ElastasesEn biologie moléculaire, les élastases sont des enzymes produites par certains animaux (dont les mammifères), appartenant à la classe des protéases.
Exclusivement produites dans le pancréas, elles sont diffusées dans le corps où elles ont un rôle immunologique (attaque des parois externes de bactéries) et où elles catalysent l’hydrolyse de l’élastine, une fibre élastique qui - avec le collagène - détermine les propriétés mécaniques du tissu conjonctif.

Toxicologie cellulaireIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Coenzyme Q10 (CoQ10) is a major ingredient in skin care products because of its anti-wrinkle effects, although it has some side effects especially at higher amounts. In this study, we compare the anti-wrinkle related properties of CoQ10 and a proprietary Commiphora mukul gum resin (guggul) and triheptanoin preparation (GU-TC7). GU-TC7 is prepared with a supercritical CO2-co-solvent extraction with ethanol, standardized to 2% guggulsterones and triheptanoin, a triglyceride composed of three 7-carbon fatty acids. Treatment of CCL-110 skin fibroblasts with GU-TC7 demonstrates a mild proliferative effect compared to CoQ10 and increased type I collagen synthesis. Additionally, GU-TC7 inhibited matrix metalloproteinase-1 (MMP-1) expression in a dose-dependent manner at 20–100 ?g mL?1 and inhibited human elastase expression by more than 50% as compared to no elastase inhibition with CoQ10 treatment. These results suggest that GU-TC7 possesses properties that are applicable to the treatment of wrinkles and may be considered for its further evaluation in skin care products. Peroxyde d'hydrogèneLe peroxyde d'hydrogène (H2O2), communément appelé eau oxygénée ou encore perhydrol (appellation industrielle), est un composé chimique liquide et visqueux, aux puissantes propriétés oxydantes (il est aussi réducteur). C'est donc un agent blanchissant efficace qui sert de désinfectant et (à haute concentration) d'oxydant ou monergol dans les fusées spatiales.

Vieillissement cutané -- PréventionIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Objective : One of the main approaches to preventing skin ageing is to protect fibroblast cells from oxidative stress. The promoting effect of the human amniotic membrane extract (hAME) on re-epithelization, proliferation and migration of cells in wound healing has been already well studied. This experimental study aimed to investigate the antioxidant activity of hAME against hydrogen peroxide (H2O2)-induced dermal fibroblast damage.
- Methods : Here, to establish the ageing model, human foreskin fibroblasts (HFFs) were exposed to 200 μM H2O2 for 2 h. HFFs were treated with 0.1 mg/ml AME for 24 or 48 h before or/and after H2O2 exposure. A total of 48 h following the H2O2 treatment, we measured cell proliferation, viability, senescence-associated β-galactosidase (SA-β-Gal), antioxidants and preinflammatory cytokine (IL-6) levels, as well as the expression of senescence-associated genes (P53 and P21).
- Results : The obtained results indicated that under oxidative stress, AME significantly increased cellular viability and not only promoted the cell proliferation rate but also attenuated apoptotic induction condition (p < 0.001). AME also significantly reversed the SA-β-Gal levels induced by H2O2 (p < 0.001). Additionally, both pre- and post-treatment regimen by AME down-regulated the expression of senescence marker genes (p < 0.001). Moreover, AME declined different oxidative stress biomarkers such as superoxide dismutase and catalase and increased the glutathione amount.

Peau -- Soins et hygièneIndex. décimale : 668.5 Parfums et cosmétiques Résumé : CONTEXTE : Le vieillissement cutané résulte de facteurs intrinsèques mais aussi extrinsèques et l'exposition aux UV en est une cause principale. Il y a donc tout intérêt à disposer de moyens pour protéger la peau des dommages induits par l'exposition aux UV. Nous avons sélectionné un extrait de la microalgue d'eau douce Scenedesmus rubescens et évalué ses capacités à protéger la peau du photo vieillissement causé par l'exposition aux UV.
METHODES : Des cellules de peau in vitro et ex vivo ont été analysées sur des marqueurs de photo vieillissement induits par l'exposition aux UV tels que la baisse de la viabilité cellulaire, la baisse de la teneur en collagène, l'hyperpigmentation et le nombre de cellules coup de soleil (« sunburn cells »).
RESULTATS : Nous avons trouvé qu'un extrait de la micro algue Scenedesmus rubescens était capable de supprimer les signes cellulaires de vieillissement induits par l'exposition aux UV. Il améliore la viabilité des fibroblastes dermique, préserve la teneur en collagène dans le derme, inhibe la formation de « sunburn cells » et inhibe l'activité de la tyrosinase.
CONCLUSION : Un extrait de Scenedesmus rubescens a montré une large activité sur les marqueurs du vieillissement cutané induit par l'exposition aux UV.
