ProtéomiqueLa protéomique désigne la science qui étudie les protéomes, c'est-à-dire l'ensemble des protéines d'une cellule, d'un organite, d'un tissu, d'un organe ou d'un organisme à un moment donné et sous des conditions données.
Dans la pratique, la protéomique s'attache à identifier de manière globale les protéines extraites d'une culture cellulaire, d'un tissu ou d'un fluide biologique, leur localisation dans les compartiments cellulaires, leurs éventuelles modifications post-traductionnelles ainsi que leur quantité.
Elle permet de quantifier les variations de leur taux d'expression en fonction du temps, de leur environnement, de leur état de développement, de leur état physiologique et pathologique, de l'espèce d'origine. The effect of photodamage on facial stratum corneum (SC) is still poorly understood.
OBJECTIVE :
To describe the SC proteome from tape strippings of Caucasian SC from photoexposed cheek and photoprotected post-auricular (PA) site, a global analysis of photodamage on the skin will be developed leading to a better understanding of keratinocyte signalling pathways and identification of new molecular targets for the treatment of photoaged skin.
METHODS :
Female Caucasian subjects had nine consecutive tape strippings taken from their cheeks and PA site. Proteins were extracted and the trypsin-digested peptides were analysed by nanochromatography coupled to a high-resolution mass spectrometer. Data-dependent acquisition allowed protein identification that was processed by Paragon algorithm of Protein Pilot software.
RESULTS :
Changes in the levels of epidermal differentiation proteins were apparent indicating poor epidermal differentiation and SC maturation (keratins, cornified envelope (CE) proteins) on photoexposed cheeks. Differences in protease–anti-protease balance were observed for corneodesmolysis (favouring desquamation) and filaggrinolysis (favouring reduced filaggrin processing). 12R-LOX, a CE maturation enzyme, was reduced in photodamaged skin but not transglutaminases. Changes in signal keratinocyte transduction pathway markers were demonstrated especially by reduced levels of downstream signalling markers such as calreticulin (unfolded protein response; UPR) and increased level of stratifin (target of rapamycin; mTOR). Evidence for impaired proteostasis was apparent by reduced levels of a key proteasomal subunit (subunit beta type-6). Finally, key antioxidant proteins were upregulated except catalase.
CONCLUSION :
Clear examples of poor keratinocyte differentiation and associated metabolic and signalling pathways together with reduced SC maturation were identified in photodamaged facial SC. Corneocyte immaturity was evident with changes in CE proteins. Particularly, the reduction in 12R-LOX is a novel finding in photodamaged skin and supports the lack of SC maturation. Moreover, filaggrinolysis was reduced, whereas corneodesmolysis was enhanced. From our results, we propose that there is a poor cross-talk between the keratinocyte endoplasmic reticulum UPR, proteasome network and autophagy machinery that possibly leads to impaired keratinocyte proteostasis. Superimposed on these aberrations is an apparently enhanced mTOR pathway that also contributes to reduced SC formation and maturation. Sensibilité cutanéeIndex. décimale : 668.5 Parfums et cosmétiques Résumé : OBJECTIF : Le but de cette étude était d'optimiser la synthèse de l'inhibiteur de plasmine et de l'urokinase (uPA) benzylsulfonyl-D-Ser-homoPhe- (4-amidino-benzylamide) (BSFAB), pour caractériser son activité et le mécanisme d'action et d’évaluer son utilisation pour améliorer la fonction de barrière du stratum corneum (SC).
MÉTHODES : Les méthodes de couplage peptidique ont été utilisées pour synthétiser BSFAB et la chromatographie liquide à haute performance-spectrométrie de masse (CLHP-SM) avec spectroscopie de résonance magnétique nucléaire 1H et 13C (RMN), qui servent à clarifier sa structure et déterminer sa pureté ont été employées. Son mode de liaison a été déterminé par l’étude d'amarrage aux domaines catalytiques de plasmine et d'uPA. Les constantes d'inhibition (Ki) ont été déterminées par des études de cinétique enzymatique et l'effet de BSFAB sur la plasmine, l'uPA et l'expression de la transglutaminase 1 a été déterminé chez les kératinocytes stimulés par les cytokines ou non. Une étude clinique contrôlée contre placebo sur la fonction barrière du SC a été réalisée sur la peau du visage des sujets à peau sensible auto-perçue.
