A rapid and sensitive flow injection spectrophotometric method is described for the determination of proteins based on the principle that Aluminum-Alizarin Red S-bovine serum albumin (BSA)-Triton X-100 complex reacts rapidly with proteins to form complexes in pH4.0 weak acid medium, which causes the change of absorption spectra, the maximum absorption peak of the system is located at 500nm. The absorbance difference is linearly related with the concentration of BSA over the range of 5-120mg l-1. The detection limit of the method was 1.42mg l-1. The method was successfully used to determine protein content in leather wastewater samples and the determined results were in good agreement with the spectrophotometry. The relative standard deviation for 11 replicate determinations of the method was 1.12% for 60mg l-1. The standard addition recovery of the method was from 101.29% to 117.32%. |
