However, the characterization of the degradation degree of elastin in skins and leathers was mostly studied through histological analysis qualitatively. There is an urgent need to develop a more efficient quantitative analytical strategy to evaluate the degradation of elastin in leather making processes. In this study, a simple and rapid HPLC method is developed for measuring elastin degradation in skins, leathers and leather processing liquors through determining the biological markers of elastin, namely desmosines. The separation of analytes was conducted on an C18 column (4.6 × 150 mm, 4.0 μm) at 30 ℃; the wavelength of diode array detector (DAD) was set at 275 nm; the mobile phases were composed of methanol and aqueous acetic acid (2.0 %, v/v). A gradient elution was carried out at a flow rate of 0.5 mL/min. It has been witnessed that Cr (III) has no effect on the retention time and peak area of desmosines, when the concentration of Cr (III) was in the range of 0 to 200 mg/L. In quantitative analysis, all of the calibration curves showed good linear regression (R2 ≥ 0.9990) within the tested ranges, and the recovery of desmosines was 100.9 % and 102.6 % for elastin hydrolysate and Cr-tanned elastin hydrolysate, respectively. The content of desmosines in elastin fiber, Cr-tanned elastin fiber and leather manufacturing liquors measured by established HPLC-DAD were comparable to the results obtained by amino acid analyzer. In the wet blue bating process, the quantitative analysis results are consistent with the histological staining results. The results demonstrate that the developed method is accurate and effective and can be readily utilized for the comprehensive process control of leather manufacturing. HydroxyprolineIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : In the area of leather development research, determining collagen content through measurement of 4-Hydroxyproline (Hyp) has been major challenging task due to the interference of various leather-processing chemicals. To overcome this problem, for the first time we have used a High-Performance Liquid Chromatography coupled with Fluorescence Detection (HPLC-FLD) method for accurate determination of Hyp in leather samples through chemical derivatization of Hyp with 7-chloro 4 nitro benzofuran (NBD-Cl) reagent. The HPLC-FLD analysis was performed on a PICO TAG column with an isocratic mobile phase (80 % of 0.1M, pH 7.2 sodium acetate and 20% of acetonitrile, v/v) at a flow rate of 0.7mL/min. The detection was carried out at an excitation wavelength of 465 nm and its emission at 535nm. The retention time of Hyp was found to be ~3.5 minutes and the total run time was about 10 minutes. The method validation indicated that this analytical method is precise (3-12% RSD), accurate (90-100%), the limit of detection 0.01μg/ml, the limit of quantification - 0.03 μg/ml and linear (R2 -0.9995) over the concentration range of 0.1 -2.0 μg/ml. The obtained result indicated that the assay linear range was acceptable for repeated analysis and suitable for the complete range of hydroxyproline levels present in leather samples. Compared to the traditional method (IUC 17:1980), this analytical method demonstrates higher simplicity, specificity, reproducibility, and it could be useful for certifying leather products as well as inspecting international trade in leather and hides. Effects of temperature on the release behaviour were investigated. The formaldehyde emission model in sealed system was established according to the law of mass transfer under several reasonable assumptions and verified by the fitting of actual release data.
The results showed that formaldehyde emission could be divided into three stages : the linear free release, barrier release and chemical release stages. The linear release .time inversely correlated with mass transfer coefficient of leather. The temperature had a significant promotion impact on release during linear release time. The higher the temperature was, the faster the formaldehyde release. Different types of leather showed different release characteristics because of porosity. The average release speed for sheepskin was faster than that of cowhide at the same temperature. However, with the increase of temperature, the increase in the rate of release speed of cowhide was greater. The columns employed in this work include a ZirChrom-DiamondBond-C18, a XTerra MS C18 and a XBridge C18 column. The quantity of methanol consumed by the greener HTLC sunscreen methods developed in this project is significantly reduced although the HTLC separation at this stage is not as efficient as that achieved by traditional HPLC. SBWC separation of sunscreens was also achieved on the XTerra MS C18 and the XBridge C18 columns using pure water at 230–250°C. Methanol was eliminated in the SBWC methods developed in this study. Produits antisolairesIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : This work aimed to develop a simple HPLC method for the simultaneous quantitative determination of the ultraviolet (UV) filters, hydrophilic benzophenone-4 and lipophilic octocrylene, in the presence of three other commonly used UV filters, avobenzone, octisalate and homosalate.
- Methods : Reverse-phased HPLC was performed on a C18 column. A scouting gradient was initially used to determine the approximate mobile phase composition required for efficient analyte elution and separation before further optimization. The assay was validated with regard to specificity, linearity, intra- and inter-day accuracy and precision, limits of detection and limits of quantification. An ultrasound dispersion extraction method for the UV filters from a commercial sunscreen was developed, and the extraction efficiencies from spiked samples were calculated.
- Results : An acetonitrile-methanol-water mixture (20:67:13, v/v/v), where the water component contained 0.2% trifluoroacetic acid (v/v), was found to be the optimal mobile phase at a flow rate of 1.0 mL/min. The assay was linear between 1.0–100 μg/mL for both benzophenone-4 and octocrylene (both correlation coefficients were above 0.999). There was no interference from the excipients of the sunscreen nor from the three other UV filters. The intra- and inter-day accuracy was between 90.0–104.6% for both analytes. Extraction recoveries from a spiked commercial sunscreen were between 95.4 ± 2.1% to 98.5 ± 2.1% for benzophenone-4, and between 87.3 ± 2.3% and 98.9 ± 3.1% for octocrylene. All validation parameters were within the acceptance criteria set out in the International Council for Harmonization (ICH) guidelines. The HPLC assay showed the extracted quantities of benzophenone-4 and octocrylene from the commercial sunscreen closely matched claimed quantities.
- Conclusion : The developed isocratic HPLC method was suitable for simultaneously determining the hydrophilic benzophenone-4 and lipophilic octocrylene in the presence of other commonly used UV filters.
