MagnésiumLe magnésium est l'élément chimique de numéro atomique 12, de symbole Mg.
Le magnésium est un métal alcalino-terreux. Il s’agit du neuvième élément le plus abondant de l'univers
. Il est le produit, dans de grandes étoiles vieillissantes, de l'addition séquentielle de trois noyaux d'hélium à un noyau carbo. Lorsque de telles étoiles explosent en tant que supernovas, une grande partie du magnésium est expulsé dans le milieu interstellaire où il peut se recycler dans de nouveaux systèmes stellaires. Le magnésium est le huitième élément le plus abondant de la croûte terrestreet le quatrième élément le plus commun de la Terre (après le fer, l'oxygène et le silicium), constituant 13 % de la masse de la planète et une grande partie du manteau de la planète. C'est le troisième élément le plus abondant dissous dans l'eau de mer, après le sodium et le chlore.
Les atomes de magnésium existent dans la nature uniquement sous forme de combinaisons avec d'autres éléments, où il présente invariablement l'état d'oxydation +2. L'élément pur est produit artificiellement par réduction ou électrolyse. Il est hautement réactif en poudre et en copeaux mais, laissé à l'air libre, il se revêt rapidement d'une mince couche d'oxyde étanche réduisant sa réactivité (passivation par oxydation). Le métal pur brûle aisément sous certaines conditions (en produisant une lumière brillante, blanche, éblouissante caractéristique). En mécanique il est utilisé principalement comme composant dans les alliages d'aluminium-magnésium (parfois appelés magnalium). Le magnésium est moins dense que l'aluminium et l'alliage est apprécié pour sa légèreté et sa résistance plus grande (mécanique et chimique). (Wikipedia)

Their biological activities directly depend on the ionic composition of the spring. However, in order to exhibit beneficial properties, the minerals need to reach viable skin layers. The present study addresses the incorporation of marketed TSW in model cosmetic formulations and the impact of the formulation on skin absorption of magnesium and calcium ions that are known to improve skin barrier function.
- Methods : Marketed TSW was introduced into five formulations. Liposomes were prepared using saturated or unsaturated phospholipids mixed with cholesterol by the thin layer evaporation technique. Emulsions water-in-oil (W/O), oil-in-water (O/W) or double: water-in-oil-in-water (W/O/W) were prepared by high-shear mixing. Skin absorption of Mg2+ and Ca2+ from those formulations was studied in vitro using static Franz diffusion cells under infinite dose condition and under occlusion of the apparatus.
- Results : Mg2+ and Ca2+ penetrate skin samples from TSW. Encapsulating TSW into double emulsion (TSW/O/W) increased skin absorption of both cations of interest and kept the Ca2+/Mg2+ ratio equal to that of TSW in each skin layer. The dermal absorption of Mg2+ from the double emulsion departs from both single emulsions. Application of liposome suspension improved the skin absorption of Ca2+ while keeping constant that of Mg2+, leading to unbalanced Ca2+/Mg2+ ratio inside skin.
- Conclusion : The beneficial effects of TSW are not only due to their action on the skin surface. Their active components, especially Ca2+ and Mg2+ cations, reach viable skin layers in a formulation-dependent manner. Sensibilité cutanéeIndex. décimale : 668.5 Parfums et cosmétiques Résumé : OBJECTIF : Le but de cette étude était d'optimiser la synthèse de l'inhibiteur de plasmine et de l'urokinase (uPA) benzylsulfonyl-D-Ser-homoPhe- (4-amidino-benzylamide) (BSFAB), pour caractériser son activité et le mécanisme d'action et d’évaluer son utilisation pour améliorer la fonction de barrière du stratum corneum (SC).
MÉTHODES : Les méthodes de couplage peptidique ont été utilisées pour synthétiser BSFAB et la chromatographie liquide à haute performance-spectrométrie de masse (CLHP-SM) avec spectroscopie de résonance magnétique nucléaire 1H et 13C (RMN), qui servent à clarifier sa structure et déterminer sa pureté ont été employées. Son mode de liaison a été déterminé par l’étude d'amarrage aux domaines catalytiques de plasmine et d'uPA. Les constantes d'inhibition (Ki) ont été déterminées par des études de cinétique enzymatique et l'effet de BSFAB sur la plasmine, l'uPA et l'expression de la transglutaminase 1 a été déterminé chez les kératinocytes stimulés par les cytokines ou non. Une étude clinique contrôlée contre placebo sur la fonction barrière du SC a été réalisée sur la peau du visage des sujets à peau sensible auto-perçue.
RÉSULTATS : Le BSFAB a été synthétisé avec une pureté élevée (97.3%). Les études in silico ont indiqué que le groupement amidine de BSFAB a été ancré dans la poche S1 de deux enzymes par liaison à Asp189, Ser190 et Gly219, alors que le squelette du résidu D-Ser fait un β-feuillet d'interaction avec Gly216 antiparallèle. BSFAB s'est montré être un inhibiteur efficace de la plasmine et de l'uPA avec des valeurs de Ki de 29 et 25 nM respectivement. BSFAB a également inhibé les activités des protéases kératinocytaires sécrétées dans les conditions de base (plasmine, l'inhibition de 37,7%, P < 0.05 et uPA inhibition de 96.6%, P < 0.01) et des conditions induites par la cytokine (plasmine inhibition de 41,1%, P < 0.05 et uPA inhibition de 97.0%, P < 0.001) et a stimulé l'expression du gène de la transglutaminase 1 dans les kératinocytes stimulés par des cytokines (une expression accrue d'environ 4,5 fois, P < 0.01). Cliniquement BSFAB a été permis d’ améliorer l'intégrité de la barrière SC (P < 0.02 le jour 29) et apporter des améliorations subjectives dans la perception de la peau saine (P < 0.05 au jour 28).
Conclusion : Le BSFAB se lie comme un inhibiteur compétitif réversible aux sites actifs de la plasmine et de l'uPA. En outre, BSFAB améliore positivement l'expression du gène de la différenciation des kératinocytes (transglutaminase 1). KératinocytesLes kératinocytes sont des cellules constituant 90 % de la couche superficielle de la peau (épiderme) et des phanères (ongles, cheveux, poils, plumes, écailles). Ils synthétisent la kératine (kératinisation), une protéine fibreuse et insoluble dans l'eau, qui assure à la peau sa propriété d'imperméabilité et de protection extérieure.
L'épiderme est divisé en 4 couches basées sur la morphologie des kératinocytes (de l'intérieur vers l'extérieur) :
1. stratum germinativum (couche basale à la jonction avec le derme)
2. stratum spinosum
3. stratum granulosum
4. stratum lucidum
5. stratum corneum
Les kératinocytes passent progressivement de la couche basale vers les couches supérieures par différenciation cellulaire jusqu'au stratum corneum ou ils forment une couche de cellules mortes nommées squames, par apoptose. Cette couche constitue une barrière de protection et réduit la perte d'eau de l'organisme.
Les kératinocytes sont en perpétuel renouvellement. Ils mettent environ 1 mois pour aller de la couche basale au stratum corneum mais ce processus peut être accéléré en cas d'hyperprolifération de kératinocyte (psoriasis).

