StatistiquesIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : Nowadays, it is recognized the need for improved safety and efficacy protocols to evaluate the human stratum corneum (SC) and its interaction with topical and cosmetic formulations by minimally or non-invasive methodologies. The aim of our research work was to streamline the HPLC-TBARS-EVSC (high-performance liquid chromatography–thiobarbituric acid reactive substances–ex vivo stratum corneum) methodology, by exploring the results of a group of 18 subjects.
- Methods : The study included nine women and nine men aged between 19 and 57 years old with phototypes from II to V. Sites in the forearm of each volunteer were randomly delimited, and the SC was collected by tape stripping. HPLC was used to quantify the MDA-TBA2 (malondialdehyde–thiobarbituric acid) adduct from the tape-stripped SC, irradiated and not by an ultraviolet (UV) simulator chamber.
- Results : Observing the findings of our present investigation, and the statistical approach applied, the use of the ratio between the treatment site and control would be an adequate strategy to better discriminate and evaluate the results. Additionally, an optimal selection of the volunteers to respond specifically to the purpose of the ex vivo assay also can be considered advantageous.
- Conclusions : It seemed that in future studies focusing on the impact of SC UV-induced lipid peroxidation, determined by the HPLC-TBARS-EVSC, the most suitable subjects are females aged less than 35 years old, with phototype II.Note de contenu : - Legal and ethical concerns, and subjects' selection

Over time, normal skin ageing is accelerated by external stresses such as smoking, pollution, chemical products and radiation. UV light, in particular UVA, causes DNA damage, apoptosis and morphological modifications, which are responsible for both premature ageing and cancer. The aim of this study was to establish a discriminatory and sufficiently reproducible cutaneous model for evaluating UVA damage, to enable testing for effectiveness of potentially protective compounds.
- Methods : The cutaneous model is based on Human skin explants irradiated with UVA. Deleterious effects on epidermis were observed and quantified by haematoxylin–eosin staining and by immunofluorescence of y-H2Ax, cytokeratin 5, involucrin and loricrin protein. Dermis deterioration was evaluated by transmission electronic microscopy and zymography in situ.
- Results : We were able to observe and quantify deleterious effects associated with UVA irradiation: epidermal and dermal disruption, appearance of Sunburn cells, increased DNA damage and induced apoptosis. The use of this model in the evaluation of protective compounds was first confirmed using sunscreens, then further validated with a panel of active ingredients which showed beneficial effects on epidermis morphology and DNA integrity after UVA exposure.

VectorisationIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Objectif : Les compartiments de la peau traditionnellement ciblés par les actifs cosmétiques – épiderme et le derme – sont ancrés et nourris par l'hypoderme sous-jacente, qui devrait donc être un objectif clé pour les formulations de rajeunissement de la peau. Toutefois, étant donné la difficulté d'atteindre même les couches superficielles cosmétique de la peau, et dû à sa composition grasse "ingrate", le potentiel cosmétique de régénération de l'hypoderme reste largement inexploité. Par conséquent, cette étude était destinée à évaluer la capacité d'un produit cosmétique à déclencher une réponse régénérative dans le derme et l'épiderme grâce à une action sélective sur hypoderme. En outre, il vise à établir l'effet d'un tel matériau cosmétique sous forme de livraison hypodermique transbuccale, sur les cellules précurseurs hypodermiques – les pré-adipocytes.
Méthodes : Une combinaison de l'extrait de pépins de raisin et des phospholipides de soja a été élaborée et normalisée pour le niveau de l'activité d'inhibition de l'élastase et des radicaux libres. Cette formulation a été ensuite utilisée pour communiquer avec la couche hypodermique de biopsies de peau humaine et – sous une forme de véhicule de livraison transbuccale – avec les pré-adipocytes 3T3-L1 ; ses effets ont été quantifiés en utilisant la PCR et l'histochimie.
