Therefore, research on skin physiology and the development of active ingredients for products for the treatment of rough and dry skin conditions are important and universal topics.
With the purpose of clarifying the role of internal factors in the process of dry skin occurs, we investigated from a pharmacological perspective as a new experimental strategy. Through this research, we have conceived a new theory explaining how dry skin occurs. Furthermore, on the basis of this theory, we have developed a new effective compound for its treatment.
In order to study on the mechanism by which dry skin occurs, we used anti-inflammatory agents and inhibitors against histological impairment mediators, as well as various substances that were considered to regulate the function of epidermal cells. The results showed that protease inhibitors, especially the plasminogen (PLG) activation system-specific inhibitors, were effective against dry skin caused by extrinsic factor. Furthermore, its effects closely correlated with the inhibition activity against the PLG activation system. In addition, we demonstrated immunohistologically that the PLG activation system, which was only located along the epidermal basal layer of normal skin, was widely distributed throughout all layers of epidermis while dry skin induction. These results strongly suggested that the occurrence of dry skin involves a cause bringing about the over-manifestation of the epidermal PLG activation system, which in turn causes abnormalities in the regulating mechanisms for the proliferation and differentiation of epidermal cells, and these result in dry skin.
We discovered trans-4-aminomethylcyclohexanecarboxylic acid (t-AMCHA), which was the most effective substance in view of the theory on the occurrence of dry skin. Then we investigated in detail the efficacy of t-AMCHA. The results of our studies confirmed that t-AMCHA strongly suppresses the over-manifestation of the PLG activation system in the epidermis when dry skin was occurring. In addition, t-AMCHA demonstrated superb effectiveness against phenomena caused by dry skin, including the loss of moisture from the horny layer, the increase in transepidermal water loss (TEWL), accelerated turnover in the horny layer, and the changes in various other indicators such as epidermal hypertrophy. These results strongly supported the effectiveness of t-AMCHA on dry skin.
Based on these research findings, we have further developed a novel active ingredient, t-AMCHA, which is being widely applied to skin care products.
Furthermore, subsequent research on the efficacy of t-AMCHA has confirmed this compound to be effective in promoting restoration and recovery of the barrier function of the stratum corneum, which is a key role of the horny layer.
We have also conducted research on the skin physiology of acne and/or pimples of the pilosebaceus unit, which are the main skin problem for Japanese females in their 20s and 30s. Examination of the skin condition of females who are prone to develop these problems demonstrates a relative lowering of several physiological functions of the horny layer. These results indicate the necessity of incorporating new skin care technology designed to targets improvement on the horny layer function.
With the background, we focused on the effectiveness of t-AMCHA in improving functions of the horny layer and developed new products by incorporating rough rough skin control technology into conventional and basic counter-acne techniques. In-use studies of these products showed favorable results.

In this study, we have discovered that a urokinase-type Plg activator (UK), a trigger of the Plg activation system, which was previously believed to work within the epidermis, also exists in stratum corneum (SC). Focusing on the UK reaction in SC, we sought to develop a method of dry skin prevention. First, the removal of UK by adsorption with inorganic powders was investigated. Screening of powders, revealed that silicia and talc afforded superior UK adsorption. Futhermore, zinc oxide showed superior UK inhibition. The combination of these two different powders enabled us to develop the composite powder, "Skin Care Powder" (SCP), which has an excellent performance in both UK adsorption and UK inhibition. The daily application of a powdery foundation containing SCP for 2 months resulted in a significant suppression of increased transepidermal water loss and a better texture compared to the control. Our results demonstrated that certain inorganic powders can bring significant biological benefit to the skin, whilst remaining on the skin surface. Sensibilité cutanéeIndex. décimale : 668.5 Parfums et cosmétiques Résumé : OBJECTIF : Le but de cette étude était d'optimiser la synthèse de l'inhibiteur de plasmine et de l'urokinase (uPA) benzylsulfonyl-D-Ser-homoPhe- (4-amidino-benzylamide) (BSFAB), pour caractériser son activité et le mécanisme d'action et d’évaluer son utilisation pour améliorer la fonction de barrière du stratum corneum (SC).
MÉTHODES : Les méthodes de couplage peptidique ont été utilisées pour synthétiser BSFAB et la chromatographie liquide à haute performance-spectrométrie de masse (CLHP-SM) avec spectroscopie de résonance magnétique nucléaire 1H et 13C (RMN), qui servent à clarifier sa structure et déterminer sa pureté ont été employées. Son mode de liaison a été déterminé par l’étude d'amarrage aux domaines catalytiques de plasmine et d'uPA. Les constantes d'inhibition (Ki) ont été déterminées par des études de cinétique enzymatique et l'effet de BSFAB sur la plasmine, l'uPA et l'expression de la transglutaminase 1 a été déterminé chez les kératinocytes stimulés par les cytokines ou non. Une étude clinique contrôlée contre placebo sur la fonction barrière du SC a été réalisée sur la peau du visage des sujets à peau sensible auto-perçue.
RÉSULTATS : Le BSFAB a été synthétisé avec une pureté élevée (97.3%). Les études in silico ont indiqué que le groupement amidine de BSFAB a été ancré dans la poche S1 de deux enzymes par liaison à Asp189, Ser190 et Gly219, alors que le squelette du résidu D-Ser fait un β-feuillet d'interaction avec Gly216 antiparallèle. BSFAB s'est montré être un inhibiteur efficace de la plasmine et de l'uPA avec des valeurs de Ki de 29 et 25 nM respectivement. BSFAB a également inhibé les activités des protéases kératinocytaires sécrétées dans les conditions de base (plasmine, l'inhibition de 37,7%, P < 0.05 et uPA inhibition de 96.6%, P < 0.01) et des conditions induites par la cytokine (plasmine inhibition de 41,1%, P < 0.05 et uPA inhibition de 97.0%, P < 0.001) et a stimulé l'expression du gène de la transglutaminase 1 dans les kératinocytes stimulés par des cytokines (une expression accrue d'environ 4,5 fois, P < 0.01). Cliniquement BSFAB a été permis d’ améliorer l'intégrité de la barrière SC (P < 0.02 le jour 29) et apporter des améliorations subjectives dans la perception de la peau saine (P < 0.05 au jour 28).
Conclusion : Le BSFAB se lie comme un inhibiteur compétitif réversible aux sites actifs de la plasmine et de l'uPA. En outre, BSFAB améliore positivement l'expression du gène de la différenciation des kératinocytes (transglutaminase 1).
