Cours de matières premières - Tome 2URLCovalent fixation of drum dyes on collagen / Gerhard Wolf in JOURNAL OF THE AMERICAN LEATHER CHEMISTS ASSOCIATION (JALCA), Vol. CII, N° 3 (03/2007)

Titre : Covalent fixation of drum dyes on collagen Type de document : texte imprimé Auteurs : Gerhard Wolf, Auteur ; Harald Kiesow, Auteur ; Laszlo Somogyi, Auteur Année de publication : 2007 Article en page(s) : p. 93-98 Note générale : Bibliogr. Langues : Américain (ame) Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : Covalent fixation of performance chemicals on various substrates of natural or synthetic origin is a well-known and thoroughly investigated principle since decades. Taking a deeper look into the literature of collagen, though, there are only a few examples found to this topic. Immobilisation of process chemicals is of great importance for leather as well. Especially dyes play a crucial role, since their mobility within the substrate matrix my lead to visible staining of other materials upon contact. In order to meet these challenges in leather manufacturing a new dyeing concept has been developed: covalent fixation of drum dyes to the collagen fibre. The dyes react with collagen under mild alkaline conditions at moderately elevated temperatures, thus reaching a fixation rate of above 95%. Due to the fact that only covalently fixed dye molecules remain in leather, unprecedented fastness properties are achieved even on leather dyed to very deep shades. TempératureIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : An organophosphorous hydrolase (OPH) was prepared and partially purified from Flavobacterium sp. The crude enzyme, with an activity of 1030U/g, was immobilized onto insoluble bovine collagen fibers (hide powder), instead of collagen membrane, through glutaraldehyde coupling. Optimal conditions of enzyme immobilization and properties of the immobilized enzyme preparation were investigated. Compared to glutaraldehyde tanned and chrome tanned hide powders, non-tanned hide powder showed higher immobilized enzyme activity. The optimal enzyme immobilization conditions are as follows: 10mg of enzyme was immobilized onto 500mg of hide powder in a 50mM phosphate buffer of pH 7.5 at 20°C, and 20% glutaraldehyde offer (based on the hide powder weight) was used to couple the enzyme and collagen fibers. The enzyme activity yield was about 35% and the hide powder immobilized OPH has an enzyme activity of about 7U/g. The immobilized enzyme showed the same temperature and pH profiles as the free enzyme, and it performed at much higher pH and with better thermal stability. The Km value of the immobilized enzyme was a somewhat higher (0.388mM) than that of the free enzyme (0.215mM). The reusability test showed that about 85% activity was retained after 10 use cycles. After storing for eight months at 20°C, the residual activity of the immobilized enzyme preparation was 98%.
(Wikipedia)

Solutions (chimie)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : This study examines the relationship of Cr(VI) formation in leather and wool fibres to the level of Cr and the ironing temperature in double-face processing. The chromium oxide levels in both the leather and wool fibres of the skins were measured. An increase in the amount of basic chromium sulphate used was shown to increase the chromium content in both leather and wool fibres. Related to this increase, parallel increases in the amount of Cr(VI) in leather and wool fibres were detected by the analysis method - DIN 53314 (IUC 18: 1997).
Furthermore, higher temperatures during the ironing processes produced higher levels of Cr(VI) in leather and wool fibres. To eliminate any false-positive results due to the interference of dyestuffs in the Cr(VI) determination with dark coloured extracts, Br2/water solutions were added to extract solutions and the measurements were repeated in the transparent extracts. A comparison of the results revealed that in the extracts with Br2/water addition, Cr(VI) values were lower and it was concluded that Br2/water solution eliminated the dyestuff interference and, therefore the false-positive results.Note de contenu : - EXPERIMENTAL PROCEDURE : Double-face processing - Cr2O3 Analysis and Cr(VI) determinations - Decolorising the extract by Br2/water addition - Reaction mechanism of Br2/water
