Sulfo-carrabioseIndex. décimale : 668.5 Parfums et cosmétiques Résumé : La plupart de femmes adultes ont de la cellulite sur les hanches, les fesses et les cuisses. Même si des troubles de la matrice extracellulaire et du système lymphatique peuvent favoriser son apparition, la cellulite résulte essentiellement d’un stockage lipidique excessif dans le tissu adipeux ce qui exerce une pression considérable sur le tissu cutané environnant et provoque une apparence superficielle irrégulière. La caféine, l’ingrédient anti-cellulite le plus largement utilisé, favorise la lipolyse en inhibant la phosphodiesterase et en favorisant un taux intracellulaire d’AMPc important. Une série d’études a montré que la spermine et la spermidine, deux polyamines omniprésentes, ont favorisé le stockage lipidique et ont ralenti la lipolyse dans le tissu adipeux. De plus, on a montré que l’ les glycosaminoglycans heparane sulfate avaient une forte affinité pour les polyamines. Afin de concevoir un nouvel ingrédient cosmétique avec des propriétés anti-cellulite, nous avons utilisé la modélisation moléculaire pour examiner plusieurs ingrédients avec une structure semblable à celle des glycosaminoglycans heparane sulfate. Ainsi, nous avons identifié le sulfo-carrabiose comme un puissant piègeur de spermine et de spermidine. Ces résultats virtuels ont été d’abord confirmés in tubo où on a montré une inactivation dose-dépendante de la psermine et la spermidine par le sulfo-carrabiose. In vitro, les adipocytes mis en culture avec le sulfo-carrabiose ont montré une réduction significative de la lipogenese et une augmentation significative de lipolyse. Quand le sulfo-carrabiose a été incorporé dans une formule cosmétique, des améliorations significatives ont été observées dans le tour de cuisse, avec de meilleurs résultats que ceux obtenus avec la caféine après 28 jours d’utilisation. En outre, une combinaison de caféine et sulfo-carrabiose a donné des résultats significativement meilleur que ceux obtenus avec la caféine seule. Mesuré par la projection de frange, le volume de la cuisse a également été significativement réduit après un traitement avec le sulfo-carrabiose. Enfin, l’aspect de la cellulite évaluée cliniquement a été également significativement améliorée en 28 jours. SéborrhéeIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - Background : Botanical ingredients are widely used in hair- and skin-care products. However, few studies have investigated the effectiveness of botanical products on counteracting sebum synthesis and secretion.
- Objective : To investigate the composition of Lotus corniculatus seed extract (LC) and its potential inhibition of lipogenesis in SZ95 sebocytes and oily human skin.
- Methods : The active components of LC solutions were identified by high-performance liquid chromatography (HPLC) and nuclear magnetic resonance (NMR). The in vitro effects of LC were evaluated using SZ95 cells treated with linoleic acid (LA) and dihydrotestosterone (DHT) and incubated with LCs for 24 h and 72 h. Lipogenesis was assessed by Oil Red O and Nile Red staining of the cells. In vivo effects were assessed on 30 subjects with oily skin who were enrolled in a randomized, blank-controlled trial and were treated with LC solution for 6 h and 4 weeks. The skin sebum contents and area on the forehead and cheeks were evaluated using a Sebumeter SM815 and Sebfix sebutape with Visioscan VC98. In addition, VISIA was used to collect half-face photos for analysis.
- Results : A novel active molecule, 5′-o-rhamnosyl uridine, was identified in LC. LC exhibited a dose-dependent inhibitory effect on LA and DHT-induced lipid synthesis. When 5% LC was applied for 3 h, the skin sebum contents and area were significantly reduced compared with the vehicle control, with an obvious reduction after 6 h. Continued use of the serum containing 5% LC for 4 weeks resulted in a significant reduction in the skin sebum contents and area. No adverse reactions were reported during the study.
- Conclusions : Topical application of LC resulted in an immediate and long-lasting reduction of the sebum contents and area of oily human skin by reducing sebaceous lipogenesis through the LA and DHT pathways. The oil or sebum attracts dirt more easily, produces even dandruff, and makes the hair stick together. To reduce the sebum and enhance the barrier efficacy on oily scalp, Mibelle Biochemistry has developed an active ingredient based on the Chinese medicinal herb Astragalus membranaceus. AstraForce is a liposomal preparation of Astragalus membranaceus root extract.
This plant is one of the most important Chinese medicinal herbs and its roots have been used for more than 2500 years in Chinese medicine to strengthen qi, the body’s life force. This liposomal form facilitates the penetration of actives into the sebaceous duct allowing the active components to target the sebaceous gland more straightforwardly. The sebum-reducing effect of AstraForce has been proven on volunteers having oily scalp and hair while maintaining the barrier effectiveness. In vitro the active ingredient has reduced the activity of enzymes involved in sebum lipid metabolism. Unfortunately, the current irritating treatments lead to skin dryness, sebaceous gland atrophy and impact the skin barrier integrity. The aim of this study was to evaluate Mangifera indica extract as a new way to solve oily condition for Caucasian, African and Asian skins. Mangifera indica significantly reduced the production of lipids on various ethnicities on human sebocytes derived from iPS. Transcriptomic analysis on all ethnicities of several genes involved in sebum production has demonstrated that Mangifera indica significantly regulated lipogenesis on iPS-derived-human sebocytes. Ex vivo studies confirmed that Mangifera indica played on lipogenesis inhibition without impacting integrity of sebaceous glands. Finally, we confirmed at the clinical level a significant reduction of sebum production on Caucasian, Asian and African skins. Our results showed that Mangifera indica is an inclusive seboregulator, efficient on the three major ethnicities through an innovative mode of action controlling lipogenesis without impacting sebaceous gland morphology. Considering that sebum overproduction is related to acne concern, our results support a role for Mangifera indica in the therapeutic strategy for prevention and treatment of acne.
