Stabilité thermiqueIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Polyvinyl alcohol (PVA) was oxidised by hydrogen peroxide to prepare oxidised polyvinyl alcohol (OPA). The chemical structure of OPA was characterised by FTIR, SEC and potentiometric titration. PVA was ruptured into fragments along with the formation of terminal aldehyde and carboxyl groups after oxidation. Then the modification effect of OPA on collagen was investigated. A Schiff's base was formed between the aldehyde group of OPA and the amino group of collagen under alkaline conditions, without altering the triple helical structure of collagen. OPA promoted the moderate aggregation of collagen through chain entanglement, and enhanced the thermal stability of collagen. The main reaction between OPA and collagen was proposed as grafting of the single aldehyde component of OPA onto the collagen chain, which was quite different from the cross-linking action of glutaraldehyde. These results suggest the prospect of designing collagen-based biomaterial with a proper range of strength by OPA. GélatineLa gélatine est une substance solide translucide, transparente ou légèrement jaune, presque sans goût et sans odeur, obtenue par l'ébullition prolongée de tissus conjonctifs (peaux) ou d'os d'animaux (principalement porc, bœuf, poisson). Elle possède de nombreuses applications dans le domaine culinaire, la médecine, les industries agroalimentaire et pharmaceutique.
En matière d’étiquetage, la gélatine est considérée par la norme européenne3 comme un ingrédient et non pas comme un additif, c'est pourquoi elle n'a pas de numéro E. Hors Union européenne, elle est considérée par certains pays comme un additif gélifiant et on peut la trouver avec la dénomination E441.
La gélatine est un mélange de protéines obtenu par hydrolyse partielle du collagène extrait de la peau comme la peau de porc (cochon), des os, des cartilages, etc. Les liaisons moléculaires entre les fibres de collagène sont alors brisées. Mélangée à de l'eau, la gélatine forme un gel colloïdal semi-solide thermo-réversible (il fond lorsqu'il est chauffé et recouvre son aspect gélatineux lorsqu'il est refroidi). Sous forme déshydratée, par contre, la gélatine n'a pas de point de fusion et devient friable ou brûle quand elle est chauffée à trop haute températureLa rhéologie de la gélatine se caractérise par un comportement viscoélastique, et des contraintes trop élevées ou appliquées trop rapidement peuvent entraîner une rupture fragile (fracturation) ou ductile6. Le caractère plutôt élastique/fragile ou plutôt visqueux/ductile dépend de la concentration en gélatine de la solution aqueuse et de la température, ainsi que de la durée de la mise sous contrainteLes acides aminés constituant la gélatine sont : la glycine (21 %), la proline (12 %), l'hydroxyproline (12 %), l'acide glutamique (10 %), l'alanine (9 %), l'arginine (8 %), l'acide aspartique (6 %), la lysine (4 %), la sérine (4 %), la leucine (3 %), la valine, la phénylalanine et la thréonine (2 %), l'isoleucine et l'hydroxylysine (1 %), la méthionine et l'histidine (< 1 %) et la tyrosine (< 0,5 %). Ces valeurs sont variables (surtout pour les constituants minoritaires) et dépendent de la source de matériaux bruts et de la technique de préparation. La gélatine est constituée à environ 98-99 % (en poids sec) de protéines et contient 18 acides aminés dont huit des neuf acides aminés essentiels à l'Homme. Elle n'a qu'une relative valeur nutritionnelle du fait de l'absence de tryptophane et de son déficit en isoleucine, thréonine et méthionine; elle possède également un taux inhabituellement élevé d'acides aminés non essentiels, la glycine et la proline (qui sont produits par le corps humain). RhéologieIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Oxidized poly (2-hydroxyethyl acrylate) (OPHEA) with multiple aldehyde groups was prepared as a gelatin crosslinker by polymerization of 2-hydroxyethyl acrylate and then 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) -mediated oxidation of poly (2-hydroxyethyl acrylate) (PHEA). The oxidation conditions were optimized so as to form aldehyde groups in OPHEA as many as possible. The maximum aldehyde content reached 3.66 mmol/g (accounts for 42.5% of the total hydroxyl groups in PHEA) when the oxidation was conducted at 5°C and pH 9.4 for 90 min, by using 30% NaClO as oxidant and 0.1% TEMPO as catalyst. FT-IR and 13C-NMR analyses demonstrated the formation of aldehyde and carboxyl groups in OPHEA. The crosslinking reactivity of OPHEA was confirmed by the enhancement of thermal stability of modified gelatin by DSC and TG analyses. Furthermore, the viscosity of the modified gelatin was significantly increased when the crosslinking reaction was performed at 40°C and pH 8.0. These results could provide support for developing novel aldehyde tanning agents without free formaldehyde. TrypsineLa trypsine (EC 3.4.21.4) est une enzyme digestive du suc pancréatique qui a pour rôle de digérer les protéines.
Elle est synthétisée par le pancréas sous forme de trypsinogène (proenzyme inactive), puis stockée dans les vésicules enzymatiques des cellules acineuses d'où elle est excrétée au moment de la digestion. L'activation du trypsinogène en trypsine est le résultat de l'hydrolyse d'un propeptide sous l'action de l'entérokinase ou par un effet d'autoactivation de la trypsine par elle-même. La cholecystokinine-pancréozymine active la sécrétion des enzymes (donc de la trypsine) dans le suc pancréatique.
La trypsine est une endoprotéase qui hydrolyse les liaisons peptidiques dans lesquelles un acide aminé basique (Lys-|-Xaa ou Arg-|-Xaa) engage sa fonction acide (sauf dans le cas où l'acide aminé suivant (schématisé ici par "Xaa") est une Proline). Elle coupe en C-terminal de ces acides aminés. En d'autres mots, elle transforme les chaînes polypeptides en chaînes protéiques plus courtes pour permettre la digestion. Efficace à pH 7,5 - 8,5, elle est inactivée et digérée en quelques heures à pH neutre (=7) dans l'intestin.
