Malassezia furfur is a lipid-dependent yeast naturally living on the skin. Despite its tolerance by the immune system under healthy condition, M. furfur overgrowth is associated with skin disorders such as dandruff.
Dandruff and its more severe form seborrheic dermatitis (SD), are common chronic inflammatory conditions characterized by an abnormal shedding of the skin in seborrheic areas of the body. While dandruff is restricted to the scalp, SD also affects oily rich areas of the face, causing itching, pruritic lesions and erythema.
Environmental and internal stress factors, dysregulation of the immune system and fungal colonization are amongst the main contributors to the development of SD and dandruff.1 It has been described that in dandruff, the quantity of Malassezia can increase up to 1.5 to 2 times its normal level.
This huge spread of yeast highly invades the epidermis, with detrimental consequences on the skin barrier function. Because the epidermis is the principal barrier against the penetration of chemicals and pathogens, any disruption increases microbial invasion and triggers both innate and acquired immune response.
Keratinocytes are playing a central role in this response by secreting key signaling molecules like antimicrobial peptides to overcome pathogenic infections, as well as various cytokines and interleukins to recruit inflammatory cells. In the case of Malassezia overgrowth, previous studies have reported the expression of some defensins and pro-inflammatory cytokines in response to infection.Note de contenu : - Colonization of RHE with M. furfur and barrier disruption

Cette enzyme est formée de quatre chaînes polypeptidiques d’environ 500 acides aminés, comportant chacune un groupe hème. Ces hèmes et leur environnement protéique sont les sites actifs de cette enzyme. Pour catalyser la réaction, l’atome de fer du groupement hème de la catalase réalise une coupure hétérolytique de la liaison O-O du peroxyde d'hydrogène, créant de ce fait une molécule d’eau et un groupement Fe(IV)=O hautement oxydant ; ce dernier peut ensuite oxyder une autre molécule de peroxyde d'hydrogène pour donner du dioxygène. Ce processus est illustré plus spécifiquement par les équations suivantes :
H2O2 + Fe(III)-E → H2O +O=Fe(IV)-E
H2O2 + O=Fe(IV)-E → O2 + Fe(III)-E +H2O
Avec une vitesse maximale de 200 000 réactions catalytiques par seconde, elle est une des enzymes les plus efficaces connues. Sa vitesse de réaction ne semble en effet limitée que par la diffusion, c’est-à -dire par la vitesse limite avec laquelle les molécules parviennent au site actif de l'enzyme. Chez les eucaryotes on la trouve dans le peroxysome des cellules pour la catalase A et dans le cytosol pour la catalase T.
On trouve la catalase chez tous les organismes aérobies et plus particulièrement dans les navets ou dans la plupart des racines des plantes, dans la levure et dans le foie des animaux.

Stress oxydatifIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Un signe évident du vieillissement est le grisonnement des cheveux ou la perte de la production de pigment et de son dépôt dans les follicules pileux. De nombreux mécanismes, agissant à différents niveaux et endroits folliculaires, contribuent aux cheveux grisonnants, allant des dommages aux cellules souches mélanocytaires à la mort des mélanocytes folliculaires. Une question clé qui est commune à ces processus concerne le dommage oxydatif. Au niveau de la niche des cellules souches du follicule, le stress oxydatif, accéléré par l'appauvrissement de BCL-2, conduit à l'apoptose et à une diminution sélective des cellules souches des mélanocytes, ce qui réduit le repeuplement des follicules anagènes nouvellement formés. Les mélanocytes bulbaires mélaniques expriment des niveaux élevés de Bcl-2 pour permettre la survie suite à l'attaque par les espèces réactives de l'oxygène (ROS) induites par la mélanogénèse et les ultraviolets A (UVA). Avec le vieillissement, l'expression dans mélanocytes du bulbe des protéines anti-oxydantes comme BCL-2, et peut-être TRP-2, est réduite, et la défense spécifique du système anti-oxydant enzymatique faiblit à travers le follicule, résultant en un plus grand stress oxydatif. Une réduction marquée de l'expression catalase et les résultats de l'activité de l'accumulation millimolaire de peroxyde d'hydrogène contribuent à un dysfonctionnement et à la mort des mélanocytes bulbaires. Fait intéressant, les mélanocytes amélanotiques à la gaine externe (SRO) sont un peu moins touchés par ces processus, et survivent pendant plus longtemps encore dans les follicules pileux blanc vieillissants. Une meilleure compréhension de la sensibilité des melanocytes au stress oxydatif dans les différents sites folliculaires pourrait donner des indices pour un éventuel traitement pour la prévention, voire l'inversion des cheveux gris. Vieillissement cutanéIndex. décimale : 668.5 Parfums et cosmétiques Résumé : La protection contre la perte en eau et la prévention de pénétration des substances et des bactéries dans le corps sont considérées comme les fonctions les plus importantes de la peau. Cette fonction de la peau que l'on appelle 'barrière' est la frontière naturelle entre l'organisme et l'environnement intérieur, et elle est principalement formée par l'épiderme. Une altération de la fonction barrière de la peau est souvent constatée dans la peau malade et abîmée. L'influence du vieillissement sur ​​la fonction barrière de la peau est largement acceptée, mais n'a pas été évaluée de manière concluante. Par conséquent, l'objectif de la présente étude clinique était d'évaluer l'influence potentielle du vieillissement sur ​​la fonction barrière de la peau,y compris la perte insensible en eau, l'hydratation du stratum corneum, le taux de sébum et la valeur du pH. 150 femmes en bonne santé âgées de 18 à 80 ans, réparties en cinq groupes d'âge de 30 sujets chacun, ont été évaluées dans cette étude. La perte insensible en eau, le niveau d'hydratation, la sécrétion de sébum et le pH du film hydro-lipidique acide ont été mesurés à l'aide des méthodes biophysiques mondialement reconnues, par des mesures sur la joue, le cou, le décolleté, l'avant bras et le dos de la main. Alors que la PIE et l'hydratation du stratum corneum aient montré seulement une corrélation très faible avec l'âge des sujets, la production de sébum diminue significativement avec l'âge, entraînant le niveau le plus bas de lipides de surface mesurés chez les sujets âgés de 70 ans. Le pH le plus élevé de surface était mesuré chez les sujets âgés de 50 à 60 ans, alors que le groupe d'âge le plus âgé montrait le plus faible pH moyen. Le dos de la main est l'endroit à la PIE la plus haute et de l'hydratation du stratum corneum la plus basse, dans tous les groupes d'âge. Les résultats montrent que seuls certains paramètres liés à la fonction barrière de la peau sont influencés par le vieillissement. Alors que la production de sébum diminue de manière significative dans la vie le et pH de la surface cutanée est significativement augmentée chez la femme ménopausée, la PIE et l'hydratation de la couche cornée ne montrent que des variations mineures avec l'âge.