Résultats : L'application du matériau de raisin/soja standardisé à la couche hypodermique de la peau déclenche la modulation de l'expression génétique dans les couches supérieures de la peau et aboutit à l'amélioration notable morphologique au niveau du derme et de l'épiderme. En outre, lorsque ce matériau a été formulé dans un film un muco-adhésif, intra-oral et appliqué sur des pré-adipocytes 3T3-L1, la modulation de l'expression des gènes qui en résulte dans ces cellules est compatible avec les effets de la différenciation et de désintoxication.
Conclusions : Ces résultats suggèrent que les formulations transbuccales de produits cosmétiques de qualité nutraceutique ont le potentiel d'induire des voies de transduction du signal dans l'hypoderme du visage, entraînant des effets anti-vieillissement dans tous les compartiments de la peau, y compris les couches dermiques et épidermiques.Note de contenu : - MATERIALS AND METHODS : Preparation of SBD, 5HC, 5HCF - Antioxidant and anti-elastolytic activity - Assay of SBD, 5HC on human skin - Assay of SBD, 5HCF on 3T3-L1 pre-adipocytes

VectorisationIndex. décimale : 668.5 Parfums et cosmétiques Résumé : OBJECTIFS : Les polyphénols sont des molécules ayant un fort pouvoir antioxydant permettant de prévenir les dommages dus au photovieillissement de la peau mais leur utilisation est limitée de par leur faible solubilité et leur toxicité envers les cellules de la peau. Nous avons postulé que la glucosylation enzymatique pourrait améliorer leurs solubilité, stabilité et, en conséquence, leur efficacité. Le but de ce travail est d'étudier les changements induits par l'addition d'un glucose sur deux polyphénols ayant une structure chimique très différente [acide caféique (CA), epigallocatechin-3-gallate (EGCG) et leurs formes glucosylées, Glc-CA et Glc-EGCG] en évaluant leurs propriétés cytotoxiques et leurs activités antioxydante et anti-inflammatoire.
METHODES : Leur pouvoir antioxydant a d'abord été évalué par la méthode classique de réduction du radical DPPH. Puis, un ensemble de cellules de peau humaine (kératinocytes, mélanocytes, fibroblastes et cellules endothéliales) a été utilisé pour évaluer leur effet sur la toxicité cellulaire et leurs activités anti-oxydantes. Pour cela, un modèle de photovieillissement basé sur l'irradiation UV de cellules de peau a été établi. L'activité des molécules a été évaluée sur la production d'espèces réactives de l'oxygène (ROS), sur les activités superoxyde dismutase (SOD) et catalase et, enfin, sur la production de facteurs anti-inflammatoires IL-6, IL-8 et IL1β.
RESULTATS : Dans un modèle acellulaire, l'activité antioxydante mesurée par la méthode au DPPH était fortement réduite pour le Glc-CA comparé au CA, alors qu'elle restait la même pour le Glc-EGCG comparé à l'EGCG. Les dérivés glucosylés ne présentaient pas plus d'effet toxique sur les différentes cellules de peau. De plus, la toxicité était même fortement réduite pour l'acide caféique suite à sa glucosylation. L'efficacité des composés glucosylés contre la production de ROS induite par les UV était préservée, à la fois lors des traitements pré et post-UV. En particulier, une meilleure efficacité antioxydante a été démontrée avec le Glc-EGCG, vs. EGCG, sur les kératinocytes. Une augmentation des activités SOD et catalase a été nettement observée pour le Glc-CA. Les deux polyphenols glucosylés montrent la même activité que leur molécule parente en diminuant la production de facteurs anti-inflammatoires.
