Tannage Résumé : A rapid and sensitive flow injection spectrophotometric method is described for the determination of proteins based on the principle that Aluminum-Alizarin Red S-bovine serum albumin (BSA)-Triton X-100 complex reacts rapidly with proteins to form complexes in pH4.0 weak acid medium, which causes the change of absorption spectra, the maximum absorption peak of the system is located at 500nm. The absorbance difference is linearly related with the concentration of BSA over the range of 5-120mg l-1. The detection limit of the method was 1.42mg l-1. The method was successfully used to determine protein content in leather wastewater samples and the determined results were in good agreement with the spectrophotometry. The relative standard deviation for 11 replicate determinations of the method was 1.12% for 60mg l-1. The standard addition recovery of the method was from 101.29% to 117.32%. Denature Starch II possessed more carboxyl groups and a smaller particle size than Denatured Starch I. The two denatured starches developed in this research and several typical fillers were applied to chrome-tanned crust leather. The contrasting results of filling showed that Denatured Starch II has some characteristics with obvious differences from other fillings. The leather sample filled with Denatured Starch II showed obvious sample filled with Denatured Starch I showed significant softness, large elongation, and low hypsochromic effects. The leather samples filled with Denatured Starch I and II showed low moisture contents, favourable water vapour permeability, and the morphological characteristics of fibre bundles. TannageIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : In order to control the pollution by nitrite, a direct spectrophotometric flow injection method for the determination of nitrite in tannery effluents has been developed. The method is based on the diazotized intramolecular coupling of a 4-amino-5hydroxynaphthalene-2,7-disulfonic acid monosodium salt hydrate (AHNDMS) by the addition of nitrite in a phosphate buffer solution of pH7.5 and the increase in absorbance of the purple complex is monitored at 550nm. Chemical factors and FIA variables affecting the system are also discussed. Under the optimal conditions, the linear range of this method was 20-1500µg l-1 ; the detection limit was 0.15µg l-1 ; the relative standard deviation was 0.67 % (n=11, 450µg l-1). This method was suitable for automatic and continuous analysis, and successfully applied to determine the content of nitrite in tannery effluent. Experiments conducted with the mobilechiller to preserve hides indicates that it is possible to chill the hides to 4°C in less than 1 hour. Studies on degradation of hides during thawing show that the safe duration of transportation of thilled hides in insulated containers might be about 15 hours. The results also confirm that a considerable reduction in TDS (total dissolved solids) and chloride pollution loads can be achieved, without any adverse effect on the final quality of the leathers, by the adoption of the chilling method in place of conventional salt curing. Tanins végétauxIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Ultraviolet-visible diffuse reflection (UV-Vis DR) spectroscopy and Attenuated Total Reflection Fourier Transform Infrared (ATR FT-IR) spectroscopy were employed to investigate the influence of deleterious UV radiation on the pure collagen fibre and plant tannin crosslinked collagen fibre. After exposure to UV radiation, the UV-Vis abosorption intensity of the pure collagen fibre is found to be dramatically increased at eh peak of 280nm, which should be attributed to the increase of photo-products of free tyrosine and phenylamine formed by UV radiation damage. However, the damage effect on collagen fibre is significantly hindered in the presence of barberry tannin. On the other hand, the study of ATR FT-IR spectroscopy also showed that the intensities of amid bands (amide A, B, I and II) of pure collagen fibre. In addition, the weakness tannin stabilized collagen fibre are less than those in the case of pure collagen fibre. In addition, the weakness of C-O-H in-plane deformation vibration and the stretching vibration of phenolic hydroxyls suggested that the UV protection effect of barberry tannin on collagen fibre should be closely related to the photoxidation of phenolichydroxyls in bayberry tannin molecule. TannageIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : In order to control the pollution by nitrate, a simple, accurate and sensitive reverse flow injection analysis (rFIA) spectrophotometric method for determination of nitrate in tannery effluent is proposed in this paper. Firstly, in alkaline medium and with copper ion as catalyst, nitrate is reduced to nitrite by hydrazine sulfate. Then in acid condition, nitrate and sulfanilamide are diazotized. The diazo material is then coupled with N-1-naphthyl ethylenediamine hydrochloride (NEH) to form a red product. The absorbance of the complex compounds is measured at 540nm. Chemical factors ad rFIA variables affecting the system are also discussed. Under the otimal conditions, the linear range of this method was 50-5000µg l-1 ; the detection limit was 7µg l-1 ; the relative standard deviation was 0.844%. This method was suitable for automatic and continuous analysis, and successfully applied to determine the content of nitrate in tannery effluent.