La trypsine participe à l'activation d'autres enzymes comme l'alpha-chymotrypsine par coupure hydrolytique de la chaîne polypeptidique du chymotrypsinogène.
Cette enzyme sert également lors de la 2e semaine du développement embryonnaire humain. Elle est sécrétée par le trophoblaste afin de digérer la zone pellucide entourant le blastocyste. However, it is prone to cause damage to hide collagen fibers and result in loose grain or damaged grain surface due to the longer enzymatic hydrolysis time of the grain layer than the middle layer caused by the slow mass transfer and the rapid hydrolysis reaction of the bating agent in hide. Considering trypsin is the most common bating agent, soybean flour (SF) that contains environmental friendly Kunitz trypsin inhibitor (KTI) was used after delimed hides were treated with trypsin for a certain time to opportunely inhibit its activity in the grain and avoid bating defects. The fluorescence micrographs of hides bated with fluorescently labelled trypsin and KTI showed that KTI could cover only the surface layers when it was added after bating for a certain time, whereas trypsin could penetrate the hide completely. When sufficient SF was added to the bating float at 2 h, the damage to hide collagen caused by trypsin was effectively reduced. In particular, the grain surface remained intact after bating for 8 h. The decrease in the strength of leather caused by bating was also reduced. In addition, the usage of SF scarcely affected the fiber dispersion of the middle layer, thereby ensuring the softness of leather. In summary, opportunely inhibiting the activity of trypsin in the grain could prevent hide damage and improve the quality of leather. Point isoélectriqueEn biochimie, le point isoélectrique (pI) ou potentiel hydrogène isoélectrique (pHI) est le pH auquel une molécule est sous forme d'ion mixte ou, en physico-chimie, le pH d'une solution aqueuse dans laquelle un solide existe sous un potentiel électrique neutre.
En physico-chimie : Selon Bolger, le caractère acide ou basique d'une surface s'exprime par son point isoélectrique " Is ou IEPS (Iso Electric point for the surface) " ou point de charge nulle " PCN ou PZC (Point of Zero Charge) ", défini comme étant le pH de la solution aqueuse dans laquelle le solide existe sous un potentiel électrique neutre. Si le pH de la solution est basique, la surface est acide, et inversement. La différence entre le PZC et l'IEPS est basée sur le phénomène d'adsorption spécifique. On peut considérer que si la grandeur mesurée ne dépend pas de la solution utilisée pour la mesurer (pH, concentration, nature des ions), alors on a affaire à un PZC. Dans le cas contraire, c'est un IEPS que l'on mesure. Par exemple, quand la mesure de goutte sessile à deux liquides est utilisée, on considère en général qu'il n'y a pas adsorption des ions de cette goutte et que la goutte déplace complètement l'alcane qui sert de deuxième liquide: on est alors en présence d’un PZC. Au contraire, dans les mesures de potentiel d'écoulement (streaming potential), la solution joue un rôle important et c'est un IEPS que l'on mesure. Enfin, la charge nette se définit grâce au pH de la solution aqueuse dans laquelle la surface métallique existe, dans un état électriquement neutre (c’est-à -dire [M-OH2+ surf]=[M-O- surf]) et au PZC.
- Si pH < PZC alors la charge nette est positive
- Si pH > PZC alors la charge nette est négative.
Il existe plusieurs méthodes expérimentales permettant de décrire l’état acido-basique de la surface : la mesure du potentiel d’écoulement, la photoélectrochimie, la mesure de l’angle de contact, et la spectroscopie XPS.

Tannage Résumé : Neutralized chrome leathers (pH 6.3) were retanned with typical anionic retanning agents acrylic resin (AC), amino resin (AR) and mimosa extract (ME) alone or together. The isoelectric points (pIs) of the retanned leathers were determined using a zeta potential analyzer for solid- state materials, and the penetratiOn as well as the uptake of the retanning agents in the leathers was analyzed using a fluorescent tracing technique. It was found that the pIs of the surface layers of the leathers retanned with 3% of AC, AR or ME alone became below 5.4, while the pIs of their middle layers were still around 7.4. The pIs of the upper, the middle and the lower layers of the leather retanned with 3% AC, 3% AR and 3% ME together were very close and below 3.6. Additionally, greater penetrations and uptakes of the retanning agents were obtained after retanning with AC, AR and ME together compared to alone. These results indicate that retanning with more and various anionic retanning agents is very useful for a more thorough decrease in the pI of the chrome-tanned leather and favors the penetration and the uptake of the retanning agents in the leather. Séparation (technologie)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : To explore the new approach of value-added utilization of tannery skin wastes, a novel column packing material, collagen fibre adsorbent (CFA), was prepared by using skin wastes as raw material. In the present study, the separation application of CFA on flavonoids and its comparison with other current packing materials (silica gel, polyamide, D101 macroporous resin and Sephadex LH-20) were investigated. Rutin and kaempferol were selected as model flavonoids. Static adsorption indicated that the adsorption capacity of CFA to rutin and kaempferol was different, and the extent of adsorption of kaempferol was lower because it has fewer hydroxyl groups/hydrogen-bond reaction sites. In CFA column chromatography separation, the mixture of rutin and kaemoferol could be well separated by stepwise elution with 90% and 50% aqueous ethanol solutions, and their recoveries were 90.02% and 94.60%, respectively.
However, in comparison with CFA, rutin and kaempferol could not be well separated and recovered by using silica gel, polyamide and D101 macroporous resin columns. In addition, the cost of CFA is much lower, especially compared with Sephadex LH-20.