CONCLUSIONS : Nos résultats démontrent que la glucosylation enzymatique de CA et EGCG conduit à la préservation de leur activité antioxydante dans un modèle cellulaire de vieillissement de la peau par les UV, en dépit de la diminution de l'activité antioxydante instantanée observe pour Glc-CA. Glc-EGCC est spécifiquement plus actif que l'EGCG sur les keratinocytes, suggérant un ciblage spécifique. Gentiane jaune et constituantsLa gentiane jaune (Gentiana lutea), encore appelée grande gentiane, est une espèce de plantes de la famille des Gentianaceae, originaire d'Europe méridionale et d'Asie mineure, présente notamment dans divers massifs montagneux européens, dont les Alpes, le Massif central, le Jura, les Pyrénées et les Vosges. Elle a toutefois été observée en 2013 dans les hautes Ardennes belges1. La gentiane jaune est aussi appelée gentiane officinale, jouvansanne, quinquina d'Europe, quinquina des pauvres, lève-toi-et-marche, jansonna, bananier des Alpes et quinquina indigène. On considère que cette plante fait partie de la flore obsidionale de France.
- Description :
Grande herbe robuste et vivace, elle peut vivre 50 ans et met 10 ans pour fleurir la première fois. Les feuilles sont opposées et nervurées. Elles sont pétiolées à la base et sessiles embrassantes sur la tige. Les grandes fleurs jaunes sont serrées à la base des feuilles supérieures. La corolle est divisée en 5 à 9 lobes presque jusqu'à la base.
Elle est parfois confondue avec le vérâtre blanc (ou hellébore blanc), qui est violemment toxique, mais dont les fleurs sont blanches et les feuilles alternées, alors que les fleurs de la gentiane sont jaunes et les feuilles opposées.
- Usages :
Le rhizome et les racines sont utilisés en phytothérapie comme le laissent supposer ses nombreux noms vernaculaires. Mais la gentiane est surtout utilisée dans des apéritifs comme la liqueur de gentiane (Suze, Salers, Avèze, etc.) ou l'alcool de gentiane, Bière de Fleurac7, et le Picon, auxquels elle apporte son amertume. Ce sont près de 1 000 à 1 500 tonnes qui sont utilisées chaque année pour satisfaire les besoins de l'artisanat et l'industrie. En volume, la gentiane est une des trois premières plantes médicinales et aromatiques utilisées en France et ses applications sont nombreuses (pharmacie et médecine humaine et vétérinaire, boissons et spiritueux, cosmétique, fabrication d'arômes et d'extraits, gastronomie…). They can be treated by various methods, particular by topical solutions. This investigation was directed towards exploring the effect of gentiopicroside (GP) on the skin around the eyes. For this, an extract of Gentiana lutea (GIE) containing GP (65% by dry matter) was evaluated on oxidant and angiogenesis parameters using in vitro and ex-vivo studies. A clinical experimentation was also realized.
- Methods : The effect of GIE at different concentrations on antioxidant gene was evaluated in vitro by RT-qPCR after treatment of NHDF. The effect of 2.93 μg mL−1 GIE on the release of VEGF-A and VEGF-C by NHDF was also studied. The effect of 87.9 μg mL−1 GIE was also evaluated on pseudotube formation in a coculture system of normal dermal microvascular endothelial cells (HMVEC-d)-NHDF stimulated or not with VEGF as pro-angiogenic factor. Prior to these assays, preliminary cytotoxicity assays were performed using a standard WST-8 reduction assay. The expressions of carboxymethyl-lysine and glyoxalase-1 were quantified on skin explants topically treated with 147 μg mL−1 GIE in basal and UVA-irradiated conditions. A clinical study was conducted in 22 subjects using topical twice daily for 14 days on eye area (split-face application: cream containing 147 μg mL−1 GIE versus placebo). 3D image acquisition and skin colour measurement were performed at D0 and D14.
- Results : Treatment of GIE upregulated the gene expression of NFE2L2 and downregulated the expression of CXCL8. GIE targeted AGEs pathways and reduced the formation of pseudotubes. A total of 147 μg mL−1 GIE gel cream significantly reduced significantly the average roughness and relief of the upper eyelid skin as well as the redness of dark circles after 14 days of application.
- Conclusion : By acting on the pathway of AGEs, VEGF-A and VEFG-C, GIE seems to allow a rejuvenation of the skin resulting, among others, in a decrease in redness.
