TyrosinaseIndex. décimale : 668.5 Parfums et cosmétiques Résumé : On trouve un bon nombre de produits proposés pour éclaircir la peau, mais peu parmi eux s'avèrent acceptables, en particulier à cause du potentiel carcinogène attribué aux ingrédients classiques tels que l'hydroquinone, l'arbutine et l'acide kojique. Pour trouver une alternative plus sure, nous avons développé deux composés d'origine végétale issus de la biodiversité Brésilienne, à savoir un extrait de Schinus terebinthifolius Raddi et une fraction d'acide linoléique isolée de l'huile de Passiflora edulis. L'effet éclaircissant de ces composes a été évalué à l'aide de tests biochimiques et de modèles in vitro, y compris de cellules et de peaux équivalentes. Les résultats montrent que l'extrait de Schinus terebinthifolius Raddi est capable de réduire l'activité de la tyrosinase in vitro, et que la combinaison de cet extrait avec l'acide linoléique permet de diminuer la quantité de mélanine produite par les cellules B16 cultivées en présence de l'hormone de stimulation des mélanocytes (MSH). La quantité de mélanine produite diminue également dans la peau humaine reconstituée contenant des mélanocytes quand elle est traitée avec ces composés. L'utilisation conjointe de ces composés pourrait apporter un effet blanchissant synergique comparé à l'utilisation isolé de chacun. Nous montrons que l'activité des ces composés est comparable à celles des substances utilisées classiquement, tel que l'acide kojique. Ce nouveau mélange pourrait représenter une alternative thérapeutique pour traiter l'hyperpigmentation et s'avérer être un composé efficace dans les produits cosmétiques éclaircissants. KératinocytesLes kératinocytes sont des cellules constituant 90 % de la couche superficielle de la peau (épiderme) et des phanères (ongles, cheveux, poils, plumes, écailles). Ils synthétisent la kératine (kératinisation), une protéine fibreuse et insoluble dans l'eau, qui assure à la peau sa propriété d'imperméabilité et de protection extérieure.
L'épiderme est divisé en 4 couches basées sur la morphologie des kératinocytes (de l'intérieur vers l'extérieur) :
1. stratum germinativum (couche basale à la jonction avec le derme)
2. stratum spinosum
3. stratum granulosum
4. stratum lucidum
5. stratum corneum
Les kératinocytes passent progressivement de la couche basale vers les couches supérieures par différenciation cellulaire jusqu'au stratum corneum ou ils forment une couche de cellules mortes nommées squames, par apoptose. Cette couche constitue une barrière de protection et réduit la perte d'eau de l'organisme.
Les kératinocytes sont en perpétuel renouvellement. Ils mettent environ 1 mois pour aller de la couche basale au stratum corneum mais ce processus peut être accéléré en cas d'hyperprolifération de kératinocyte (psoriasis).

StatistiqueIndex. décimale : 668.5 Parfums et cosmétiques Résumé : The human skin, and mainly the upper layer of the epidermis, plays the role of a barrier, but is also one of the first and major targets of air pollutants, pollutants contributing to wrinkle and dark sports occurrence through the redox imbalance. A possible approach to attack ROS-mediated disorders for both preventive and treatment means is based on the use of substances, which can be found in plants as secondary metabolites, lignans being a promise candidate. The present study was aimed to better understand the cellular mechanisms beyond the oxidative changes induced by urban pollution (Urban dust 1649b, NIST) and the effect of Schisandra chinensis (S. chinensis) extract in reconstructed human epidermis, by a transcriptomic approach and secondly through the evaluation of Nrf2, AhR, NF-kB, and DJ-1 pathways using an in vitro model. Finally, we evaluated the effect of S. chinensis on skin hydration, homogeneity, radiance and luminosity in Chengdu (China). Urban dust (80pg.ml") was able to activate the cytoplasmic expression of NF-kB and AhR when compared to control. S.chinensis extract attenuated the urban dust-induced oxidative stress, the protective mechanism being associated, at least in part, with the modulation of the Nrf2 and AhR pathways and the activation of DJ-1. S. chinensis extract, named Urbalys protects from prolonged pollution aggression since it improves hydration, protects skin homogeneity, increases skin radiance and attenuates skin spot intensity after 21 days of pollution exposition. Vaccinium bracteatum ThunbergIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Screening for tyrosinase (TYR) inhibitors potentially useful for control of skin pigmentation has been hampered by the limited availability of human TYR. To overcome this hurdle, we have established human embryonic kidney (HEK293)-TYR cells that constitutively express human TYR. In the current study, we assayed human TYR inhibition activities of 50 plant extracts using the lysates of transformed HEK293-TYR cells. The strongest inhibition of human TYR was shown by the extract of Vaccinium bracteatum Thunberg, followed by the extract of Morus bombycis Koidzumi. The former extract did not inhibit mushroom TYR activity whereas significant inhibition was observed with the latter extract, demonstrating the importance of using human TYR in the screening for human TYR inhibitors. Upon liquid-liquid partitioning of the extract from V. bracteatum, the active constituents were enriched in the ethyl acetate fraction, and the subsequent preparatory thin-layer chromatography identified p-coumaric acid (PCA) as the main active constituent. The hypo-pigmentation of PCA was verified in the MelanoDermâ„¢ Skin Model. This study demonstrates that transformed HEK293-TYR cells could expedite the discovery of human TYR-specific inhibitors from natural sources which might be useful in the control of skin pigmentation.