RÉSULTATS : Le BSFAB a été synthétisé avec une pureté élevée (97.3%). Les études in silico ont indiqué que le groupement amidine de BSFAB a été ancré dans la poche S1 de deux enzymes par liaison à Asp189, Ser190 et Gly219, alors que le squelette du résidu D-Ser fait un β-feuillet d'interaction avec Gly216 antiparallèle. BSFAB s'est montré être un inhibiteur efficace de la plasmine et de l'uPA avec des valeurs de Ki de 29 et 25 nM respectivement. BSFAB a également inhibé les activités des protéases kératinocytaires sécrétées dans les conditions de base (plasmine, l'inhibition de 37,7%, P < 0.05 et uPA inhibition de 96.6%, P < 0.01) et des conditions induites par la cytokine (plasmine inhibition de 41,1%, P < 0.05 et uPA inhibition de 97.0%, P < 0.001) et a stimulé l'expression du gène de la transglutaminase 1 dans les kératinocytes stimulés par des cytokines (une expression accrue d'environ 4,5 fois, P < 0.01). Cliniquement BSFAB a été permis d’ améliorer l'intégrité de la barrière SC (P < 0.02 le jour 29) et apporter des améliorations subjectives dans la perception de la peau saine (P < 0.05 au jour 28).
Conclusion : Le BSFAB se lie comme un inhibiteur compétitif réversible aux sites actifs de la plasmine et de l'uPA. En outre, BSFAB améliore positivement l'expression du gène de la différenciation des kératinocytes (transglutaminase 1). EnzymesUne enzyme est une protéine dotée de propriétés catalytiques. Pratiquement toutes les biomolécules capables de catalyser des réactions chimiques dans les cellules sont des enzymes ; certaines biomolécules catalytiques sont cependant constituées d'ARN et sont donc distinctes des enzymes : ce sont les ribozymes.
Une enzyme agit en abaissant l'énergie d'activation d'une réaction chimique, ce qui accroît la vitesse de réaction. L'enzyme n'est pas modifiée au cours de la réaction. Les molécules initiales sont les substrats de l'enzyme, et les molécules formées à partir de ces substrats sont les produits de la réaction. Presque tous les processus métaboliques de la cellule ont besoin d'enzymes pour se dérouler à une vitesse suffisante pour maintenir la vie. Les enzymes catalysent plus de 5 000 réactions chimiques différentes2. L'ensemble des enzymes d'une cellule détermine les voies métaboliques qui peuvent avoir lieu dans cette cellule. L'étude des enzymes est appelée enzymologie.
Les enzymes permettent à des réactions de se produire des millions de fois plus vite qu'en leur absence. Un exemple extrême est l'orotidine-5'-phosphate décarboxylase, qui catalyse en quelques millisecondes une réaction qui prendrait, en son absence, plusieurs millions d'années3,4. Comme tous les catalyseurs, les enzymes ne sont pas modifiées au cours des réactions qu'elles catalysent, et ne modifient pas l'équilibre chimique entre substrats et produits. Les enzymes diffèrent en revanche de la plupart des autres types de catalyseurs par leur très grande spécificité. Cette spécificité découle de leur structure tridimensionnelle. De plus, l'activité d'une enzyme est modulée par diverses autres molécules : un inhibiteur enzymatique est une molécule qui ralentit l'activité d'une enzyme, tandis qu'un activateur de cette enzyme l'accélère ; de nombreux médicaments et poisons sont des inhibiteurs enzymatiques. Par ailleurs, l'activité d'une enzyme décroît rapidement en dehors de sa température et de son pH optimums.Index. décimale : 668.5 Parfums et cosmétiques Résumé : - Background : Terminally differentiated keratinocytes acquire corneocyte protein envelopes (CPE) complexed with corneocyte lipid envelopes (CLE). These two structural components of the corneocyte envelopes (CEs) undergo maturation by gaining in hydrophobicity, rigidity and surface area. Linoleoyl acylceramides are processed by 12R-lipoxygenase (12R-LOX) and other enzymes before transglutaminase (TG) attaches w-hydroxyceramides to involucrin in the CPE. Concurrently, structural proteins are cross-linked by TG that has been activated by cathepsin D (CathD).
- Objectives : The primary aim of this work was to demonstrate the impact of relative humidity (RH) during ex vivo CE maturation. Low, optimal and high RH were selected to investigate the effect of protease inhibitors (PIs) on CE maturation and TG activity ; in addition, 12R-LOX and CathD activity were measured at optimal RH. Finally, the effect of glycerol on ex vivo CE maturation was tested at low, optimal and high RH.
- Methods : The first and ninth tape strip of photo-exposed (PE) cheek and photo-protected (PP) post-auricular sites of healthy volunteers were selected. Ex vivo CE maturation was assessed via the relative CE maturity (RCEM) approach based on CE rigidity and hydrophobicity. The second and eighth tapes were exposed to RH in the presence of inhibitors.
