidiques. On parle de polypeptide lorsque la chaîne contient entre 10 et 100 acides aminés. Au-dessus de 100 acides aminés on parle généralement de protéine.
Parmi eux, les peptides multi-cycliques, les peptides phosphorylés ou encore ceux intégrant des liaisons non-peptidiques ou des peptides conjugués.
Certains polypeptides constituent une famille d'antibiotiques dont les molécules très toxiques n'en permettent qu'un usage très limité. D'autres s’imposent aujourd’hui comme des éléments essentiels à la mise au point de vaccins ou de principes actifs destinés aux traitements de certaines pathologies telles que les cancers, les déficiences hormonales, l’ostéoporose.

Tests de toxicitéIndex. décimale : 547 Chimie organique : classer la biochimie à 574.192 Résumé : Supramolecular peptides exhibit obvious similarities with collagen fibers in terms of self-assembly characteristics, nanofibrous structure, and responsiveness to external stimuli. Here, a series of supramolecular peptides were developed by altering the amino acid sequence, enabling the self-assembly of three types of 4-biphenylacetic acid (BPAA)-tripeptides into fibrous hydrogel through hydrogen bonding and π–π stacking under the influence of ion induction. Transmission electron and scanning electron microscopies revealed that the diameter of the fiber within nanofibrous hydrogels was ~ 10 and ~ 40 nm, respectively, which was similar with the self-assembled collagen fibers. For this reason, these hydrogels could be considered as a biomimetic extracellular substitute. Meanwhile, the gelation concentration induced by ions was even lower than 0.66 wt%, with an elastic modulus of ~ 0.27 kPa, corresponding to a water content of 99.34 wt%. In addition, the three supramolecular hydrogels were found to be good substrates for L929 cell adhesion and MC-3T3 cell proliferation. The overall results implied that BPAA-based hydrogels have a lucrative application potential as cell carriers. Toxicologie cellulaireIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Quantitative and qualitative determination of skin irritation and skin protection through market formulations using an in vitro skin model, the perfused bovine udder (BUS) and different analytical methods, was the objective of the present research. Various noxae that are percutaneously absorbed cause skin irritation and damage through different mechanisms. Cell vitality, signs of inflammation, and the presence of inducible free radicals among the analytical parameters that enable quantification of skin damage. In this study cytotoxic effects were determined using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, and tissue levels of the pre-inflammatory mediator prostaglandin E2 in treated/untreated whole skin biopsies were assessed using an enzyme immunoassay. The inducibility of free radicals was determined by electron resonance spectroscopy techniques. Noxae (standardized chemicals and products) like detergents, coolants, toluene, and n-octane caused reduced cell vitality and metabolic activity, inflammation as evidenced by the presence of prostaglandins, and enhanced values in the inducible free radical status in in vitro bovine udder skin. The effect of the noxae is dose-dependent and permeation time-dependent, allowing construction of dose-effect curves. The induced cytotoxicity and free radical status show a positive correlation, while the inflammatory actions analyzed by the prostaglandin E2 tissue concentration do not.
As a consequence, the skin compatibility of noxae and performance of skin protection formulations can be evaluated by the measurement of different cell physiological and biophysical parameters at the very early stages of skin damage.Note de contenu : - Chemicals