Rapid urbanization exposed us to a variety of unhealthy ambient air pollutants at high concentrations. The emergence of airborne ultrafine particles has added an additional dimension to this already complex problem of air pollution. The skin has different functions, one of them being the protection against the deleterious effect of external agents. The aim of this study is to evaluate the impact of airborne ultrafine particles (UFP) pollution on skin aging and on keratinocyte differentiation.
- Methods : Ex vivo human skin biopsies and cultured keratinocytes stem cells (KSC) were submitted to diesel exhaust-derived UFP. Reactive oxygen species (ROS) production was assessed with the MitoSOXâ„¢ probe. Keratinocyte stemness potential was evaluated by the immunodetection of keratin 15 (K15) and p63 (∆N isoforms). Effect of UFP on the epithelial niche maintenance was evaluated by immunodetection of Sox9. Reconstructed epidermis model was used to assess the impact of UFP on keratinocyte differentiation and aging.
- Results : UFP exposure induced ROS production and disturbed K15, ∆Np63 and Sox9 expression in KSC or ex vivo skin. Finally, investigations on reconstructed epidermis revealed a phenotype marked by impaired keratinocyte differentiation.

This study investigated the effect of different hair-removal techniques on biophysical parameters and the concentrations of key inflammatory biomarkers in the axillae of female Thai subjects. Axillary hair was removed by shaving, plucking or waxing.
- Methods : Following a 2-week washout phase without hair removal, subjects underwent visual assessment for erythema and skin dryness in one (randomized) axilla, then, hair was removed from the axilla by shaving, plucking or waxing according to each subject’s established habit. Erythema and dryness were assessed again 30 min after hair removal, and buffer scrubs collected from depilated and non-depilated axillae and analysed for inflammatory cytokines; after a further 48 h, erythema, dryness and post-inflammatory hyperpigmentation (PIHP) were assessed in the depilated axilla. Biophysical assessments (skin hydration, barrier integrity, elasticity and roughness) were made in depilated and non-depilated axillae.
- Results : All three hair-removal techniques induced an increase in axillary erythema and skin dryness. Shaving was associated with significantly less erythema (P < 0.01), but significantly greater skin dryness (P < 0.05) versus the other techniques 30 min after hair removal. There were no between-technique differences in PIHP or biophysical parameters. Interleukins IL-1α and IL-1RA concentrations increased, and IL-8 concentration decreased following hair removal by each technique.
- Conclusion : This is the first study to identify the principal cytokines associated with the inflammatory process triggered by axillary hair removal.
