Toxicologie cellulaireIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Sur 107 produits cosmétiques dont l'indice d'irritation oculaire maximum (IOMA) a été établi par le test de Draize, des études ont été effectuées en parallèle par une méthode de cytotoxicité en culture (CFIO) et par le test sur membrane chorio-allantoïdienne d'oeuf de poule (HET-CAM). Les résultats montrent une corrélation satisfaisante entre les deux méthodes alternatives et le test de Draize sur l'oeil du lapin. Des résultats non concordants ont été obtenus dans 12 cas sur 107 par la méthode HET-CAM (six faussement positifs, deux faussement négatifs, quatre impossibilités d'effectuer le test) et dans 14 cas sur 107 par la méthode CFIO (dix faussement positifs et quatre faussement négatifs). Cependant, les produits fortement irritants ont toujours été détectés, les faux négatifs ne concernant que ceux dont l'indice d'irritation oculaire était proche de la limite (IOMA = 15) entre les produits faiblement et moyennement irritants. De plus, aucun des produits étudiés ne s'est révélé faussement négatif en même temps par les deux méthodes alternatives. Le risque de sous-estimer le potentiel irritant oculaire des produits a ainsi été évité. L'association de ces deux méthodes semble donc intéressante pour l'étude du potentiel d'irritation oculaire des produits cosmétiques. The concentrated extract was partitioned with EtOAc, n-BuOH, and H2O. From the EtOAc fractions, three flavonoid compounds were isolated through the repeated SiO2 and ODS column chromatographies. The isolated compounds were identified new compound on the basis of NMR, MS, and IR spectroscopic data, respectively. In order to investigate antiatopic dermatitis potential of Nymphaea alba, we evaluate the anti-inflammatory effect of Compound 1-3 from the flowers of N. alba. Compound 1-3 dose-dependently showed inhibition effects for the productions of nitric oxide (NO) in lipopolysaccharide (LPS)-induced macrophages (Compound 1: IC50 µM ; Compound 2: IC50 pM ; Compound 3: IC50 µM). And we conducted the mRNA expression related with thymus- and activation-regulated chemokine (TARC/CCL17) and macrophage-derived chemokine (MDC/CCL22) in HaCaT keratinocyte cells by Compound 1. Compound 1 inhibited the expression of nnRNA on HaCaT cells which treated with TARC/CCL17 and MDC/CCL22.

TyrosinaseIndex. décimale : 668.5 Parfums et cosmétiques Résumé : There has been a tenacious search for pharmaceuticals of natural origin, as they are cost-effective and are noted for having little or no side effects. The rate at which diseases are developing resistance to synthetic drugs is quite alarming, and the side effects of these drugs remain an excruciating agony to the pharmaceutical industry. Gold nanoparticles (AuNPs) have wide applications in current technology. However, their use in medicine has not been adequately explored. Chemical methods for the synthesis are associated with environmental benignity and tissue toxicity on in vivo administration. For the first time, we have synthesized AuNPs from leaf extracts of Teraxacum officinale that were found to have significant anti-melanoma, tyrosinase inhibitory and anti-microbial effects, and hence stand as promising candidates for use in cosmetics medical and food industries. QuinolonesLes quinolones et fluoroquinolones1,2,3 forment une large classe d'antibactériens de synthèse qui comprennent les dérivés de l'acide nalidixique découvert en 1962 et utilisé chez l'homme dès l'année suivante. Cette famille d'antibactériens a fait l'objet de recherches très importantes aboutissant au dépôt de plus de 10 000 brevets. L'ajout de l'atome de fluor dans les années 1970 a permis d'augmenter fortement la pénétration des molécules quinolones dans les cellules (jusqu'à 200 fois plus) : ce fut la naissance des fluoroquinolones, puissants antibiotiques capables de lutter contre une grande variété de germes chez l'homme et l'animal.
Toxicologie cellulaireIndex. décimale : 667.2 Colorants et pigments Résumé : The sensing properties of dyes based on 2(1H) quinolones containing dicyanoethylene groups towards selected anions such as CN−, F−, I−, Cl−, SCN−, CH3COO−, NO−2and CIO−4were evaluated. The spectroscopic responses of these compounds were investigated in a solution of acetonitrile/water (1:9, v/v) and phosphate buffer at a pH of 7.4. These dyes were found to act as efficient colorimetric and high turn–off fluorescent sensors to cyanide anions. The fluorescence intensity was linearly related to the concentration of CN− from 1 to 10 µmol/L, and the detection limits reached a micromolar level of 6.7, 11.1, 3.8 and 7.2 µmol/L for 1-4, respectively. The cytotoxic effect of the dyes against human neuroblastoma cells (SH-SY5Y) was also determined by MTT assay. The examined compounds exhibited slight cytotoxicity against cells under experimental conditions.