- Results : Irrespective of tape stripping depth, CEs from PE samples attained CE rigidity to the same extent as mature CEs from the PP site, but such improvement was lacking for CE hydrophobicity. 70% RH was optimal for ex vivo CE maturation. The inhibition of 12R-LOX activity resulted in enhanced CE rigidity which was reduced by the TG inhibitor. CE hydrophobicity remained unchanged during ex vivo maturation in the presence of TG or 12R-LOX inhibition. CE hydrophobicity was enhanced in the presence of glycerol at 44% RH and 100% RH but not at 70% RH. Furthermore, TG activity was significantly diminished at 100% RH compared to the commercial inhibitor LDN-27219. However, a protease inhibitor mix reversed the negative effect of overhydration.

Peau -- Soins et hygièneIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Age‐related differences in maturation parameters of corneocyte envelopes (size, hydrophobicity and rigidity) were examined at several facial test sites in young and old female Caucasians. In addition, the effect of topically applied niacinamide on these parameters was evaluated in a 4‐week placebo‐controlled study.
The stratum corneum (SC) undergoes a variety of catabolic and anabolic reactions towards its outer surface layers in preparation for its external assault from the terrestrial environment. These events are essential for the formation of a healthy barrier. Key is the maturation of the corneocyte envelope (CE) (Fig. 1). Morphologically, CE’s appear fragile in the deeper layers of the SC and more rigid in the outer layers, the mechanics of which have been confirmed with several biomechanical approaches. Early methods to assessing the maturity of the CE’s were based upon staining with tetramethylrhodamine isothiocyanate (TRITC) where rigid CEs stain more intensely than fragile ones. Later, their protein content was assessed by involucrin immunochemistry and their lipid content by Nile red staining which was then expressed as a ratio. Using these approaches, improvements in CE maturity of the volar forearm and the legs have been observed with moisturizers containing glycerol and niacinamide.Note de contenu : - Fig. 1 : Model of the corneocyte envelope (CE). An early step in the cornification process is the formation of the intercellular cytoskeleton mainly composed of keratin filaments and filaggrin. Cross-linking of proteins, mainly of loricrin and involucrin form the rigid inner corneocyte protein envelope (CPE). Then, a lipid monolayer, the corneocyte lipid envelope (CLE), is covalently attached to the CPE. The CLE serves as a scaffold for the lamellar organization of the extracellular lipid matrix.

ProtéinesIndex. décimale : 668.5 Parfums et cosmétiques Résumé : - OBJECTIFS : La quantification des protéines sur des strippings du stratum corneum (SC) est fréquemment utilisée pour étudier les conditions de la peau, pour corriger les quantités de protéines SC retirées lors des études de biomarqueurs du SC et de déterminer la distribution d'ingrédients appliqués localement. Au cours des dernières années, une méthode rapide et pratique pour la quantification des protéines SC à partir de strippings est devenue disponible par densitométrie infrarouge (IRD). Cependant, des courbes standard ont seulement été générées pour le SC de l'avant-bras et de l'épaule de la peau “caucasienne” et ont été supposées être correctes, non seulement pour le SC du visage mais aussi pour les échantillons de SC d'autres groupes ethniques. Le but de la présente étude était de déterminer si l'utilisation de l'IRD pour la mesure des protéines du SC est valable pour d'autres sites de l'organisme tels que la joue et pour mesurer la teneur en protéines SC des types de peau à pigmentation foncée.
- METHODES : Dix sujets féminins caucasiens et dix sujets noirs africains avec la peau normale selon l'auto-évaluation ont participé à l'étude. Des strippings ont été recueillis à partir de deux sites du corps différents (avant-bras et des joues). D'abord l'absorption optique des strippings SC a été déterminée par densitométrie. Cette absorption obtenue (en%) a été comparée à la quantité des protéines de SC absolue, extraite des mêmes strips en utilisant un dosage colorimétrique à l'acide bicinchoninique micro (μBCA).
- RESULTATS : Des quantités plus élevées de protéine SC ont été retirées de l'avant-bras par rapport à la joue (P < 0.01). La concentration absolue en protéines de SC quantifiée par dosage μBCA et l'absorption des protéines SC par l'IRD ont suivi un profil similaire. Il n'y avait pas de différence significative entre les deux ethnies en protéines SC (P > 0.05). Le coefficient global de détermination (R2) montre un bon ajustement à la ligne de régression entre les deux méthodes dans les deux sites (avant-bras = 0.82, joue = 0.77). Aussi, les deux ethnies ont montré une bonne corrélation (R2 ≥ 0.69, P = 0.01).
- CONCLUSIONS : Le SC du visage est morphologiquement distincte de l'avant-bras, comme en témoignent les différences de quantités de protéines SC prélevées. Bien que la distribution des données dans les différents groupes de matières varie, la régression était toujours assez similaire entre les deux parties du corps et les deux groupes ethniques. Aussi les corrélations étaient similaires à celles déjà publiées sur d'autres sites de l'organisme. Les courbes d'étalonnage résultantes peuvent donc être utilisées comme une évaluation indirecte rapide des protéines de SC obtenues par stripping à partir de la joue.Note de contenu : - Materials
